‘晚大新高’梨授粉及受精过程的显微动态研究   总被引:4,自引:1,他引:3  
应用荧光显微法和石蜡切片解剖法对‘晚大新高’梨授粉受精过程进行了系统观察研究。结果表明:‘晚大新高’梨自花授粉不结实;异花最佳授粉品种为‘黄花’,其次为‘翠冠’和‘丰水’。与选用‘黄花’为异花授粉品种相比,自花和异花的授粉受精过程存在明显差异,自花花粉在授粉后2h开始萌发,8h花粉管生长至离柱头约1/3处停止生长,顶端膨大呈球形,表现出自交不亲和性;异花花粉在授粉后1h开始部分萌发,8h花粉管生长至花柱中部,24h到达花柱基部并进入子房,48h进入胚囊,72h完成双受精过程。  相似文献

Anthracnose disease was detected from dragon fruit (Hylocereus undatus) at a market of Yuanjiang County, Yunnan Province, China. The results of pathogenicity test, morphology studies and sequence analyses based on ITS and β‐tubulin loci indicated that the disease was caused by Colletotrichum truncatum. The pathogen produced elliptic, yellow spots with chlorotic halos on the surface of the fruit, and the lesion become depressed gradually. Grey to black acervuli appeared on the lesion surface in concentric circles later. This is the first report of dragon fruit anthracnose caused by this pathogen in China.  相似文献

We investigated how the plant‐inhabiting, anthocorid predator, Anthocoris nemoralis, copes with variation in prey, host plant and associated herbivore‐induced plant volatiles and in particular whether the preference for these plant odours is innate or acquired. We found a marked difference between the olfactory response of orchard‐caught predators and that of their first generation reared on flour moth eggs in the laboratory, i.e. under conditions free of herbivory‐induced volatiles. Whereas the orchard‐caught predators preferred odour from psyllid‐infested pear leaves, when offered against clean air in a Y‐tube olfactometer, the laboratory‐reared first generation of (naive) predators did not. The same difference was found when a single component (methyl salicylate) of the herbivore‐induced plant volatiles was offered against clean air. After experiencing methyl salicylate with prey, however, the laboratory‐reared predators showed a pronounced preference for this volatile. This acquired preference did not depend on whether the volatile had been experienced in the juvenile period or in the adult phase, but it did depend on whether it had been offered in presence or absence of prey. In the first case, they were attracted to the plant volatile in subsequent olfactometer experiments, but when the volatile had been offered during a period of prey deprivation, the predators were not attracted. We conclude that associative learning is the most likely mechanism underlying acquired odour preference.  相似文献

对药食同源植物苦瓜(Momordica charantia L.)的鲜汁冻干粉对动物的降血糖作用及急性毒性进行了药理实验,苦瓜冻干粉(生药量20g/kg和40g/kg)对药物性高血糖小鼠具有显著的降血糖作用,同时对正常小鼠血糖没有影响;苦瓜冻干粉的小鼠口服LD50为生药量704.8g/kg(95%可信限生药量618.4~804g/kg),其产生降糖药效的剂量仅为其LD50的1/35~1/17。  相似文献

Stylar soluble proteins in self-incompatible “Nijisseiki” (S2S4), self-compatible “Osa-Nijisseiki” ( S2SSM4, SM means stylar-part mutant) and its progeny were analyzed by isoelectric focusing polyacrylamide gel electrophoresis (IEF-PAGE).SSM4-allele associated protein, SSM4-protein, existed in the style of “Osa-Nijisseiki” and its progeny. The SSM4-protein expressed only in the stigma of “Osa-Nijisseiki”, whereas in its original variety “Nijisseiki”, S4-protein expressed in the upper and lower parts of the style as well as in the stigma, and its expression amount decreased from the upper part to the lower part. The protein bands analyzed by IEF-PAGE were subjected to RNase activity staining. The results showed that the S4- and the SSM4-proteins have the similar molecular weights (approximately 30 kD) and RNase activity. The specific-activities measured with yeast RNA were similar, equivalent to approximately 275 U·min-1·mg-1 protein. The S SM4-protein showed almost the same inhibitory effects as the S4-protein on the pollen germination and the pollen tube growth with S4- and SSM4-alleles in vitro . From the above results, the reasons of the self-compatibility of “Osa-Nijisseiki” are considered as (1) low expression of the SSM4-gene and (2) the SSM4-gene expression only in stigma.  相似文献

为研究‘砀山酥梨’及其褐皮芽变木葡聚糖转葡糖苷酶基因(PbXET)表达水平差异,该实验利用RACE技术,克隆了梨PbXET基因;采用实时荧光定量PCR技术,分析了梨树叶片、果皮和果肉等不同组织及花后不同时期果皮中PbXET基因表达差异。结果表明:(1)梨PbXET3(KJ690921)和PbXET4(KJ690922)开放阅读框分别为903bp和891bp,分别编码300和296个氨基酸;氨基酸序列聚类分析显示,PbXET3与苹果MdXET-3以及PbXET4与苹果MdXET-5的亲缘关系最近。(2)半定量PCR分析显示,花后150d,PbXET3和PbXET4基因在‘砀山酥梨’和‘锈酥’不同组织中均有表达,且PbXET3在叶片中表达量很低,在果皮、果肉中表达相对较强,其中叶片中PbXET3表达量低于PbXET4,而果肉和果皮中PbXET3的表达均明显高于PbXET4。(3)荧光定量PCR分析发现,在‘砀山酥梨’和‘锈酥’果皮中,PbXET3和PbXET4基因不同时期的表达量变化趋势不同;与‘砀山酥梨’相比,果皮颜色发生变化(花后100d)之后,‘锈酥’果皮中PbXET3表达量骤减;而果皮颜色发生变化(花后100d)之前,PbXET4表达量均显著降低。由此推测,PbXET3和PbXET4基因参与了‘锈酥’果皮褐色形成的调节,其表达水平差异可能是改变‘锈酥’表皮细胞结构的重要原因之一。  相似文献

根据鸭梨多酚氧化酶基因序列设计引物,PCR扩增该基因3′端450bp的片段,并将该片段反向插入真核表达载体pB I121的CaMV 35S启动子和NOS终止子之间,首次构建了鸭梨PPO基因的反义表达载体;其后,在农杆菌EHA105的介导下,成功实现了PPO反义基因对鸭梨组培苗的遗传转化。经Northern杂交和酶活检测证实,转基因鸭梨植株体内的多酚氧化酶基因转录和翻译水平均得到明显抑制,从而为耐褐化梨新品种的培育奠定了基础。  相似文献

Summary Mesophyll protoplasts of wild pear (Pyrus communis var. pyraster L., Pomoideae) were chemically fused with cell suspension protoplasts of cherry rootstock Colt (Prunus avium x pseudocerasus, Prunoideae), following an electroporation treatment of the separate parental protoplast systems. Fusion-treated protoplasts were cultured, on modified K8P medium, where it had been previously established that neither parental protoplasts were capable of division. Somatic hybrid calli were recovered and, following caulogenesis on MS medium with zeatin and after rooting of regenerated shoots, complete trees were obtained and grown in vivo. Hybridity of these trees was confirmed based on morphological characters, chromosome complement and isozyme analysis. Two separate cloned lines of this intersubfamilial rootstock somatic hybrid (wild pear (+) Colt cherry) were produced. This is the first report of the production of somatic hybrid plants of two woody species, of agronomic value, within the order Rosales.  相似文献

梨果实是由下位民衍生,有发达的肉质果壁,是一种假果。不同品种梨果实结构有一;定判别表现在果实表现和果肉细胞及细胞内物质成分的不同。果实角质厝较厚,且后期深入表皮细胞间隙,表皮下4~5层细胞2含有大量易被染色的单宁类物质,定藏,角质膜较薄,表皮下细胞不含单宁类物质,不耐贮藏。不同品种梨果实中石细胞团的大小不同,糖分含含量也存在一定差异。石细胞团的大小与果肉细腻程度及糖分含量呈负相关,是影响果实品质的  相似文献
