The aim of this work was to study, in a rhizobox experiment, the phytoextraction of metals by the hyperaccumulator plant Thlaspi caerulescens in relation to the heterogeneity of metal pollution. Six treatments were designed with soils containing various levels of metals. Homogeneous soils and inclusions of soils in other soil matrices were prepared in order to vary metal concentration and localization. Growth parameters of the plant (rosette diameter and shoot biomass) and localization of roots and shoot uptake of Zn, Cd, Ca, and Mg were determined after 10 weeks of growth. The plants grown on the polluted industrial soils provided a larger biomass and had lower mortality rates than those grown on the agricultural soil. Moreover, these plants accumulated more Zn and Cd (up to 17,516 and 375 mg kg(-1) DM, respectively) than plants grown on the agricultural soil (up to 7300 mg Zn kg(-1) and 83 mg Cd kg(-1) DM). The roots preferentially explored metal-contaminated areas. The exploration of polluted soil inclusions by the roots was associated with a higher extraction of metals. Zinc and Cd in the shoots of Thlaspi caerulescens were negatively correlated with Ca and Mg concentrations; however, the soil supply for these two elements was identical. This suggests that there is competition for the uptake of these elements and that Zn is preferentially accumulated.  相似文献

In this work we have studied the accumulation of heavy metals in two brown trout (Salmo trutta) populations in their natural environment and the participation of metal binding to metallothionein (MT) in this process. Cd, Cu and Zn concentrations, total MT (including Cu MT) and Cd/Zn MT were measured in the gills, liver and kidney of trout inhabiting two rivers, one Cu-contaminated and the other Cd/Zn-contaminated, located at Røros, Central Norway. In both populations, high levels of Cu were found in the liver, whereas Cd was accumulated in liver and particularly in the kidney. The proportions of Cd/Zn MT and Cu MT in liver and kidney, but not in gills, reflected the accumulated and the environmental concentrations of these metals. The total Cu MT concentrations in the investigated tissues, however, were highest in trout from the river with the lowest ambient Cu concentration. It is suggested that MTs are of less importance in Cu-acclimated trout. The data also suggest that acclimation to a Cu-rich environment involves reduced Cu accumulation or increased Cu elimination. In trout from the Cd-rich environment, this metal was mainly bound to MT, whereas in trout from the Cu-rich environment Cd was also associated with non-MT proteins. These findings emphasize the importance to determine both Cd/Zn MT and Cu MT levels, when the participation of this protein in metal handling in trout tissues is investigated.  相似文献

采用室内培养实验(25℃),研究了不同培养时间下重金属Cd、Zn、Cu、Pb(浓度分别为50,800,400,800mg.kg-1)污染对土壤微生物和酶活性的影响。结果表明,土壤蔗糖酶、过氧化氢酶和脱氢酶活性随着培养时间的增加而显著下降,在培养20d的时候达到最小值,然后酶活性缓慢升高。Cu对脲酶活性以及Cd对酸性磷酸酶和脲酶活性的抑制作用随时间增加而增加。土壤微生物生物量碳、细菌、真菌和放线菌数量随培养时间的增加均表现出先降低后升高的变化趋势。Cd和Cu对微生物生物量氮的抑制作用则随着培养时间的增加而增强,在培养30d时微生物生物量氮到达最低值,分别较培养10天减少了12.6%和16.5%。  相似文献

建立了电感耦合等离子体质谱法(ICP-MS)测定茵栀黄注射液中Pb、As、Cd、Hg、Cu 5种元素的方法。样品经微波消解后,直接用ICP-MS同时测定上述5种元素,结果5种元素的检出限分别在5~1250 ng/L之间;线性良好,线性相关系数均为r≥0.999;精密度RSD3.5%;回收率在95.7%~107.5%之间。方法操作简便、分析速度快、灵敏度高,各项分析性能指标均达到要求,适用于茵栀黄注射液中有害元素的测定。  相似文献

Uptake of lipophilic metal complexes by freshwater algae has recently been shown to be pH dependent. Here we look at different physiological aspects that could influence the diffusion of the lipophilic Cd complex, Cd(diethyldithiocarbamate)20 (Cd(DDC)20), into algal cells at different exposure pH values. Changes in cell membrane permeability were assessed as a function of pH for three species of green algae [Chlamydomonas reinhardtii P. A. Dang., Pseudokirchneriella subcapitata (Korshikov) Hindák, and Chlorella fusca var. vacuolata Shihira et R. W. Kraus] using two neutral, nonionic probes, fluorescein diacetate (FDA) and D‐sorbitol. In parallel experiments, we exposed algae to inorganic Cd or to Cd(DDC)20 and monitored Cd intracellular metal distribution, together with phytochelatin synthesis. For the three algal species acclimated at pH 5.5 (w/wo DDC 1 μM) and exposed at this pH, their permeability to FDA and D‐sorbitol was consistently lower than for algae growing at pH 7.0 and exposed at this pH (P < 0.001). The ratio of the FDA hydrolysis rate measured at pH 7.0 with respect to the rate measured at pH 5.5 (both in the presence of DDC) correlated with the ratio of the Cd(DDC)20 initial internalization rate constant obtained at pH 7.0 versus that obtained at pH 5.5 (three algae species, n = 9, r = 0.85, P = 0.004). Our results strongly suggest that acidification affects metal availability to algae not only by proton inhibition of facilitated metal uptake but also by affecting membrane permeability.  相似文献

Phagocytosis is a crucial event in the immune system that allows cells to engulf and eliminate pathogens. This is mediated through the action of immunoglobulin (IgG)-opsonized microbes acting on Fcγ receptors (FcγR) on macrophages, which results in sustained levels of intracellular Ca(2+) through the mobilization of Ca(2+) second messengers. It is known that the ADP-ribosyl cyclase is responsible for the rise in Ca(2+) levels after FcγR activation. However, it is unclear whether and how CD38 is involved in FcγR-mediated phagocytosis. Here we show that CD38 is recruited to the forming phagosomes during phagocytosis of IgG-opsonized particles and produces cyclic-ADP-ribose, which acts on ER Ca(2+) stores, thus allowing an increase in FcγR activation-mediated phagocytosis. Ca(2+) data show that pretreatment of J774A.1 macrophages with 8-bromo-cADPR, ryanodine, blebbistatin, and various store-operated Ca(2+) inhibitors prevented the long-lasting Ca(2+) signal, which significantly reduced the number of ingested opsonized particles. Ex vivo data with macrophages extracted from CD38(-/-) mice also shows a reduced Ca(2+) signaling and phagocytic index. Furthermore, a significantly reduced phagocytic index of Mycobacterium bovis BCG was shown in macrophages from CD38(-/-) mice in vivo. This study suggests a crucial role of CD38 in FcγR-mediated phagocytosis through its recruitment to the phagosome and mobilization of cADPR-induced intracellular Ca(2+) and store-operated extracellular Ca(2+) influx.  相似文献

以分布广泛的沉水植物——黑藻为实验材料,对比研究了Cd胁迫和施加适宜浓度的外源Ca后,黑藻体内Cd积累、矿质营养、光合色素、可溶性蛋白、渗透调节物质、抗氧化能力以及非蛋白巯基(NP-SH)和植物络合素(PCs)的变化,以探讨Ca缓解水生植物Cd毒害的生理生化机制。结果表明:(1)Cd胁迫使黑藻体内Cd含量极显著增加,并造成明显的矿质营养失衡,主要表现为显著降低了P、K、Fe、Cu、Mn的含量,而外源Ca则削弱了黑藻对Cd的蓄积,并在一定程度上减轻了Cd胁迫所造成的矿质元素失衡;(2)Cd处理使黑藻体内叶绿素含量、叶绿素a/b值和可溶性蛋白含量大幅度下降并显著降低了黑藻的总抗氧化能力(T-AOC)和小分子保护物质[谷胱甘肽(GSH)、抗坏血酸(AsA)]的含量,而外源Ca延缓了黑藻的失绿症状,促进了可溶性蛋白的合成并提高了黑藻的抗氧化能力;(3)Cd胁迫使黑藻体内脯氨酸积累显著,而外施Ca减缓了其积累;可溶性糖变化趋势与之相反;(4)Cd胁迫诱导了NP-SH和PCs在黑藻体内的大量累积,外源Ca处理后,其增加幅度减小。以上结果说明外源Ca能通过抑制Cd的吸收,促进光合色素、可溶性蛋白和可溶性糖的合成,维持高的总抗氧化能力和抗氧化物质含量以及矿质营养平衡等途径来增强黑藻对Cd胁迫的抗性。  相似文献

  • Environmental cadmium (Cd) sources have increased in mangrove sediments in recent decades, inducing cellular damage to many plants. Avicennia schaueriana is abundant in mangrove sites and has been subject to Cd contamination. The possible effects of Cd toxicity and the structural and physiological disturbances to this plant were studied. Can this plant express early cellular tolerance mechanisms to such metal contamination?
  • Seedlings of A. schaueriana were collected from sites of their natural occurrence, placed in plastic pots containing nutrient solution for 60 days, and subsequently exposed to increasing Cd concentrations for 5 days under experimental conditions. The anatomical, ultrastructural and physiological changes induced by Cd were analysed.
  • Cd accumulated mainly in the root system and in pneumatophores, stems and leaves, induced differential accumulation of mineral nutrients, but did not induce necrosis or changes in leaf anatomy. However, there was a decrease in starch grains and an increase in deposited electron‐dense material in the cortex and vascular bundles. Cd induced both increases in calcium (Ca) content in shoots and Ca oxalate crystal precipitation in leaf mesophyll and was detected in crystals and in the secretion of salt glands.
  • Our observations and experimental results provide evidence of Cd tolerance in A. schaueriana. As a new feature, despite the clear cellular physiological disorders, this plant is able to eliminate Cd through leaf salt glands and immobilise it in Ca crystals, representing fast mechanisms for Cd exclusion and complexation in leaves in heavy metal coastal polluted marine ecosystems.

  • This study applied Ca in various forms (Ca acetate and CaCl2) and concentrations to reduce cadmium (Cd) concentration in rice and propose a possible mechanism through which Ca acts to control the Cd concentration in rice.
  • The results showed that supplementation of Cd‐contaminated soil with Ca acetate reduced the Cd concentration in rice after exposure for 7 days in both hydroponic and soil conditions. The possible involvement of the auto‐inhibited Ca2+‐ATPase gene (ACA) might act to control the primary signal of the Cd stress response. The messages from ACA3 and ACA13 tended to up‐regulate the low‐affinity cation transporter (OsLCT1) and down‐regulate Cd uptake and the Cd translocation transporter, including the genes, natural resistance‐associated macrophage protein 5 (Nramp5) and Zn/Cd‐transporting ATPase 2 (HMA2), which resulted in a reduction in the Cd concentration in rice. After cultivation for 120 days, the application of Ca acetate into Cd‐contaminated soil inhibited Cd uptake of rice.
  • Increasing the Ca acetate concentration in the soil lowered the Cd concentration in rice shoots and grains. Moreover, Ca acetate maintained rice productivity and quality whereas both aspects decreased under Cd stress.

The effects of Ethylenediamine disuccinic acid (EDDS) (0 and 5?mmol·kg?1) as a synthetic chemical amendment, vermicompost (0 and 5%w/w) as an organic amendment and their combined application were evaluated for the phytoextraction by sunflower (Helianthus annuus L.) of cadmium (Cd) and lead (Pb) at three artificial contamination levels in soils (0, 50, and 100?mg·kg?1 for Cd and 0, 100, and 200?mg·kg?1 for Pb). The results showed that the application of EDDS was the most effective method to increase Pb and Cd concentrations in both parts of the plant. The results also showed that the application of EDDS increased 9.27% shoot Pb content at 200?mg·kg?1 but decreased 15.95% shoot Cd content at 100?mg·kg?1 contamination level with respect to the respective controls. The bioavailable concentrations of Cd at 100?mg·kg?1 and Pb at 200?mg·kg?1 contamination level in the soil at the end of experiment increased 25% and 26%, respectively after the application of EDDS but vermicompost decreased 43.28% the bioavailable Pb concentration relative to their controls. Vermicompost increased the remediation factor index of Cd, thus making it the best treatment for the phytoextraction of Cd. The combined application of EDDS and vermicompost was the best amendment for Pb phytoextraction.  相似文献
