Habitat loss is the single greatest threat to persistence of the critically threatened California tiger salamander (Ambystoma californiense). To aid management plans that designate critical habitat for this species, I developed and characterized 21 tetranucleotide microsatellite markers using two native populations in Santa Barbara and Alameda Counties. Allelic variation and average heterozygosities were lower in the endangered Santa Barbara population (allele range 1–4, mean 2.4; H O = 0.308 H E = 0.288) compared with the threatened Alameda population (allele range 2–10, mean 6.7; H O = 0.712, H E = 0.722). In-depth population studies using these markers will provide vital information for plans to assign critical habitat that optimize gene flow among breeding populations, as well as for identifying non-native hybrid genotypes that threaten native A. californiense stocks. Beyond the conservation goals for A. californiense, the close phylogenetic relationships within the tiger salamander complex also suggest a broad utility for population studies using these markers.  相似文献

A sudden and nearly synchronous emergence of the red tide forming dinoflagellate Cochlodinium along more than 800 km of California coastline was initially observed in late summer 2004. Thereafter high cell concentrations have been detected on an annual basis. Here, we present quantitative and semi-quantitative data indicating that Cochlodinium was uncommon in the phytoplankton community in California prior to 2004 and is now persisting as a more regular component and one that seasonally can cause red tides. The quantitative portion of this study was primarily conducted in Monterey Bay, where cell densities reached at least 6 × 104 cells L−1 during the initial outbreak. A semi-quantitative comparison of California coastal counties by the California Department of Health Services (CDHS) was also made: of the 15 counties surveyed (most with multiple sites per county), cells were detected only from Los Angeles County in the south to San Mateo County in the central region (seven counties), but not in the northern part of the state (six counties). Two counties in the central region of the state, San Luis Obispo and Santa Cruz, displayed intense and frequent periods of elevated Cochlodinium cell abundances. Although not observed in the state-wide CDHS survey, we occasionally found cells in San Diego County with densities up to 2.7 × 104 cells L−1. Though these colonial dinoflagellates have been recognized in California for over 80 years, with several “blooms” recorded prior to 2004, the species’ geographic range and abundance in recent years suggest significant shifts in the nearshore phytoplankton community of this region of the eastern Pacific.  相似文献

Diversity of the filamentous green algae in the genus Spirogyra (Zygnematophyceae) was investigated from more than 1,200 stream samples from California. We identified 12 species of Spirogyra not previously known for California (CA), including two species new to science, Spirogyra californica sp. nov. and Spirogyra juliana sp. nov. Environmental preferences of the Californian species are discussed in the light of their restricted distribution to stream habitats with contrasting nutrient levels. We also investigated the systematic relationships of Spirogyra species from several continents using the chloroplast‐encoded genes ribulose‐1,5‐bisphosphate carboxylase/hydrogenase large subunit (rbcL) and the beta subunit of the ATP synthase (atpB). Californian species were positioned in most major clades of Spirogyra. The phylogeny of Spirogyra and its taxonomic implications are discussed, such as the benefits of combining structural and molecular data for more accurate and consistent species identification. Considerable infraspecific genetic variation of globally distributed Spirogyra species was observed across continental scales. This finding suggests that structurally similar species from distant regions may be genetically dissimilar and that Spirogyra may contain a large number of cryptic species. Correlating the morphological and genetic variation within the genus will be a major challenge for future researchers.  相似文献

Understanding the mechanisms that determine the distribution and abundance of plants is a major problem in ecology. However, very few studies have explored the factors controlling the abundance of columnar cacti throughout their range of distribution. In this paper, we describe the density and size structure of 26 populations of Pachycereus pringlei throughout its distribution range in the Sonoran Desert. Major differences in abundance were detected between island and mainland and peninsular areas, with islands sustaining significantly larger densities than mainland and peninsular populations. Within peninsular populations, the abundance was negatively associated with latitude and positively associated with annual and seasonal rainfall. In contrast, the abundance in mainland populations showed neither latitudinal trend nor an association with rainfall. In peninsular populations, mean height and basal diameter of branched plants showed a negative association with population density whereas mainland populations showed no significant association. None of the populations exhibited a population structure that fitted the log-normal distribution expected for young, growing populations with constant recruitment. Insular, peninsular and␣mainland populations showed a population structure with an uneven size distribution typical of populations experiencing regeneration pulses.  相似文献

Aspects of the biology and host range of Sibinia fastigiata Clark (Coleoptera: Curculionidae) were studied to assess its safety for release in Australia as a biological control agent of the weed Mimosa pigra L . (Mimosaceae) . Larvae feed on the seeds and adults on open flowers of their host . Adults oviposit on to immature seeds 3 mm long or less and hence seeds of this length and maturity were used in the host range tests and for rearing . Females are shown to avoid previously attacked seeds enhancing their effectiveness as seed destroyers . Survival of adults was higher when provided with open flowers . The host range was determined using laboratory control - choice oviposition tests on excised plant material and , in the field in the native range , no - choice oviposition tests on living plants , surveys of adults on plants , and breeding of insects from pods of plants of various legume species . The control - choice oviposition tests employed a new design in which the control plant alone was offered to the insects followed by a choice of test plants species . Other than M. pigra, only one plant species was acceptable for oviposition , the closely related M. asperata. Larval development also occurs on M. asperata and this host is occasionally used in the field . This insect was approved for release in March 1997 .  相似文献

PurposeThe purpose of this study was to design and develop a new range optimization for target and organs at risk (OARs) in dynamic adaptive proton beam therapy (PBT).MethodsThe new range optimization for target and OARs (RO-TO) was optimized to maintain target dose coverage but not to increase the dose exposure of OARs, while the other procedure, range optimization for target (RO-T), only focused on target dose coverage. A retrospective analysis of a patient who received PBT for abdominal lymph node metastases was performed to show the effectiveness of our new approach. The original plan (OP), which had a total dose of 60 Gy (relative biological effectiveness; RBE), was generated using six treatment fields. Bone-based registration (BR) and tumor-based registration (TR) were performed on each pretreatment daily CT image dataset acquired once every four fractions, to align the isocenter.ResultsBoth range adaptive approaches achieved better coverage (D95%) and homogeneity (D5%−D95%) than BR and TR only. However, RO-T showed the greatest increases in D2cc and Dmean values of the small intestine and stomach and exceeded the limitations of dose exposure for those OARs. RO-TO showed comparable or superior dose sparing compared with the OP for all OARs.ConclusionsOur results suggest that BR and TR alone may reduce target dose coverage, and that RO-T may increase the dose exposure to the OARs. RO-TO may achieve the planned dose delivery to the target and OARs more efficiently than the OP. The technique requires testing on a large clinical dataset.  相似文献

Dendrochronological studies of large and old Sequoia sempervirens are limited by access and complex crossdating, but core sampling at regular height intervals along the main trunks of five standing trees allowed for reconstruction of growth, height, and age while providing within-tree replication for crossdating. We developed a crossdated ring-width chronology (1453–2015) for redwoods growing in an easternmost old-growth forest in the Napa Range of California, determined aboveground tree attributes, investigated the inter-annual climate-growth relationships since the late 19th century, and documented long-term growth trends. Age, height, f-DBH (functional diameter at breast height), and aboveground biomass of these co-dominant trees ranged from 241 to 783 years, 45.7 to 61.5 m, 117.0 to 226.9 cm, and 9.34 to 33.62 Mg, respectively. Bootstrapped correlation and response function analysis showed radial growth positively related to May through August Palmer Drought Severity Index (PDSI) and negatively related to maximum June temperature (r ≥ │0.47│, P < 0.0001), explaining 33.3% of ring-width variation. Bootstrapped correlations over a moving 40-year window indicated strengthening relationships with PDSI and minimum temperature. The long-term growth trend, reflected by the size-detrended metric of residual wood volume increment (RWVI), varied over time and showed an average one-year decrease of 13.3% for 20th and 21st century droughts. A fire detected in August 1931 corresponded with a one-year decrease in RWVI of 43.1% followed by >100% increase within five years. Growth dynamics for redwoods in this interior forest provide a point of comparison for redwoods previously studied in old-growth forests along the latitudinal gradient, highlighting range-wide trends and site-specific differences in responses to climate and fire.  相似文献

Ligaments assist trunk muscles in balancing external moments and providing spinal stability. In absence of the personalized material properties for ligaments, finite element (FE) models use dispersed data from the literature. This study aims to investigate the relative effects of eight different ligament property datasets on FE model responses. Eight L4-L5 models distinct only in ligament properties were constructed and loaded under moment (15 N m) alone or combined with a compressive follower load (FL). Range of motions (RoM) of the disc-alone model matched well in vitro data. Ligament properties significantly affected only sagittal RoMs (∼3.0–7.1° in flexion and ∼3.8–5.8° in extension at 10 N m). Sequential removal of ligaments shifted sagittal RoMs in and out of the corresponding in vitro ranges. When moment was combined with FL, center of rotation matched in vivo data for all models (3.8 ± 0.9 mm and 4.3 ± 1.8 mm posterior to the disc center in flexion and extension, respectively). Under 15 N m sagittal moments, ligament strains were often smaller or within the in vitro range in flexion whereas some posterior ligament forces approached their failure forces in some models. Ligament forces varied substantially within the models and affected the moment-sharing and internal forces on the disc and facet joints. Intradiscal pressure (IDP) had the greatest variation between models in extension. None of the datasets yielded results in agreement with all reported measurements. Results emphasized the important role of ligaments especially under larger moments and the need for their accurate representation in search for valid spinal models.  相似文献
