We have investigated the localization of atrial natriuretic factor (ANF)-like immunoreactivity in the central nervous system of the cartilaginous fish, Scyliorhinus canicula, using the indirect immunofluorescence technique. Immunoreactive perikarya and fibers were observed in two regions of the telencephalon, the area superficialis basalis and the area periventricularis ventrolateralis. In the diencephalon, the hypothalamus exhibited a moderate number of ANF-containing neurons and fibers located in the preoptic and periventricular nuclei and in the nucleus lateralis tuberis. The most important group of ANF-immunoreactive cells was observed in the nucleus tuberculi posterioris of the diencephalon. In contrast, the mesencephalon showed only a few ANF-positive nerve processes located in the tegmentum mesencephali. Numerous fine fibers and nerve terminals were found in the dorsal area of the neurointermediate lobe of the pituitary. These results provide the first evidence for the presence of ANF-related peptides in the brain of a cartilaginous fish. The widespread distribution of ANF-positive cells and fibers in the brain and pituitary suggests that this peptide may act both as a neurotransmitter and (or) a neurohormone in fish. 相似文献

Summary Previous studies have shown that cell density can regulate the binding of several growth factors. To determine whether cell
density exerts a uniform effect on the expression of epidermal growth factor (EGF) receptors, seven cell lines were examined
in detail. For each cell line, EGF binding was found to decrease as cell density increases. Scatchard analysis of the binding
data reveals that decreases in EGF binding are due to reductions in the number of cell surface EGF receptors. The only apparent
exception is the effect of cell density on the binding of EGF to A-431 cells. For these cells, increases in cell density lead
to two effects: decreases in the number of high affinity EGF receptors and increases in the total number of EGF receptors.
In addition to the effects of cell density on EGF receptors, it was determined that increases in cell density can coordinately
down-regulate receptors for as many as four different growth factors. Overall, the findings described in this report for EGF
and those previously described for transforming growth factor type-β (TGF-β) and fibroblast growth factor (FGF) demonstrate
the existence of a common mechanism for down-regulating growth factor receptors.

Summary Weibel-Palade bodies are ultrastructurally defined organelles found only in vascular endothelial cells. Because endothelium
in corpo is very dispersed, isolation and further characterization of this organelle has been dependent on increasing the
number of cells in culture. However, primary isolates of endothelial cells have a limited replication potential and tend to
senesce in culture. In this report, EA.hy926, a continuously replicating cell line derived from human endothelium, is shown
to contain Weibel-Palade bodies. Electron micrographs demonstrate the ultrastructural characteristics of these tissue-specific
organelles and their cytoplasmic distribution in EA.hy926 cells. Von Willebrand factor, which has been shown to exist in Weibel
Palade bodies, is demonstrated by immunofluorescence in discrete rod-shaped organelles whose size, shape, and distribution
are consistent with that of Weibel-Palade bodies in primary endothelial cell cultures. Rapid release of von Willebrand factor
can be induced by calcium ionophore, and large multimeric forms of the protein are found in EA.hy926 cells. These two properties
are consistent with the function currently ascribed to Weibel Palade bodies: storage of multimerized von Willebrand factor.
Thus ultrastructural, immunologic, and functional data establish the existence of this as yet poorly understood tissue-specific
organelle in a continuous, vigorously replicating human cell line. 相似文献

Summary The adult mouse submandibular salivary gland provides a good model system to study gene regulation during normal and abnormal
cell behavior because it synthesizes functionally distinct products ranging from growth factors and digestive enzymes to factors
of relevance to homeostatic mechanisms. The present study describes the long-term growth and differentiation of submandibular
salivary epithelial cells from adult male mice as a function of the culture substratum. Using a two-step partial dissociation
procedure, it was possible to enrich for ductal cells of the granular convoluted tubules, the site of epidermal growth factor
synthesis. Long-term cell growth over a period of 2 to 3 mo. with at least 3 serial passages was obtained only within three-dimensional
collagen gels. Cells grew as ductal-type structures, many of which generated lumens with time in culture. Electron microscopic
analysis in reference to the submandibular gland in vivo revealed enrichment for and maintenance of morphologic features of
granular convoluted tubule cells. Reactivity with a keratin-specific monoclonal antibody established the epithelial nature
of the cells that grew within collagen. Maintenance of cell differentiation, using immunoreactivity for epidermal growth factor
as criterion, was determined by both cytochemical and biochemical approaches and was found to be dependent on the collagen
matrix and hormones. Greater than 50% of the cells in primary collagen cultures contained epidermal growth factor only in
the presence of testosterone and triiodothyronine. In contrast, cells initially seeded on plastic or cycled to plastic from
collagen gels were virtually negative for epidermal growth factor. Biochemical analysis confirmed the presence of a protein
with an apparent molecular weight of 6000 which comigrated with purified mouse epidermal growth factor. Epidermal growth factor
was also present in detectable levels in Passage 1 cells. This culture system should permit assessment of whether modulation
of submandibular gland ductal cell growth can be exerted via a mechanism that in itself includes epidermal growth factor and
its receptor and signal transduction pathway.

The population of bream in L. Peipsi was studied with respect to age, growth rate, condition factor (according to Fulton) and length-weight relationship in 1994. That autumn the bream population in L. Peipsi consisted of fishes aged from 0+ to 15+. During the first year bream reached an average body length of 7.9 cm (the commercial legal size (30 cm) was usually attained by the end of the 5th–6th year. The condition of bream in this lake was above the average of Estonian lakes. The relatively good growth rate and condition of bream in the lake indicates that the waterbody is appropriate for this fish. 相似文献

Lipopolysaccharide (LPS) has been recognized as a potent antitumor agent in animal tumor models; however, its use in human
cancer therapy has been limited to only one trial, in which LPS from Salmonella was given intravenously. It was not very successful because of poor tumor response and was also toxic. We originally developed
LPS prepared from Pantoea agglomerans (LPSp), and this was a well-purified, small-molecular-mass (5 kDa) agent. We chose intradermal rather than intravenous administration
in the hope that the former would release LPS slowly into the bloodstream, and thus be less toxic while preserving antitumor
activity. In our animal tumor models, intradermal administration was indeed less toxic and more beneficial for tumor regression
than intravenous administration. We made a pilot study with intradermal administration of LPSp on the treatment of ten advanced
cancer patients. Five of them had evaluable tumor, which had failed earlier to respond to conventional chemotherapy. Cyclophosphamide
was also administered in this trial, in anticipation of its synergistic effect with LPSp. In this study LPSp was injected
intradermally into each patient twice a week, starting with an initial dose of 0.4 ng/kg, and raising it to 600 or 1800 ng/kg.
A 400-mg/m2 dose of cyclophosphamide was given intravenously every 2 weeks. After completion of the dose escalation, the treatment was
continued for at least 4 months, and it was found that 1800 ng/kg LPSp was well tolerated. A significant level of cytokines
was observed in the sera for at least 8 h. These results indicate higher tolerable doses and remarkably more continuous induction
of the cytokines than were reported in a previous study by others using intravenous administration. Three of the five evaluable
tumors showed a significant response to our combined therapy. Intradermally administered, LPS was less toxic and elicited
a tumor response in combination with cyclophosphamide; it can thus can be applied to cancer treatment even in humans.

Reactivation of herpes simplex virus type 1 (HSV-1) occurred rapidly in cells of latently infected adult mouse trigeminal ganglia which were cultured in serum-free medium in the presence of sufficient nerve growth factor (NGF). However, HSV-1 reactivation was delayed significantly in ganglionic cultures in the absence of exogenous NGF or in cultures treated with 2-aminopurine in the presence of NGF. The delayed viral reactivation in ganglionic cultures without NGF was accelerated by treatment with phorbol myristate acetate or dibutyryl cyclic AMP. Culture conditions which affected HSV-1 reactivation did not affect replication of HSV-1 in normal ganglionic cultures. 相似文献
