Human serum albumin (HSA) is an essential protein for maintaining human health. Accurate detection and quantification of HSA are of great significance for disease diagnosis and biochemical research. Here, a new HSA fluorescent probe BNPE based on the 1,8-naphthalimide fluorophore was designed and synthesized. The probe could recognize HSA through a twisted intramolecular charge transfer mechanism, effectively avoid the interference of most substances, and realize HSA fluorescence imaging in living cells.  相似文献

摘要 目的:探讨磁共振成像(MRI)弥散张量成像(DTI)参数联合血清神经元特异性烯醇化酶(NSE)、脂蛋白相关磷脂酶A2(Lp-PLA2)在脑梗死患者的诊断和预后不良风险评估中的应用价值。方法:选择2021年3月至2022年9月吉林大学中日联谊医院收治的106例脑梗死患者作为脑梗死组,另选同期62例体检健康志愿者作为对照组,比较两组DTI参数,血清NSE和Lp-PLA2水平。脑梗死组出院90d后,采用改良Rankin量表(mRS)进行预后评估,分为预后良好组与预后不良组,并比较两组上述指标水平。受试者工作特征(ROC)曲线分析DTI参数联合血清NSE、Lp-PLA2诊断脑梗死和预测脑梗死患者预后的价值。结果:脑梗死组表观弥散系数(ADC)值、部分各向异性指数(FA)值低于对照组(P<0.05),血清NSE、Lp-PLA2水平高于对照组(P<0.05)。预后不良组FA值、ADC值低于预后良好组(P<0.05),血清NSE、Lp-PLA2水平高于预后良好组(P<0.05)。联合FA值、ADC值、NSE和Lp-PLA2诊断脑梗死以及预测脑梗死患者预后不良的曲线下面积(AUC)分别为0.852、0.874,均高于各因素单独诊断和预测。结论:脑梗死DTI参数FA值、ADC值降低,血清NSE、Lp-PLA2水平增高,联合DTI参数和血清NSE、Lp-PLA2检测在脑梗死诊断和预后预测中具有较高价值。  相似文献

Bone scintigraphy is a nuclear imaging scan using a radiopharmaceutical composed of a bisphosphonate coupled to a radionuclide (technetium 99m). Radiopharmaceutical uptake is particularly important at the level of the bone structures having a strong osteoblastic activity. These uptakes can be due to a benign pathology (fracture, loosening of prosthesis, rheumatic pathologies, etc.) or to a malignant pathology (primary or secondary bone lesion). The high sensitivity of bone scintigraphy makes it particularly interesting at the initial stage of the pathology, especially when X-rays are normal. In addition, its specificity has clearly improved in recent years with the increasingly use of tomoscintigraphy coupled with X-ray scanning (SPECT/CT). We describe the operating principle of bone scintigraphy, normal uptakes with its variants as well as pathological uptake features in traumatic, rheumatic, prosthetic or cancerous pathologies.  相似文献

Synovial sarcoma (SS) is a mesenchymal tumor which generally affects the soft tissues of the extremities. Primary pleural synovial sarcoma (PPSS) is a very rare and aggressive subtype of SS. A 73-year-old male patient presented with chest-back pain and dyspnea. Hypermetabolic diffuse pleural lesions were detected in 18F-FDG PET/CT performed after pleural nodular thickenings were observed on CT. As the result of the molecular analysis performed in the excisional biopsy, SYT-SSX mutation was detected and the patient was diagnosed as SS. Pazopanib treatment was commenced. We are reporting a very rare case of PPSS with diffuse pleural involvement.  相似文献

Recently, we selected a novel anti-hPD-L1-specific HCAb named Nb6 with high affinity (EC50 = 0.65 ng/mL) for potential hPD-L1 targeted non-invasive PET imaging. In this research, Nb6 was conjugated with the bifunctional chelator NCS-Bz-NOTA ((2-[(4-Isothiocyanophenyl) methyl]-1,4,7-triazacy-clononane-1,4,7-triacetic acid)) and further labeled with radio-nuclide 64Cu. 64Cu-NOTA-Nb6 was prepared with over 95% labeling yield, over 99% radiochemical purity and 14–16 GBq/μmol specific activity after PD-10 column purification. It shows good stability in 0.01 M PBS and 5% HSA solutions. 64Cu-NOTA-Nb6 has a high binding affinity to 3.60 nM which was tested by human lung adenocarcinoma A549 cell lines. Tumor lesion can be clearly observed from 20 h to 38 h by Micro-PET equipment after 64Cu-NOTA-Nb6 administration. The study revealed that 64Cu-NOTA-Nb6 has good lesion detection ability, high ratios between tumor and non-tumor signal and can specifically target A549 xenografted tumor model. Taken together of good stability, high binding affinity, and tumor detection ability, 64Cu labeled Nb6 is a promising radio-tracer in diagnosing of hPD-L1 overexpression tumor, supposed to monitor PD-L1overexpression tumor progression and guide targeted therapy with PET molecular imaging.  相似文献

Recently, oxytocin (OT) has been studied as a potential modulator of endogenous analgesia by acting upon pain circuits at the spinal cord and supraspinal levels. Yet the detailed action mechanisms of OT are still undetermined. The present study aimed to evaluate the action of OT in the spinal cord dorsal horn network under nociceptive-like conditions induced by the activation of the N-methyl-d-aspartate (NMDA) receptor and formalin injection, using calcium imaging techniques. Results demonstrate that the spontaneous Ca2+-dependent activity of the dorsal horn cells was scarce, and the coactivity of cells was mainly absent. When NMDA was applied, high rates of activity and coactivity occurred in the dorsal horn cells; these rates of high activity mimicked the activity dynamics evoked by a neuropathic pain condition. In addition, although OT treatment increased activity rates, it was also capable of disrupting the conformation of coordinated activity previously consolidated by NMDA treatment, without showing any effect by itself. Altogether, our results suggest that OT globally prevents the formation of coordinated patterns previously generated by nociceptive-like conditions on dorsal horn cells by NMDA application, which supports previous evidence showing that OT represents a potential therapeutic alternative for the treatment of chronic neuropathic pain.  相似文献

Nitrogen doped carbon dots (N-CDs) are well documented as an outstanding fluorogenic material for protein tags, live cell imaging and protein-receptor based fluorescence sensors owing to its good optical features with less cytotoxicity and better water solubility. In this regard, the present work describes the synthesis of nitrogen rich blue fluorescent carbon dots (NR-CDs) through hydrothermal treatment of citric acid monohydrate (CA) and 2-aminopyridine (2-AP). The optical properties of NR-CDs are further analyzed by common analytical methods viz., Fourier transform infrared (FT-IR), UV–visible (UV–vis) and Fluorescence spectroscopies. The surface chemical composition and morphology of NR-CDs are acquired by X-ray photo electron spectroscopy (XPS) and high resolution transmission electron microscopy (HR-TEM), respectively. The NR-CDs produce blue fluorescent at 421 nm at the excitation wavelength of 310 nm, the calculated quantum yield is about 18% with respect to standard quinine sulfate. The synthesized NR-CDs contains 15.03 wt % of N revealed by XPS results. Further, the NR-CDs are used as a fluorescence staining agent for cell imaging of Candida albicans (C. albicans) and the cytotoxicity are also measured. All the outcomes proposed that the NR-CDs act as good staining agent for C. albicans with less cytotoxicity.  相似文献

PurposeThe objective of this study was to assess synthesized effective atomic number (Zeff) values with a new developed tissue characteristic phantom and contrast material of varying iodine concentrations using single-source fast kilovoltage switching dual-energy CT (DECT) scanner.MethodsA newly developed multi energy tissue characterisation CT phantom and an acrylic phantom with various iodine concentrations of were scanned using single-source fast kilovoltage switching DECT (GE-DECT) scanner. The difference between the measured and theoretical values of Zeff were evaluated. Additionally, the difference and coefficient of variation (CV) values of the theoretical and measured values were compared with values obtained with the Canon-DECT scanner that was analysed in our previous study.ResultsThe average Zeff difference in the Multi-energy phantom was within 4.5%. The average difference of the theoretical and measured Zeff values for the acrylic phantom with variation of iodine concentration was within 3.3%. Compared to the results for the single-source Canon-DECT scanner used in our previous study, the average difference and CV of the theoretical and measured Zeff values obtained with the GE-DECT scanner were markedly smaller.ConclusionsThe accuracy of the synthesized Zeff values with GE-DECT had a good agreement with the theoretical Zeff values for the Multi-Energy phantom. The GE-DECT could reduce the noise and the accuracy of the Zeff values than that with Canon-DECT for the varying iodine concentrations of contrast medium.Advances in knowledgeThe accuracy and precision of the Zeff values of the contrast medium with the GE-DECT could be sufficient with human equivalent materials.  相似文献
