细胞表面MHCⅠ类分子在CTL的产生和作为CTL受体的配体清除病毒感染细胞中发挥着重要作用。因此,许多病毒在其生活周期的不同阶段干扰MHCⅠ类抗原呈递并不足为奇。深入理解病毒利用的干扰策略不仅有助于揭示病毒的致病机理,而且有助于制定新的对策避免病毒逃逸,这些研究最终可能建立有效控制病毒感染的免疫疗法。因此,本文将就病毒干扰MHCⅠ类抗原呈递策略的研究进展做一综述。  相似文献

Protein adsorption onto solid substrates usually takes place in an irreversible fashion and this irreversible adsorption also occurs in some enzymatic reactions. In this work the adsorption behavior of intact β-1, 4-glucan-cellobiohydrolase (CBH I) from Trichoderma reesei onto microcrystalline cellulose was monitored by surface plasmon resonance and UV-spectral method. It was found that there existed reversible binding and irreversible binding of CBH I during its interaction with cellulose substrate. To evaluate the influence of adsorption on cellulose enzymatic hydrolysis, the reaction dynamics on pure cellulose were determined. A plot of the hydrolysis rate against the surface density of irreversibly adsorbed CBH I, revealed an inverse relationship in which an apparent decrease in the hydrolysis rate was observed with increasing surface density. Taken together, results presented here should be useful for modifying the binding characteristics of CBH I and making them more effective in cellulose hydrolysis.  相似文献

Capillary isoelectric focusing (CIEF) was used to profile the cellulase composition in complex fermentation samples of secreted proteins from Trichoderma reesei. The enzyme cellobiohydrolase I (CBH I, also referred to as Cel7A), a major component in these extracts, was purified from different strains and characterized using analytical methods such as CIEF, high-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC–PAD), and capillary liquid chromatography–electrospray mass spectrometry (cLC–ESMS). ESMS was also used to monitor the extent of glycosylation in CBH I isolated from T. reesei strain RUT-C30 and two derivative mutant strains. Selective identification of tryptic N-linked glycopeptides was achieved using LC–ESMS on a quadrupole/time-of-flight instrument with a mixed scan function. The suspected glycopeptides were further analyzed by on-line tandem mass spectrometry to determine the nature of N-linked glycans and their attachment sites. This strategy enabled the identification of a high mannose glycan attached to Asn270 (predominantly Man8GlcNAc2) and single GlcNAc occupancy at Asn45 and Asn384 with some site heterogeneity depending on strains and fermentation conditions. The linker region of CBH I was shown to be extensively glycosylated with di-, and tri-saccharides at Thr and Ser residues as indicated by MALDI-TOF and HPAEC–PAD experiments. Additional heterogeneity was noted in the CBH I linker peptide of RUT-C30 strain with the presence of a phosphorylated di-saccharide.  相似文献

 从中国珍稀毒蛛种虎纹捕鸟蛛 (Selenocosmia huwena)毒腺中分离纯化的虎纹捕鸟蛛毒素 - (Huwentoxin ,HWTX- ) ,其一级结构、二级结构和溶液构象均已阐明 ,生理功能实验已证实 HWTX- 是一种作用于突触前膜的神经毒素和高阈值钙通道抑制剂 .为深入开展对 HWTX- 的应用研究 ,根据 HWTX- 多肽氨基酸序列 ,设计其 N-端、C-端和中间段 3组简并引物 (引物 、引物 和引物 ) .从虎纹捕鸟蛛毒腺提取制备 m RNA,逆转录合成 c DNA.以引物 和引物 为引物 ,采用 PCR方法对虎纹捕鸟蛛毒腺总 c DNA进行简并引物扩增 ,得到两条相对特异性的 DNA片段 ,产物直接与 PCR克隆载体 p UC- T连接 .重组子经原引物再次 PCR扩增和同位素标记引物 进行 Southern分子杂交鉴定 .对 4个重组子测序结果表明 ,有 1个重组子所对应的氨基酸顺序与 HWTX- 一致 ,Gen Bank数据检索说明 HWTX- c DNA编码序列确实是一个从未报道的序列 .本研究结果为 HWTX- 在真核细胞表达 ,大量获取蜘蛛毒素组分奠定了基础 .  相似文献

The filamentous fungus Penicillium brasilianum IBT 20888 was cultivated on a mixture of 30 g l−1 cellulose and 10 g l−1 xylan for 111 h and the resulting culture filtrate was used for protein purification. From the cultivation broth, five cellulases and one xylanase were purified. Hydrolysis studies revealed that two of the cellulases were acting as cellobiohydrolases by being active on only microcrystalline cellulose (Avicel). Three of the cellulases were active on both Avicel and carboxymethyl cellulose indicating endoglucanase activity. Two of these showed furthermore mannanase activity by being able to hydrolyze galactomannan (locust bean gum). Adsorption studies revealed that the smaller of the two enzymes was not able to bind to cellulose. Similarity in molecular mass, pI and hydrolytic properties suggested that these two enzymes were identical, but the smaller one was lacking the cellulose-binding domain or an essential part of it. The basic xylanase (pI>9) was only active towards xylan. Two of the purified cellulases with endoglucanase activity were partly sequenced and based on sequence homology with known enzymes they were classified as belonging to families 5 and 12 of the glycosyl hydrolases.  相似文献

A novel triterpenoid, kudinchagenin Ⅰ was isolated from the leaves of Ilex kudincha C. J. Tseng and its structure was determined to be 3β-hydroxy- 11 (12), 13 ( 18 ), 19 ( 29 ) - triene-28,20β-lactonic-ursane by a combination of spectroscopic methods and single crystal X-ray analysis.  相似文献

The effects of Mg2+ concentration on chlorophyll a light-in-duced variable fluorescence and eleetrophoretie velocity of chloroplasts obtained from two varieties of soybean (Ji-Pei No. 1 and Feng-Shou Yellow) were recorded. The light induced variable fluorescence yield and the eleetrophoretie mobility of two kinds of ehloroplasts in the suspending medium containing low level of salt differs from each other. But, the fluorescence yield of chloroplast membranes from two varieties of soybean is relative to that of eleetrophoretic mobility. The pattern of kinetic change of Mg2+ induced variable fluorescence intensity is similar to that of eleetrophoretie mobility. The optimum concentrations of Mg2+ required for the effect on chlorophyll fluorescence and the electrophoretie velocity change are also similar These results indicate that Mg2+ induced excitation energy distribution change between the two photosystems is intimately related to Mg2+-indueed alteration of thylakoil membrane surface eleetrostat.ie property. The possible role of earboxyl group of proteins on Mg2+ induced effects has been discussed in this paper.  相似文献

为了检验已记录的4个溞属种类((隆线溞Daphnia carinata、透明溞D. hyalina、蚤状溞D. pulex和大型溞D. magna)的系统分类, 用试剂盒法分别提取4种溞类的基因组DNA。利用特异性引物, 通过PCR扩增了4种溞属种类的16S rDNA和COⅠ基因部分序列, 并与来自GenBank中每个种类相似度较高的同种序列进行分析。结果表明, 基于16S rDNA和COⅠ基因, 4个溞属种类的平均种间相似度分别为85.56%和80.67%, 碱基中A+T含量均明显高于G+C含量。基于COⅠ基因, 巢湖隆线溞与来自GenBank上的拟同形溞((Daphnia similoides AB549199))相似度为99%, 分歧度为0.4%0.9%。就16S rDNA和COⅠ基因而言, 巢湖透明溞更接近于盔形溞(Daphnia galeata)。基于16S rDNA和COⅠ基因构建的NJ树和贝叶斯树, 两者的主要分枝基本一致。结果暗示, 巢湖隆线溞与拟同形溞为一个种, 透明溞应属于盔形溞。由于缺乏核基因的研究, 有关巢湖溞属的分类地位还需进行更深入的探讨。    相似文献

桔小实蝇幼体及成虫残体DNA条形码识别技术的建立与应用   总被引:1,自引:0,他引:1  
实蝇类害虫多为国内外检疫对象, 其鉴定识别方法主要依据成虫的外部形态特征, 而传统的形态学识别法对口岸经常截获的幼体及残缺的虫体, 则无能为力。本研究以桔小实蝇Bactrocera dorsalis的幼体(卵、 幼虫、 蛹)以及成虫残体(足、 翅、 头部、 胸部、 腹部)为对象, 利用 DNA 条形码技术, 构建实蝇类害虫快速鉴定技术体系, 并以其他4种常见实蝇(包括番石榴实蝇B. correcta、 瓜实蝇B. cucurbitae、 南亚果实蝇B. tau、 柑桔大实蝇B. minax)为对象对该技术体系进行应用验证。结果显示, 桔小实蝇幼体以及成虫残体的碱基序列与数据库中靶标种COⅠ基因碱基序列的一致性为99.51%~99.84%, 其他4种实蝇相应序列与数据库中靶标种COⅠ基因序列的一致性分别为100%, 100%, 99.81%~99.83%和100%; 以邻接法(NJ法)构建系统发育树, 靶标种实蝇均与数据库中对应种实蝇聚为一支, 且置信度均为100%。以K2-P模型计算种内及种间遗传距离得出, 5种实蝇的种间遗传距离为0.0597~0.2363, 平均为0.1693; 种内遗传距离为0.0000~0.0041, 平均为0.0019。这些结果表明, 基于DNA条形码的物种识别技术完全可用于口岸截获的实蝇类害虫幼体及残体的准确鉴定。  相似文献
