Two seaweed concentrates were made from the kelps Ecklonia maxima and Macrocystis pyrifera using a cell burst method. Cytokinin- and auxin-like activities were measured using the soybean callus and mungbean bioassays,
respectively. Cytokinin-like activity was detected in both seaweed concentrates, being equivalent to approximately 50 μg L−1 kinetin. Auxin-like activity was also detected in both concentrates with the E. maxima derived concentrate having higher biological activity, equivalent to 10−5–10−4 M indole-butyric acid. Two replicates of each concentrate were stored at 54 °C for 14 days to accelerate the effects of storage.
Both fresh and stored samples of the two seaweed concentrates were analysed for their endogenous cytokinin and auxin content.
The samples were purified using a combined DEAE-Sephadex octadecylsilica column and immunoaffinity chromatography based on
wide-range cytokinin and IAA specific monoclonal antibodies. These extracts were analysed by HPLC linked to a Micromass single
quadrupole mass spectrophotometer. Eighteen and nineteen different cytokinins were detected, respectively, in the two concentrates,
with trans-zeatin-O-glucoside being the main cytokinin present. Accelerated storage of the concentrates caused an increase in the total
cytokinin concentration with a large increase in the aromatic meta-topolin. Indole-3-acetic acid was the main auxin in both seaweed concentrates. Indole conjugates, including amino acid conjugates,
were also quantified. The total auxin concentration decreased with accelerated storage for both concentrates.

Fifty male Merino lambs (6 to 8 weeks, 14.1 kg; n=10 per group) were used to study the effect of feeding system: barley straw in long form and concentrate pellets in separate troughs (Control), ad libitum alfalfa supplemented with concentrate in separate troughs (Alfalfa) or including various levels of ground barley straw in concentrate pellets (B05, B15 and B25 for 50, 150 and 250 g barley straw/kg), on rumen characteristics, acid-base status, blood cell counts and lymphocyte stimulation. Alfalfa lambs had the heaviest digestive tract contents, highest rumen pH values, lowest volatile fatty acid concentration, highest papillae counts and best mucosa colour and the greatest blood pCO2 values, lowest sodium and chloride and highest potassium concentrations (P<0.05). Including ground barley straw in the concentrate pellet or providing straw in long form separately from the concentrate reduces rumen pH and darkens ruminal mucosa as compared with alfalfa-fed lambs, thus affecting acid-base status. 相似文献

A fluorescent in situ hybridization assay has been developed for the detection of the human-pathogenic microsporidian, Encephalitozoon hellem in water samples using epifluorescence microscopy. The assay employs a 19-nucleotide species-specific 6-carboxyfluorescein-labeled oligonucleotide probe, HEL878F, designed to be complementary to the nucleic acid sequence 878-896, a highly variable segment of the 16S ribosomal RNA of E. hellem spores. The specificity of this probe for its ribosomal RNA target site was confirmed using RNA degradation, ribosomal RNA target site competition, and nucleotide base mismatch control probe assays. Furthermore, the specificity of the HEL878F oligonucleotide probe for E. hellem spores was established when it was evaluated on spores from all three species of the genus Encephalitozoon that had been seeded in reagent water and environmental water concentrates. The specificity of the HEL878F oligonucleotide probe was further corroborated when tested on algae, bacteria, and protozoa commonly found in environmental water. The study demonstrates the applicability of a fluorescent in situ hybridization assay using a species-specific fluorescent-labeled oligonucleotide probe for the detection of E. hellem spores in water samples. 相似文献

According to the characteristics of the reverse osmosis concentrate (ROC) generated from iron and steel company, we used three sets of parallel horizontal subsurface flow (HSF) constructed wetlands (CWs) with different plants and substrate layouts to treat the high-salinity wastewater. The plant growth and removal efficiencies under saline condition were evaluated. The evaluation was based entirely on routinely collected water quality data and the physical and chemical characteristics of the plants (Phragmites australis, Typha latifolia, Iris wilsonii, and Scirpus planiculmis). The principal parameters of concern in the effluent were chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP). The results showed that the CWs were able to remove COD, TN, and TP from ROC. S. planiculmis was not suitable for the treatment of high-saline wastewater. The sequence of metals accumulated in CW plants was K>Ca>Na>Mg>Zn>Cu. More than 70% of metals were accumulated in the aboveground of P. australis. The CW filled with gravel and manganese ore and planted with P. australis and T. latifolia had the best performance of pollutant removal, with average removal of 49.96%, 39.45%, and 72.01% for COD, TN, and TP, respectively. The effluent water quality met the regulation in China. These results suggested that HSF CW planted with P. australis and T. latifolia can be applied for ROC pollutants removal. 相似文献

Soy protein concentrates and soy protein isolates act as ingredients in bakery, meat and dairy products, baby formulas, starting materials for spun textured vegetable products, and other nutritional supplements. In this study, the effectiveness of a liquid-solid circulating fluidized bed (LSCFB) ion exchanger is demonstrated for the recovery of soluble soy proteins from full fat and defatted soy flour. Under steady-state operating conditions, about 50% of the proteins could be recovered from the feed streams entering the ion exchanger. The LSCFB was shown to be a promising system for the recovery of soy protein from both defatted and full fat soy flour solutions. As the ion exchange process captures dissolved proteins, the system may offer a less damaging form of processing compared with the acid precipitation process where soy protein aggregates form and functionality is affected. In addition, the LSCFB allows simultaneous adsorption and desorption of the proteins allowing for a continuous operation. No prefiltration of feed containing suspended particles is required as well, because fluidization is used in place of packed bed technology to improve on current ion exchange processes. 相似文献

X-ray diffraction analysis revealed that pentlandite and chalcopyrite were the prominent mineral phases in a South African sulfidic nickel ore concentrate that hosted nickel and copper. Cobalt was found to be closely associated with the nickel-bearing pentlandite phase of the ore sample. Microbial batch leaching experiments designed according to a central composite design model were run for 15 days in a shaking incubator (150 rpm) at a constant temperature (30°C) with variations in experimental parameters like ore pulp density, particle size, bacterial inoculum, pH of the culture medium, and residence time. Quadratic mathematical models were developed to predict the rate of metal extractions. The suitability of the model of the microbial leaching process was confirmed from normal probability curves. An analysis of variance indicated that the residence time, pulp density of the ore, and particle size were the most significant factors. Bacterial inoculum size hardly showed any effect on the total metal extractions. Maximum nickel (82%), cobalt (76%) and copper (25.6%) extractions were achieved under optimum conditions, operated for 15 days at pulp density of 2% and particle size of ?75 µm at pH 1.5. 相似文献

In organic pig production systems, one of the main challenges is to meet the demand for resources rich in protein. Among the resources available, temperate green plants, such as forage legumes, are potential sources of energy and protein. The aim of the study was to determine the nutritional value of silages (S) from the whole plant of lucerne (L) and red clover (R) and protein pastes (PPs) obtained from L and R leaves. In a first trial, 30 pigs were used in a factorial design to determine the total tract digestibility (TTD) of dietary nutrients and energy in five dietary treatments. The control group was fed a control diet (C1). The lucerne silage (LS) and red clover silage (RS) groups were fed a 78%:22% mixture (on a DM basis) of the C1 diet and LS or RS. The lucerne protein paste (LPP) and the red clover protein paste (RPP) groups were fed an 81%:19% mixture (on a DM basis) of the C1 diet and LPP or RPP. In the second trial, five pigs were used in a 5 × 5 Latin square design to evaluate the standardised ileal digestibility (SID) of amino acids (AAs) in the four legume products. The control diet (C2) was formulated with casein as the sole protein source. The LS and RS groups were fed an 85%:15% mixture (on a DM basis) of the C2 diet and LS or RS. The LPP and RPP groups were fed an 80%:20% mixture (on a DM basis) of the C2 diet and LPP or RPP. Regardless of the plant species, silages obtained from L and R leaves contained less AA and more fibre than protein pastes. While the fresh forages contained the same percentage of protein N in total N (63.6%), lucerne lost more protein N during ensiling than red clover (?75.5 vs ?33.8%). The calculated TTD coefficient of energy was higher in silages than in protein pastes and lower in R than in L products (72.8, 71.5, 67.7, and 61.3 for LS, RS, LPP and RPP, respectively). The SID of total essential AA was higher in LPP than in RPP (87.2 vs 79.2%) whereas it was lower in LS than in RS (33.2 vs 56.8%). The lower SID values in silages were explained by the protein degradation during the ensiling process and a high proportion of AA linked to the NDF fraction. The results of the present study show that protein pastes obtained from lucerne and red clover are valuable protein sources for pig. In contrast, legume silages have to be considered as an energy source rather than a protein source. 相似文献
