Pyrophosphate-dependent phosphofructokinase from pollen: properties and possible roles in sugar metabolism 总被引:1,自引:0,他引:1
To evaluate the role of pyrophosphate-dependent phosphofructokinase (PFP. EC 2.7.1.90) in the sugar metabolism of pollen. its occurrence and properties were studied in pollen grains of several plants including camellia ( Camellia japonica L.). In all pollen samples, PFP was strongly activated by fructose-2,6-bisphosphate (F2,6BP), and the activity of F2,6BP-activated PFP was higher than that of phosphofructokinase (PFK. EC 2.7.1.11). PFP partially purified from camellia pollen required Mg2+ for activity with an optimum at 1 m M . and was almost unaflected by a variety of metabolites at 1 m M . Its molecular mass was around 220 kDa, and apparent Km values for F6P, PPi . F1, 6BP and Pi were 294, 4, 20 and 580 u M , respectively. The levels of F2.6BP. PPi and F6P in camellia pollen were sufficent to support the forward reaction by PFP, and PFP, was 20- to 40-fold more active than PFK during pollen growth. These results suggest that pollen PFP plays a role in glycolysis but not gluconeogenesis. and the possible relevance of this to pollen tube growth is discussed. 相似文献

The tea plant (Camellia sinensis) is a thermophilic cash crop and contains a highly duplicated and repeat-rich genome. It is still unclear how DNA methylation regulates the evolution of duplicated genes and chilling stress in tea plants. We therefore generated a single-base-resolution DNA methylation map of tea plants under chilling stress. We found that, compared with other plants, the tea plant genome is highly methylated in all three sequence contexts, including CG, CHG and CHH (where H = A, T, or C), which is further proven to be correlated with its repeat content and genome size. We show that DNA methylation in the gene body negatively regulates the gene expression of tea plants, whereas non-CG methylation in the flanking region enables a positive regulation of gene expression. We demonstrate that transposable element-mediated methylation dynamics significantly drives the expression divergence of duplicated genes in tea plants. The DNA methylation and expression divergence of duplicated genes in the tea plant increases with evolutionary age and selective pressure. Moreover, we detect thousands of differentially methylated genes, some of which are functionally associated with chilling stress. We also experimentally reveal that DNA methyltransferase genes of tea plants are significantly downregulated, whereas demethylase genes are upregulated at the initial stage of chilling stress, which is in line with the significant loss of DNA methylation of three well-known cold-responsive genes at their promoter and gene body regions. Overall, our findings underscore the importance of DNA methylation regulation and offer new insights into duplicated gene evolution and chilling tolerance in tea plants. 相似文献

Camellia oleifera is believed to exhibit a complex intraspecific polyploidy phenomenon. Abnormal microsporogenesis can promote the formation of unreduced gametes in plants and lead to sexual polyploidy, so it is hypothesized that improper meiosis probably results in the formation of natural polyploidy in Camellia oleifera. In this study, based on the cytological observation of meiosis in pollen mother cells (PMCs), we found natural 2n pollen for the first time in Camellia oleifera, which may lead to the formation of natural polyploids by sexual polyploidization. Additionally, abnormal cytological behaviour during meiosis, including univalent chromosomes, extraequatorial chromosomes, early segregation, laggard chromosomes, chromosome stickiness, asynchronous meiosis and deviant cytokinesis (monad, dyads, triads), was observed, which could be the cause of 2n pollen formation. Moreover, we confirmed a relationship among the length–width ratio of flower buds, stylet length and microsporogenesis. 针对尺度对地物空间结构的限制以及传统分水岭分割易产生树冠过分割等问题,选择长沙县明月村油茶基地为研究区,提出一种基于多尺度标记优化分水岭分割油茶树冠的方法。首先使用高分辨率无人机影像采集图像,分析影像特征,构建油茶分类体系,提取油茶林分布区域。其次,运用多尺度区域迭代增长方法提取树冠标记,将标记应用于多阈值尺度的分水岭变换,并结合Johnson指数选取树冠标记增长和分水岭阈值的最优尺度,实现油茶单木的准确识别。结果表明:多尺度标记优化分水岭方法在分离油茶单木时,树冠面积提取值与目视解译参考值的相对误差为9.4%;单木总体识别精度为89.4%,相对于传统的分水岭分割方法精度提高了34.8%;通过Johnson指数确定的最优迭代增长尺度为20,分水岭分割阈值尺度为85,对比不同尺度组合下的油茶冠幅提取结果,最优尺度下的油茶冠幅提取精度最高(R2=0.75)。多尺度标记优化分水岭方法能较准确地分离油茶树冠,将该方法应用于无人机影像树冠分割,可有效提高经济林调查的效率。 相似文献

以普通油茶(Camellia oleifera) 4个无性系为材料,结合油茶成花的动态观察和花芽分化过程的石蜡切片形态观察,采用酶联免疫吸附分析法测定花芽中玉米素核苷(ZR)、脱落酸(ABA)、生长素(IAA)、赤霉素(GA) 4种内源激素含量,探讨油茶花芽分化与内源激素的关系。油茶花芽分化过程可分为6个时期:前分化期(10 d)、萼片形成期(20 d)、花瓣形成期(30 d)、雌雄蕊形成期(20 d)、子房与花药形成期(10 d)和雌雄蕊成熟期(20 d),历时3~4个月。油茶不同无性系的花芽分化时间略有不同。油茶花芽中ZR含量相对较低(5.102~16.412 ng·g–1 FW),ABA含量相对较高(76.815~137.648 ng·g–1 FW)。其中,粤华5号和湘林8号的ZR、ABA含量变化趋势一致,岑软3号和岑软2号含量变化趋势一致。油茶花芽中IAA含量相对较高,为49.072~135.622 ng·g–1 FW,随着花芽分化进程,IAA含量均呈先升后降再升的变化趋势。GA含量相对较低,为5.616~13.720 ng·g–1 FW,随时间变化,呈现出不断降低的趋势。其中,不同无性系的IAA、GA含量变化趋势一致,而ZR、ABA含量变化趋势有所差异。ZR有利于花器官形成;高浓度IAA促进油茶花芽分化,低浓度IAA有利于开花;花芽中IAA与ABA存在明显的颉颃作用;GA抑制花芽分化。 相似文献

利用生物信息学方法,从茶树(Camellia sinensis(L.)O.Ktze.)全基因组数据库中分析获得DELLA蛋白的家族成员,并对它们的系统进化关系、蛋白序列特征、基因表达特异性及其与茶树次生代谢物的相关性进行分析。结果显示:茶树基因组中共有5个DELLA基因,分别为:TEA009882(CsGAI)、TEA022818(CsRGA1)、TEA010112(CsRGL1)、TEA008736(CsRGL2)和TEA020933(CsRGL3);其编码的氨基酸数量在525~594之间,均定位于细胞核。该蛋白的二级和三级结构分析结果表明,茶树DELLA蛋白结构中含有大量的α螺旋及少量β转角结构。蛋白保守结构域分析结果显示该蛋白与拟南芥(Arabidopsis thaliana(L.)Heynh.)具有高度的同源性,均具有GRAS、DELLA等保守结构域。基因的表达特异性分析结果表明,在茶树不同组织部位中,TEA009882、TEA022818和TEA010112基因的表达量均较高,而TEA020933和TEA008736的表达量在各组织中均较低;茶树DELLA基因的表达受到干旱、NaCl、低温及茉莉酸甲酯等非生物逆境胁迫的调控,且其表达量与茶树次生代谢物的积累间存在相关性。推测茶树DELLA基因广泛参与了茶树生长发育及非生物逆境胁迫的响应,以及对次生代谢物生物合成过程的调控。 相似文献

Tea root lesion nematode, Pratylenchus loosi, is one of the most important crop pests in Iran, which causes loss in quantity and quality of tea. This study was carried out to identify the relationship between soil pH and population level of P. loosi. One hundred and eighty-three soil and root samples were taken randomly from all of the tea plantations in Iran. The nematodes were extracted from root samples with Coolen and d’Herd methods and from soil samples by sieving and centrifugation. Soil pH was measured by two methods: 1:1 dilution of soil:deionised water and 1:1 dilution of soil:dilute calcium chloride 0.01?M (Cacl2) solution. Range of mean population of P. loosi per gram of root was 0.66–884 and per 100?g of soil was 1–186 in all samples. The highest population of P. loosi was observed at pH 3.5–4.5 as if 71.33% of infested samples were observed at soil pH 3.5–4.5. The results of regression analyses showed that at pH ranges between 3.5 and 4.5, there is a correlation (P???0.05) between soil pH and mean population of nematode per gram of feeder roots. This study demonstrated that the population density and damage potential of this nematode likely increases at pH?<?4.5. 相似文献

The present study was performed to observe histopathological changes in tissues of Bithynia siamensis goniomphalos (Gastropoda, Bithyniidae) incubated in crude extract solutions of camellia (Camellia oleifera) seed and mangosteen (Garcinia mangostana) pericarp, and furthermore to estimate the molluscicidal effects of 2 plant substances. Substantial numbers of bithyniid snails were incubated in various concentrations of 2 plant solution for 24 hr. As the positive control, snails incubated in various concentrations of niclosamide, a chemical molluscicide, were used. The histopathological findings were observed in sectioned snail specimens of each experimental and control groups. The results showed that both camellia and mangosteen extracts had molluscicidal effects at 24 hr with 50% lethal concentration (LC50) at concentrations of 0.003 and 0.002 g/ml, respectively, while niclosamide had LC50 at concentrations 0.599 ppm. B. siamensis goniomphalos snail tissues (foot, gill, and digestive system) showed disruption of columnar muscle fibers of the foot, reduction of the length and number of gill cilia, numerous mucous vacuoles, and irregularly shaped of epithelial cells. Irregular apical and calciferous cells, dilatation of the digestive gland tubule, and large hemolymphatic spaces, and irregular apical surfaces, detachment of cilia, and enlargement of lysosomal vacuoles of epidermis were also shown in all groups. By the present study, it is confirmed that 2 plants, camellia and mangosteen, are keeping some substance having molluscicidal effects, and histopathological findings obtained in this study will provide some clues in further studies on their action mechanisms to use them as natural molluscicides. 相似文献

Eight chloroplast markers were developed from Japanese and snow camellia (Camellia japonica and C. rusticana). Six markers were based on mononucleotide repeats, while the other two resulted from indels of larger units. Polymorphisms were screened using 15 individuals from all over the Japanese archipelago, including C. japonica, C. japonica var. macrocarpa, C. japonica var. hozanensis, and C. rusticana. Polymorphisms within a single population were searched in 22 and 26 individuals of C. japonica and C. rusticana, respectively. The number of alleles per locus ranged from two to three, resulting in eight haplotypes, two of which were specific to C. rusticana. No polymorphisms were detected within a single population for both C. japonica and C. rusticana. The eight markers developed in the present study will be useful for analyzing the genetic diversity and tracing maternal origins of Japanese and snow camellias. 相似文献
