Exocellular pyruvate-containing galactoglucan ofAchromobacter spp.

Exopolysaccharides of a number of slime-producingAchromobacter strains, isolated from activated sludge, were prepared and analysed. They consist ofD-glucose,D-galactose, pyruvic acid and O-acetyl in the approximate molar ratio of 1:1:1:1/2. The polysaccharides were shown by methylation and partial hydrolysis to consist of unbranched chains of alternately arrangedD-glucose andD-galactose residues, exclusively linked by -1,3-linkages, and with pyruvate substituents linked by acetal bonds at positions 0–4 and 0–6 of theD-galactose residues. The significance of the exopolysaccharides in relation to some relevant properties of activated sludge organisms is discussed.     

Localization of cells capable of transdetermination in a specific region of the male foreleg disk ofDrosophila

Summary In the male foreleg disk ofDrosophila melanogaster the cells capable of transdetermination are clustered in a specific region within the upper half of the disk. Cells outside this region cannot transdetermine under any of the experimental conditions thus far applied.Transdetermination occurs when cells capable of transdetermination are stimulated to a certain extent of additional proliferation. This can be achieved either by exposing these cells at a wound surface of an intact fragment, or by dissociation.     

Design,synthesis, and biological evaluation of histone deacetylase inhibitors possessing glutathione peroxidase-like and antioxidant activities against Alzheimer’s disease

A series of hybrids containing the pharmacophores of the histone deacetylase (HDAC) inhibitor, SAHA, and the antioxidant ebselen were designed and synthesized as multi-target-directed ligands against Alzheimer’s disease. An in vitro assay indicated that some of these molecules exhibit potent HDAC inhibitory activity and ebselen-related pharmacological effects. Specifically, the optimal compound 7f was found to be a potent HDAC inhibitor (IC₅₀?=?0.037?μM), possessing rapid hydrogen peroxide scavenging activity and glutathione peroxidase-like activity (ν₀?=?150.0?μM?min^?1) and good free oxygen radical absorbance capacity (value of ORAC: 2.2). Furthermore, compound 7f showed significant protective effects against damage induced by H₂O₂ and the ability to prevent ROS accumulation in PC12 cells.     

Thermal liquid biopsy for monitoring melanoma patients under surveillance during treatment: A pilot study

Background

Differential Scanning Calorimetry (DSC) is a technique traditionally used to study thermally induced macromolecular transitions, and it has recently been proposed as a novel approach for diagnosis and monitoring of several diseases. We report a pilot study applying Thermal Liquid Biopsy (TLB, DSC thermograms of plasma samples) as a new clinical approach for diagnostic assessment of melanoma patients.

Thiols are Main Determinants of Total Antioxidant Capacity of Cellular Homogenates

While the total antioxidant capacity (TAC) of blood plasma is mainly accounted for by urate, TAC of cell interior can be expected to depend more on other antioxidants, especially glutathione and protein -SH groups. We studied TAC of homogenates of several lines of cultured cells subjected to the action of thiol-modifying agents. Comparison of changes of TAC of the homogenates and of the level of total thiols (determined with a biradical spin label) demonstrates that alterations in cellular thiol content is the main determinant of changes of TAC of cell homogenates. These results show that estimation of TAC of cell extracts may be a useful parameter of assessment of oxidative stress, primarily of oxidation of thiol groups, yielding information different than TAC of body fluids.     

Legal and regulatory concerns about transgenic plants in Brazil

Brazil has a biosafety law that was approved in 1995. This law provides for a horizontal type of regulation that coordinates other existing regulatory frameworks in the areas of agriculture, health and environment. Various federal government departments are responsible for implementing the law. The National Technical Biosafety Commission is the national competent authority on biosafety with overall responsibility. In the case of Bt plants or any insecticidal organism, the Agrochemical Law also applies and authorization for laboratory, greenhouse and field studies must be obtained from the Plant Protection Secretariat, the Brazilian Institute of Environment and the National Agency of Health. Furthermore, the National Environmental Council must issue a license for commercialization of any GMO. There is pressure needed for capacity building and to harmonize the regulatory and administrative frameworks among the different federal departments involved. Some perspectives and challenges for the commercial registration of transgenic crops are discussed.     

Effect of histidine on autotaxin activity in experimentally induced liver fibrosis

The aim of this study was to explain whether serum autotaxin (ATX) activity might be a target for regulation of liver fibrosis and to evaluate the hepatoprotective and antifibrotic effects of histidine in thioacetamide (TAA)-induced liver fibrosis in rats. This study was carried out on 100 Wistar Albino rats, classified into five groups, each containing 20 rats: Group I (control group), Group II: rats were given histidine intraperitoneally, Group III: rats were injected intraperitoneally with TAA, Group IV: rats were injected with L-histidine together with TAA, and Group V: rats were injected with TAA for 1 month then treated with intraperitoneal injection of L-histidine for another month. At the end of experiment, blood and liver were collected for determination of some liver enzymes, plasma total antioxidant capacity (TAC), serum ATX activity, and liver tissue hydroxyproline. Thioacetamide treatment caused significant increases in liver enzymes, ATX activities, and liver hydroxyproline, but a significant decrease in plasma's TAC. Upon treatment with histidine, a significant decrease in liver enzymes, ATX activities, and liver hydroxyproline was observed with a significant increase in plasma TAC in Group IV and a significant decrease in Group V. Histidine as an antioxidant has a protective effect on TAA-induced liver fibrosis; it is beneficial in rats not only by inhibition of collagen synthesis and increasing TAC but also by inhibition of ATX activities thus reducing its capacity to produce lysophosphatidic acid, which has a role in liver fibrosis.     

Electrorotation of Erythrocytes Treated with Dipicrylamine: Mobile Charges within the Membrane Show their ``Signature' in Rotational Spectra

In this study, electrorotation spectra of individual cells (that is, frequency dependence of cell rotation speed) have been proved to yield information not only about the passive electric properties of cell constituents, but also about the presence of mobile charges within the plasma membrane being part of ion carrier transport systems. Experiments on human erythrocytes pretreated with the lipophilic anion dipicrylamine (DPA) gave convincing evidence that these artificial mobile charges adsorbed to the plasma membrane contributed significantly to the rotational spectrum at relatively low conductivity of the external medium (2–5 mS m⁻¹). Theoretical integration of the mobile charge concept into the single-shell model (viewing the cell as a homogenous sphere surrounded by a membrane) led to a set of equations which predicted electrorotational behavior of DPA-treated cells in dependence on medium conductivity. The quantitative data on the partition and the transmembrane translocation rate of the DPA anion extracted from the experimental rotational spectra agreed well with the corresponding literature values. Low concentrations of medium-sized HDL particles predict incident CVD in chronic kidney disease patients

Patients with chronic kidney disease (CKD) are at high risk for CVD. However, traditional CVD risk factors cannot completely explain the increased risk. Altered HDL proteome is linked with incident CVD in CKD patients, but it is unclear whether other HDL metrics are associated with incident CVD in this population. In the current study, we analyzed samples from two independent prospective case-control cohorts of CKD patients, the Clinical Phenotyping and Resource Biobank Core (CPROBE) and the Chronic Renal Insufficiency Cohort (CRIC). We measured HDL particle sizes and concentrations (HDL-P) by calibrated ion mobility analysis and HDL cholesterol efflux capacity (CEC) by cAMP-stimulated J774 macrophages in 92 subjects from the CPROBE cohort (46 CVD and 46 controls) and in 91 subjects from the CRIC cohort (34 CVD and 57 controls). We tested associations of HDL metrics with incident CVD using logistic regression analysis. No significant associations were found for HDL-C or HDL-CEC in either cohort. Total HDL-P was only negatively associated with incident CVD in the CRIC cohort in unadjusted analysis. Among the six sized HDL subspecies, only medium-sized HDL-P was significantly and negatively associated with incident CVD in both cohorts after adjusting for clinical confounders and lipid risk factors with odds ratios (per 1-SD) of 0.45 (0.22–0.93, P = 0.032) and 0.42 (0.20–0.87, P = 0.019) for CPROBE and CRIC cohorts, respectively. Our observations indicate that medium-sized HDL-P—but not other-sized HDL-P or total HDL-P, HDL-C, or HDL-CEC—may be a prognostic cardiovascular risk marker in CKD.     

Relationship of the urban fabric of Lawrence,Kansas with satellite-derived thermal infrared data

Many of the twelve land-use types and ten surface materials that comprise Lawrence, Kansas correlate significantly with afternoon and evening temperatures derived from thermal infrared data of the Heat Capacity Mapping Mission satellite. Three afternoon temperature patterns and six evening patterns were analyzed covering late spring through the middle of fall. Land uses account for a mean of 49% of the temperature variance in the daytime; the mean explained variance for the evening data is 37%. The temperature variation accounted for by surface materials is nearly the reverse of that of land use: the daytime and evening mean values are 32% and 49%, respectively. With the primary exception of trees, the correlations closely coincide with previously reported results relating the land uses and surface materials with temperatures for the city obtained from autotraverses.Abbreviations LST local standard time - CDT central daylight time
