Numerous attempts to capture the morphological variability of the genus Caulerpa have resulted in an unstable classification of numerous varieties and formae. In the present study we attempted to test taxon boundaries by investigating morphological and genetic variation within and between seven taxa of Caulerpa, supposedly belonging to four species, sampled at different sites in a Philippine reef system. Using both field and culture observations, we described the relation between the variability of a set of morphological characters and ecological parameters, such as wave exposure, light intensity, and substrate type. Statistical analyses showed that the limits between two (out of three) ecads of the C. racemosa (Forsskål) J. Agardh complex were obscured by the presence of morphological plasticity. Other studied taxa of Caulerpa (i.e. C. cupressoides [Vahl] C. Agardh, C. serrulata [Forsskål] J. Agardh, and two formae of C. sertularioides [S. Gmelin] Howe) could be grouped based on morphology despite the presence of morphological plasticity. Our results indicated a strong association between light intensity and several quantitative morphological variables. Genetic diversity of these taxa was assessed by partial sequencing chloroplast rbcL and tufA genes and the ycf10‐chlB chloroplast spacer. In all phylogenetic analyses, C. serrulata, C. cupressoides, C. sertularioides, and the three ecads of C. racemosa emerged as distinct genetic units. Despite the presence of morphological plasticity and morphological convergence, a subset of morphological characters traditionally used in taxonomic delimitation still had sufficient discriminative power to recognize the terminal phylogenetic clades.  相似文献

Chemical investigations of two new species of Udotea, reinvestigation of Tydemania expeditionis and Chlorodesmis fastigiata from Guam and Saipan, Marianas Islands, as well as studies of Caulerpa brownii from Southern Australia, have led to the isolation of four new related diterpenoids and several previously described metabolites. These metabolites possess antimicrobial activities and inhibit cell division in the fertilized sea urchin egg bioassay.  相似文献

Morphological and photosynthetic performance were analyzed in species of the genus Caulerpa from an exposed coral reef and a sheltered reef lagoon. Morphological characters, such as distance between modules, number of modules, stolon branches and rhizoid clusters per centimeter of stolon, indicated a uniformity among species within a specific habitat. "Guerilla," or diffusive, growth forms were characteristic for lagoon species and "phalanx," or compact, growth forms for reef species. Differences in photosynthesis were found between Caulerpa species. Sun-tolerant species were found on the reef, and both sun- and shade-tolerant species were present in the lagoon. In the lagoon, shade-tolerant species, such as C. lanuginosa J. Agardh, were found growing in the understory, and sun-tolerant species, such as C. paspaloides (Bory) Greville, formed the canopy. C. cupressoides (West in Vahl) C. Agardh was the only species found in both environments; it showed higher photosynthetic rates and a compressed morphology when growing on the reef and lower photosynthetic rates and expanded morphology for lagoonal ramets. These results suggest that C. cupressoides possesses a broad phenotypic ability to acclimate to lagoonal and reef settings in comparison to other Caulerpa species, enhancing its ecological success in this particular system.  相似文献

A method for isolating high‐quality DNA is presented for the green algae Caulerpa sp. (C. racemosa, C. prolifera, and C. taxifolia) and the brown alga Sargassum muticum. These are introduced, and invasive species in Europe, except for the native C. prolifera. Previous methods of extraction, using cetyl trimethyl ammonium bromide or various commercial kits, were used to isolate genomic DNA but either no DNA or DNA of very low quality was obtained. Genomic libraries were attempted with Caulerpa sp. on three occasions and either the restriction enzyme, the Taq polymerase, or the T4 ligase was inhibited, probably by the large amount of polysaccharides in these algae. The method presented here consists of the rapid isolation of stable nuclei, followed by DNA extraction. Yields of 6–10 μ g genomic DNA from 1 g fresh blades were obtained. After genomic DNA was isolated from fresh material, the quality was checked by agarose gel. Quantification of DNA concentration was performed using UV spectrophotometric measurement of the A 260/ A 280 ratio. The DNA was suitable for PCR, cloning, and hybridization. The DNA isolated using this method allowed successful construction of microsatellite libraries for Caulerpa species and S. muticum . The technique is inexpensive and appropriate for the isolation of multiple samples of DNA from a small amount of fresh material.  相似文献

Biological invasions threaten biodiversity worldwide. Nonetheless, a unified theory linking disturbance and resistance to invasion through a mechanistic understanding of the changes caused to biodiversity is elusive. Building on different forms of the disturbance‐biodiversity relationship and on the Biotic Resistance Hypothesis (BRH), we constructed conceptual models showing that, according to the main biodiversity mechanism generating invasion resistance (complementary vs. identity effects), disturbance can either promote or hinder invasion. Following the Intermediate Disturbance Hypothesis (IDH), moderate levels of disturbance (either frequency or intensity) are expected to enhance species richness. This will promote invasion resistance when complementarity is more important than species identity. Negative effects of severe disturbance on invasion resistance, due to reductions in species richness, can be either overcompensated or exacerbated by species identity effects, depending on the life‐traits becoming dominant within the native species pool. Different invasion resistance scenarios are generated when the diversity‐disturbance relationship is negative or positive monotonic. Predictions from these models were experimentally tested on rocky reefs. Macroalgal canopies differing in species richness (1 vs. 2 vs. 3) and identity, were exposed to either a moderate or a severe pulse disturbance. The effects of different canopy‐forming species on the seaweed, Caulerpa cylindracea, varied from positive (Cystoseira crinita) to neutral (Cystoseira barbata) to negative (Cystoseira compressa). After 2 years, severely disturbed plots were monopolized by C. compressa and supported less Ccylindracea. Our study shows that the effects of disturbance on invasion depend upon its intensity, the main mechanism through which biodiversity generates invasion resistance and the life‐traits selected within the native species pool. Disturbance can sustain invasion resistance when promoting the dominance of competitively subordinate species possessing traits that allow outperforming invaders.  相似文献

Polysaccharides (PF) from marine macroalgae, Caulerpa scalpelliformis were extracted and tested for its potential immunostimulatory and disease resistance properties in fish. Five groups of Nile tilapia (n = 6), Oreochromis niloticus (Linnaeus, 1758) were intraperitoneally administered with the different doses of PF (2, 20 or 200 mg/kg body weight) or with yeast‐derived commercial immunostimulant, Macrogard? (20 mg/kg body weight), to compare the effectiveness. An untreated control group was also maintained. A total of fifteen fibre reinforced plastic tanks (150 L, ambient temperature and light conditions) were used, with triplicate tanks for each group. Only four fish per tank (totally 12 fish from a group) were taken at random and assayed. PF enhanced all the tested non‐specific serum immune responses namely lysozyme, myeloperoxidase, antiprotease, and bactericidal activities. There was an upregulation of the genes encoding IL‐1β, lysozyme and TNF‐α in the spleen of PF injected fish as compared to the control group. In order to study the overall functional immunity, disease resistance test was conducted. Another five groups of fish (n = 10) were treated by intraperitoneal injection with different doses of PF or Macrogard? or untreated as mentioned earlier in triplicates (30 fish per group in three tanks, totally 150 fish in 15 tanks). Seven days post treatment, fish were challenged by intraperitoneal administration of live virulent Aeromonas hydrophila. PF treated fish were protected with significant reduction in the mortality and the consequent increased relative percent survival (RPS) of 92 in the least (2 mg/kg) and middle dose (20 mg/kg) groups. The disease resistance experiment was repeated again but this time, fish were challenged 21 days post treatment that resulted in RPS of 50 for the middle dose. The results clearly show that the intraperitoneal administration of the polysaccharide fraction had a stimulating effect on the non‐specific immune responses, immune gene expression and disease resistance.  相似文献

The distribution of radioactivity into storage and intermediary metabolites during photosynthesis over periods of up to three hours was followed in the siphonous green alga Caulerpa simpliciuscula C. Ag. After the first hour, almost all the carbon assimilated was recovered as insoluble 1,4-α-glucan. There was rapid movement of carbon into insoluble 1,3-β-glucans during the first 20 min of photosynthesis, but little additional carbon moved into these compounds after that time. This contrasted with the movement of carbon into the soluble 1,3-β-glucan fraction, which continued for 2 h. Sucrose accumulated very little 14C during the entire period of photosynthesis. There was a very slow transfer of 14C into lipid throughout the period but the rate was linear. Protein labelling showed a lag of 2 h before it reached the same rate of 14C accumulation as shown by the lipid initially. It is suggested that the distribution of radioactivity can be explained in part by proposing that the tissue is in the latter stages of recovery from the wounding which look place at the time of dividing the thallus into sections.  相似文献

Growth of the invasive algae Caulerpa racemosa var. cylindracea in shallow habitats may influence the faunal assemblage composition. We studied its effects on caprellid assemblages associated with shallow-water habitats of hard and soft bottoms from the SE Iberian Peninsula (native rocky-bottom algae, C. racemosa from hard and soft bottoms, and Caulerpa prolifera, Cymodocea nodosa and Posidonia oceanica from soft bottoms). Samples were taken in two different sampling periods (September 2004 and March 2005). A total of seven caprellid species were identified, with important differences in their distribution in different habitats. Total abundance of caprellids was very high in March on native algae on hard bottoms, and on C. racemosa on both soft and hard bottoms. On both hard and soft bottoms, abundances of Caprella hirsuta recorded from C. racemosa were low. On the other hand, a higher abundance of other species, namely C. acanthifera, C. santosrosai, Phtisica marina and Pseudoprotella phasma, was recorded from C. racemosa. The results indicate that C. racemosa may have a positive influence on some caprellid species, while seasonal changes are also evident. It is concluded that introduced C. racemosa may serve as a new habitat, promoting and maintaining caprellid populations in shallow Mediterranean habitats.  相似文献

The life history of Rhodophysema elegans is described from laboratory culture and the field in Newfoundland, Canada. Sexual plants are lacking and tetraspores germinate to produce new tetrasporangial plants. Approximately 30–35 chromosomes were counted in vegetative cells of successive generations. There was no evidence of chromosome pairing in the early stages of division of tetrasporangial initials, indicating that the tetrasporangia are apomeiotic.  相似文献

为了揭示南海长茎葡萄蕨藻Caulerpa lentillifera共附生真菌的群落结构,并分析自来水清洗对真菌群落多样性的影响,本研究以收集于深圳南海各月份长茎葡萄蕨藻为材料,利用Illumina MiSeq 2500测序平台进行真菌ITS1扩增子测序,分析自来水清洗前后真菌类群组成和多样性的差异。研究发现,从所有18个样品中总共获得914个OTUs,检测出真菌3门12纲32目50科74属106种,其中优势属为曲霉属Aspergillus、单胞瓶霉属Phialemonium、枝孢属Cladosporium、链座菌属Catenulostroma和茎点霉属Phoma;不同月份真菌Alpha多样性指数显示南海长茎葡萄蕨藻共附生真菌丰度和多样性均随着温度降低而降低;自来水清洗前后南海长茎葡萄蕨藻共附生真菌的多样性及群落结构存在一定差异,清洗后真菌OTUs和Alpha多样性指数均有所降低,其中Chao1和Ace指数为显著性下降,表明自来水清洗对真菌群落组成丰度及多样性影响显著,这对长茎葡萄蕨藻的食品安全具有积极意义。  相似文献
