Abstract

Four ruminally cannulated Holstein cows (BW 615 kg) in mid lactation were used to evaluate the effects of fibre level (39, 32, 28, 24 and 19% physically effective NDF [peNDF] in DM) in diets consisting of hay and slowly degradable concentrate on chewing activity, under a constant intake level (18 kg DM · d?1). The different dietary fibre concentrations were achieved by adjusting the hay to concentrate ratio. The above-mentioned levels of peNDF corresponded to 25, 40, 50, 60 and 70% concentrate respectively. The diets with decreasing share of concentrate were offered in sequence according to the progressive lactation of the cows. A decrease of the peNDF from 39 – 28% reduced total chewing and rumination time, total number of chews and number of boluses per day as well as chewing time spent per unit of DM. No significant change in chewing behaviour occurred between peNDF level of 28 and 24%. This peNDF level (i.e., 24%) might be considered as a tolerable fibre level. A further reduction of peNDF to 19% led to a significant reduction of chewing activity. This level can be considered as critical for chewing activity and presumably for the rumen health. The chewing parameters correlated linearly to fibre and non-fibre carbohydrates with R2 of 23 – 51%.  相似文献

Abstract

A simple method is described for preparative cell electrophoresis. Solid glass beads (? 0.1 mm diameter) are used as packing material in a cylindrical glass column, the interstices of which contain a citric acid-phosphate buffer of pH 7.6 and 0.015 ionic strength. Cell mixtures containing ? 6 × 109 erythrocytes in 0.2 ml are electrophoresed at 25 V/cm for 1–2 hours after which the column contents are extruded and the separated cell fractions recovered. Complete separation was achieved between mixtures of human and chicken erythrocytes and untreated and papain-treated human erythrocytes. The electrophoretic mobilities of cells subjected to this procedure are in good agreement with previously published mobilities, obtained by microelectrophoresis.  相似文献

研究了牛蒡提取物对副溶血性弧菌的抑制作用。采用80%乙醇分别浸提两个牛蒡品种柳川理想及黄肌牛蒡根的皮及去皮根,四种提取物分别命名为LP、LR、HP和HR。采用滤纸片扩散和最低抑菌浓度法筛选了四种提取物的抑菌效果,时变抑菌研究了常温(25℃)和低温(4℃)下牛蒡提取物在无菌蒸馏水(DW)以及脑心浸汤(BHI)中对副溶血性弧菌的抑制效果。结果显示,四种提取物中LP的抑菌活性最好;经1000μL/mL LP提取液7小时处理,在4℃和25℃DW环境均未有副溶血性弧菌检出;经各浓度提取液1天处理,在4℃和25℃BHI环境均未有副溶血性弧菌检出。LP对副溶血性弧菌弧菌具有良好的抑制作用。  相似文献

The packed‐bed adsorption and elution of aqueous solutions of whey concentrate powders were investigated at pH 3.7 using a 5‐mL SP Sepharose FF column to separate and isolate two major proteins namely, α‐lactalbumin (ALA) and β‐lactoglobulin (BLG) from these solutions. ALA displaced and eluted BLG from the column in a pure form. Pure ALA could then be eluted with good recovery. A novel consecutive two‐stage separation process was developed to separate ALA and BLG from whey concentrate mixtures. Almost all of the BLG in the feed was recovered, with 78% being recovered at 95% purity and a further 20% at 86% purity. Soybean meal is rich in soybean isoflavones, which exhibit antioxidant, anti-inflammatory, antiviral and anticancer functions in humans and animals. This study was conducted to investigate the effects of soybean isoflavones on the growth performance, intestinal morphology and antioxidative properties in pigs. A total of 72 weaned piglets (7.45 ± 0.13 kg; 36 males and 36 females) were allocated into three treatments and fed corn-soybean meal (C-SBM), corn-soy protein concentrate (C-SPC) or C-SPC supplemented with equal levels of the isoflavones found in the C-SBM diet (C-SPC + ISF) for a 72-day trial. Each treatment had six replicates and four piglets per replicate, half male and half female. On day 42, one male pig from each replicate was selected and euthanized to collect intestinal samples. The results showed that compared to pigs fed the C-SPC diet, pigs fed the C-SBM and C-SPC + ISF diets had higher BW on day 72 (P < 0.05); pigs fed the C-SBM diet had significantly higher average daily gain (ADG) during days 14 to 28 (P < 0.05), with C-SPC + ISF being intermediate; pigs fed the C-SBM diet tended to have higher ADG during days 42 to 72 (P = 0.063), while pigs fed the C-SPC + ISF diet had significantly higher ADG during days 42 to 72 (P < 0.05). Moreover, compared to pigs fed the C-SPC diet, pigs fed the C-SBM diet tended to have greater villus height (P = 0.092), while pigs fed the C-SPC + ISF diet had significantly greater villus height (P < 0.05); pigs fed the C-SBM and C-SPC + ISF diets had significantly increased villus height-to-crypt depth ratio (P < 0.05). Compared with the C-SPC diet, dietary C-SPC + ISF tended to increase plasma superoxide dismutase activity on days 28 (P = 0.085) and 42 (P = 0.075) and reduce plasma malondialdehyde (MDA) content on day 42 (P = 0.089), as well as significantly decreased jejunal mucosa MDA content on day 42 (P < 0.05). However, no significant difference in the expression of tight junction genes among the three groups was found (P > 0.05). In conclusion, our results suggest that a long-term exposure to soybean isoflavones enhances the growth performance, protects the intestinal morphology and improves the antioxidative properties in pigs.  相似文献

The changes taking place with age in energy turnover of dairy cattle are largely unknown. It is unclear whether the efficiency of energy utilization in digestion (characterized by faecal and methane energy losses) and in metabolism (characterized by urine and heat energy losses) is altered with age. In the present study, energy balance data were obtained from 30 lactating Brown Swiss dairy cows aged between 2 and 10 years, and 12 heifers from 0.5 to 2 years of age. In order to evaluate a possible dependence of age effects on diet type, half of the cattle each originated from two herds kept at the same farm, which were fed either on a forage-only diet or on the same forage diet but complemented with 5 kg/day of concentrate since their first calving. During 2 days, the gaseous exchange of the animals was quantified in open-circuit respiration chambers, followed by an 8-day period of feed, faeces, urine and milk collection. Daily amounts and energy contents were used to calculate complete energy balances. Age and feeding regime effects were analysed by parametric regression analysis where BW, milk yield and hay proportion in forage as consumed were considered as covariates. Relative to intake of gross energy, the availability of metabolizable energy (ME) increased with age. This was not the result of an increasing energy digestibility, but of proportionately lower energy losses with methane (following a curvilinear relationship with the greatest losses in middle-aged cows) and urine (continuously declining). The efficiency of utilization of ME for milk production (kl) increased with age. Potential reasons include an increase in the propionate-to-acetate ratio in the rumen because of a shift away from fibre degradation and methane formation as well as lower urine energy losses. The greater kl allowed older cows to accrete more energy reserves in the body. As expected, offering concentrate enhanced digestibility, metabolizability and metabolic utilization of energy. Age and feeding regime did not interact significantly. In conclusion, older cows seem to have digestive and metabolic strategies to use dietary energy to a certain degree more efficiently than younger cows.  相似文献

本文采用纳滤分离低浓度乙醇水溶液中迷迭香酸,分析其分离行为并探索富集可行性。结果表明:乙醇浓度对纳滤膜通量和迷迭香酸截留率均有影响,通过响应面法建立二次回归模型,在保障分离效率及截留率的前提下,优化参数为溶质浓度0.41 mg/mL,乙醇浓度20.00%,截留分子量450 Da,迷迭香酸截留率93.81%,预测值较接近与理论值误差较小,数学模型准确可行。通过工艺对比,纳滤富集工艺相较于传统减压浓缩,迷迭香酸保留率提高35.1%,技术优势明显。结果还表明,乙醇水溶液环境下,长周期使用纳滤膜存在溶胀或污染的不可逆性,迷迭香酸截留率未发生明显变化,说明纳滤膜组件分离性能的耐用性较好。  相似文献

Microalgae concentrates, prepared by centrifuging axenic (bacteria-free) cultures of Chaetoceros calcitrans (Paulsen) Takano, were processed and stored under different experimental conditions. The content of ascorbic acid was examined in the concentrates, to assess potential changes in their nutritional properties. In algae pastes stored at 4 °C, it reduced by 29% after 4 weeks storage. As most of the ascorbic acid was retained intracellularly (92%) after resuspension, most of the cells had remained intact. In frozen and dried paste preparations, the losses of ascorbic acid ranged from minor (11% after liquid nitrogen storage for 4 weeks) to major (≥94% after drying at 100 °C for 2 h or at 60 °C overnight). However, most of the remaining ascorbic acid (>85%) in these preparations was rapidly leached from cells upon resuspension. Therefore, pastes stored at 4 °C may have the best potential as an ‘off-the-shelf’ microalgal food product for mariculture. Pastes should now be assessed in animal feeding trials, before being recommended for widespread use in the industry.  相似文献

Although seaweeds and various seaweed products have been utilized in agricultural practices for many years, the precise mechanism by which they elicit their beneficial growth responses is still not fully understood. The amount of mineral nutrients in commercial preparations cannot account for the magnitude of the responses. Some other factor, such as the presence of endogenous plant growth regulators is, therefore, thought to be involved.
