通过培养高山被孢霉利用糖蜜来发酵生产花生四烯酸(ARA),研究了不同甘蔗糖蜜预处理方法对ARA发酵生产的影响。研究表明:H2SO4法是最利于ARA发酵生产的糖蜜预处理方法。利用预处理的甘蔗糖蜜发酵生产ARA,通过单因素实验设计,确定了最优的培养条件,包括初始还原糖80 g/L,N源6 g/L,接种量20%,初始pH6.0和培养温度25℃,在此条件下发酵,干细胞质量、油脂含量、ARA产量和糖利用率分别达到28.5 g/L、11.7g/L、3.68 g/L和94.5%。  相似文献

AIMS: To isolate and characterize bacteria from nature capable of producing poly-beta-hydroxyalkanoates in high yields from soy molasses oligosaccharides. METHODS AND RESULTS: Several strains of bacteria were obtained from enrichment cultures employing raffinose as major carbon source and inoculated with soybean field soil, lake sediment, or lake water. Many of the isolates were Bacillus species and produced polyhydroxyalkanoates (PHAs) to high yield. The raffinose-degrading isolates produced endospores, were highly saccharolytic, and both respired and fermented a variety of mono-, di-, tri- and tetrasaccharides. Strain CL1 produced 90% of cell dry mass as PHA from various sugars, including raffinose, and did so without requiring a nutrient limitation. CONCLUSIONS: Strain CL1 could be the catalyst for an industrial fermentation converting soy molasses and other waste carbohydrates to PHAs. The properties of this organism that make it ideally suited for such a fermentation include (i) its ability to use a wide variety of plant-associated carbohydrates as PHA feedstocks; (ii) its rapid growth; (iii) its ability to grow under anoxic conditions; and (iv) its ability to produce spores. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report of bacteria capable of making biodegradable plastics to high yield from soy molasses oligosaccharides.  相似文献

Fourteen yeast strains were screened for production of 2-phenylethanol from l-phenylalanine with molasses as carbon source. Up to 1 g 2-phenylethanol l–1 was obtained. Using oleyl alcohol as a second phase for in situ product removal to enhance the production of 2-phenylethanol increased the yield to about 3 g 2-phenylethanol l–1 at 35 °C. The most productive strains were Kluyveromyces marxianus CBS 600 and CBS 397.  相似文献

A new osmotolerant mutant strain of Zymomonas mobilis was successfully used for ethanol production from beet molasses. Addition of magnesium sulfate to hydrolyzed molasses allowed repeated growth without the need of yeast extract addition. The kinetics and yields parameters of fermentation on media with different molasses concentrations were calculated. The anabolic parameters (specific growth rate, mu, and biomass yield, Y(X/S)) were inhibited at elevated molasses concentrations while the catabolic parameters (specific ethanol productivity, q(p), and ethanol yield, Y(p/s)) were not significantly affected. In addition to ethanol and substrate inhibition, osmotic pressure effects can explain the observed results.  相似文献

The by-product of sugar refinery—waste molasses was explored as alternative to glucose-based medium of Chlorella protothecoides in this study. Enzymatic hydrolysis is required for waste molasses suitable for algal growth. Waste molasses hydrolysate was confirmed as a sole source of full nutrients to totally replace glucose-based medium in support of rapid growth and high oil yield from algae. Under optimized conditions, the maximum algal cell density, oil content, and oil yield were respectively 70.9 g/L, 57.6%, and 40.8 g/L. The scalability of the waste molasses-fed algal system was confirmed from 0.5 L flasks to 5 L fermenters. The quality of biodiesel from waste molasses-fed algae was probably comparable to that from glucose-fed ones. Economic analysis indicated the cost of oil production from waste molasses-fed algae reduced by 50%. Significant cost reduction of algal biodiesel production through fermentation engineering based on the approach is expected.  相似文献

Production of lactic acid from beet molasses by Lactobacillus delbrueckii NCIMB 8130 in static and shake flask fermentation was investigated. Shake flasks proved to be a better fermentation system for this purpose. Substitution of yeast extract with other low cost protein sources did not improve lactic acid production. The maximum lactic acid concentration was achieved without treatment of molasses. A Central Composite Design was employed to determine the maximum lactic acid concentration at optimum values for the process variables (sucrose, yeast extract, CaCO3). A satisfactory fit of the model was realized. Lactic acid production was significantly affected both by sucrose–yeast extract and sucrose–CaCO3 interactions, as well as by the negative quadratic effects of these variables. Sucrose and yeast extract had a linear effect on lactic acid production while the CaCO3 had no significant linear effect. The maximum lactic acid concentration (88.0 g/l) was obtained at concentrations for sucrose, yeast extract and CaCO3 of 89.93, 45.71 and 59.95 g/l, respectively.  相似文献

Ethanol production from sweet sorghum juice by Saccharomyces cerevisiae NP01 was investigated under very high gravity (VHG) fermentation and various carbon adjuncts and nitrogen sources. When sucrose was used as an adjunct, the sweet sorghum juice containing total sugar of 280 g l−1, 3 g yeast extract l−1 and 5 g peptone l−1 gave the maximum ethanol production efficiency with concentration, productivity and yield of 120.68 ± 0.54 g l−1, 2.01 ± 0.01 g l−1 h−1 and 0.51 ± 0.00 g g−1, respectively. When sugarcane molasses was used as an adjunct, the juice under the same conditions gave the maximum ethanol concentration, productivity and yield with the values of 109.34 ± 0.78 g l−1, 1.52 ± 0.01 g l−1 h−1 and 0.45 ± 0.01 g g−1, respectively. In addition, ammonium sulphate was not suitable for use as a nitrogen supplement in the sweet sorghum juice for ethanol production since it caused the reduction in ethanol concentration and yield for approximately 14% when compared to those of the unsupplemented juices.  相似文献

The fungus Mucor indicus is found in this study able to consume glucose and fructose, but not sucrose in fermentation of sugarcane and sugar beet molasses. This might be an advantage in industries which want to selectively remove glucose and fructose for crystallisation of sucrose present in the molasses. On the other hand, the fungus assimilated sucrose after hydrolysis by the enzyme invertase. The fungus efficiently grew on glucose and fructose and produced ethanol in synthetic media or from molasses. The cultivations were carried out aerobically and anaerobically, and manipulated toward filamentous or yeast-like morphology. Ethanol was the major metabolite in all the experiments. The ethanol yield in anaerobic cultivations was between 0.35 and 0.48 g/g sugars consumed, depending on the carbon source and the growth morphology, while a yield of as low as 0.16 g/g was obtained during aerobic cultivation. The yeast-like form of the fungus showed faster ethanol production with an average productivity of 0.90 g/l h from glucose, fructose and inverted sucrose, than the filamentous form with an average productivity of 0.33 g/l h. The biomass of the fungus was also analyzed with respect to alkali-insoluble material (AIM), chitin, and chitosan. The biomass of the fungus contained per g maximum 0.217 g AIM and 0.042 g chitosan in yeast-like cultivation under aerobic conditions.  相似文献

Gordonia polyisoprenivorans CCT 7137 was isolated from groundwater contaminated with leachate in an old controlled landfill (São Paulo, Brazil), and cultured in GYM medium at different concentrations of sugarcane molasses (2%, 6%, and 10%). The strain growth was analyzed by monitoring the viable cell counts (c.f.u. mL?1) and optical density and EPS production was evaluated at the end of the exponential phase and 24 h after it. The analysis of the viable cell counts showed that the medium that most favored bacterial growth was not the one that favored EPS production. The control medium (GYM) was the one that most favored the strain growth, at the maximum specific growth rate of 0.232 h?1. Differences in bacterial growth when cultured at three different concentrations of molasses were not observed. Production of EPS, in all culture media used, began during the exponential phase and continued during the growth stationary phase. The highest total EPS production, after 24 h of stationary phase, was observed in 6% molasses medium (172.86 g L?1) and 10% (139.47 g L?1) and the specific total EPS production was higher in 10% molasses medium (39.03 × 10?11 g c.f.u.?1). After the exponential phase, in 2%, 6%, and 10% molasses media, a higher percentage of free exopolysaccharides (EPS) was observed, representing 88.4%, 62.4%, and 64.2% of the total, respectively. A different result was observed in pattern medium, which presented EPS made up of higher percentage of capsular EPS (66.4% of the total). This work is the first study on EPS production by G. polyisoprenivorans strain in GYM medium and in medium utilizing sugarcane molasses as the sole nutrient source and suggests its potential use for EPS production by G. polyisoprenivorans CCT 7137 aiming at application in biotechnological processes.  相似文献

A polymeric water-insoluble material, the so called canjica, was collected in nine Brazilian sugar and alcohol plants. It was found that all the samples were very similar to each other regarding the moisture and variable in ash contents, whereas CHNS analysis showed that they were carbohydrates. Trace amounts of nitrogen and sulphur compounds were also found to be present, and their source were attributed to proteinaceous material from sugarcane juice contaminants. Paper chromatography and gas chromatography-mass spectrometry (GC-MS) analysis showed that the polymers are formed by repeated glucose moieties bond by 1,6-linkages. Infrared spectroscopy revealed a dextran polysaccharide structure, while X-ray fluorescence determined that various other elements (Ca, Al, Fe, K, Cl, Mn, P and traces of Si and Ti) were also associated with the polymer, in amounts that varied considerably from sample to sample. X-ray diffraction analysis confirmed the polysaccharide dextran structure while identifying a certain amount of different (polysaccharide) material present in the canjica.  相似文献
