应用玻璃微电极的电生理实验技术,研究日本血吸虫雄性成虫的细胞电活动。结果显示:日本血吸虫雄性成虫皮层细胞膜电位为-47±5. 6mV(X±SE;n=103);肌细胞膜电位为-30.2±4.3mV(X±SE;n=126).肌细胞可记录到自发放电活动,电位去极化幅值为9—28mV。微电极记录的部分细胞(皮层细胞 n=25;肌细胞 n=22)用辣根过氧化酶(Horeradish peroxidase,HRP)标记,进行了细胞组织学鉴定. 相似文献

Abstract The plasmid pGC91.14 was used to introduce via conjugation the Escherichia coli lac operon into fast-growing and slow-growing strains of Rhizobium japonicum . Exconjugants now expressed higher levels of β-galactosidase activity which was still inducible by isopropyl-β- d -thiogalactoside (IPTG). The presence of the lac operon allowed the slow-growing strain 61A76 to grow on lactose as the sole carbon source; the fast-growing strains grew poorly on lactose but growth was not inhibited by lactose as had been reported for Rhizobium meliloti . β-galactosidase could be detected in nodule extracts and bacteroid preparations from soybean plants ( Glycine max L. The morphology of strains of Skeletonema Greville emend Sarno et Zingone was examined in LM, TEM, and SEM and compared with sequence data from nuclear small subunit rDNA and partial large subunit rDNA. Eight distinct entities were identified, of which four were known: S. menzelii Guillard, Carpenter et Reimann; S. pseudocostatum Medlin emend. Zingone et Sarno; S. subsalsum (Cleve) Bethge; and S. tropicum Cleve. The other four species were new: S. dohrnii Sarno et Kooistra sp. nov., S. grethae Zingone et Sarno sp. nov., S. japonicum Zingone et Sarno sp. nov., and S. marinoi Sarno et Zingone sp. nov. Skeletonema species fell into four morphologically distinct groups corresponding to four lineages in the small subunit and large subunit trees. Lineage I included S. pseudocostatum, S. tropicum, S. grethae, and S. japonicum. All have external processes of the fultoportulae with narrow tips that connect with those of sibling cells via fork‐, knot‐, or knuckle‐ like junctions. Lineage II included only the solitary species S. menzelii. Lineage III comprised S. dohrnii and S. marinoi. This latter pair have flattened and flared extremities of the processes of the fultoportulae, which interdigitate with those of contiguous valves without forming knots or knuckles. Lineage IV only contained the brackish water species S. subsalsum. Some species also differ in their distribution and seasonal occurrence. These findings challenge the concept of S. costatum as a single cosmopolitan and opportunistic species and calls for reinterpretation of the vast body of research data based on this species. 相似文献

Floral morphogenesis and the development of Cercidiphyllumjaponicum Sieb. et Zucc. were observed by scanning electron microscopy (SEM). The results showed that the pistillate inflorescences were congested spikes with the flowers arranged opposite. Great differences between the so-called "bract" and the vegetative leaf were observed both in morphogenesis and morphology. In morphogenesis, the "bract" primordium is crescent-shaped, truncated at the apex and not conduplicate, has no stipule primordium at the base but does have some inconspicuous teeth in the margin that are not glandular. The leaf primordium is triangular, cycloidal at the apex, conduplicate, has two stipule primordia at the base, has one gland-tooth at the apex occurring at first and some gland-teeth in the margin that occur later. In morphology, the "bract" is also different to the vegetative leaf in some characteristics that were also illustrated in the present paper. Based on the hypothesis that the bract is more similar to the vegetative leaf than the tepal, we considered that the so-called "bract" of C.japonicum might be the tepal of the pistillate flower in morphological nature. Therefore, each pistillate flower contains a tepal and a carpel. We did not find any trace of other floral organs in the morphogenesis of the pistillate flower. Therefore we considered that the unicarpellate status of extant Cercidiphyllum might be to highly reduce and advance characteristics that make the extant Cercidiphyllum isolated from both fossil Cercidiphyllum-like plants and its extant affinities. 相似文献

A method based on the use of resazurin (RSZ) is described to determine the number of viable Bradyrhizobium japonicum cells in culture medium. The observation of RSZ reduction can be done spectrophotometrically or visually. B. japonicum strains behaved differently when the reducing time was considered.
