生发泡迁移(GVM)是大多数两栖类动物中卵母细胞成熟之前都可以观察到的、涉及细胞核行为的现象。本实验在光镜水平上对激素诱导下的黑眶蟾蜍卵母细胞的GVM现象、以及细胞骨架解聚剂类药物———秋水仙素、细胞松弛B(CB)对这种激素诱导作用的影响进行了研究。同时,采用AZAN染色法观察了GVM过程中生发泡周边纤维骨架的结构变化。将取自刚脱离冬眠期雌体的卵母细胞按不同的培养液、分三个实验组,体外培养不同的时间后,固定、染色、观察。对照组培养液成分为Ringer液中加入人绒毛膜促性腺激素和脑垂体;实验组分别增加秋水仙素或CB。Tab.1和PlateI1,4,5,6,7,8,9表明:经过体外培养4h,各组生发泡均向动物极表面发生了迁移。但是,秋水仙素的作用在培养的前2h,对GVM表现为促进效应(PlateI5);而培养的后2h,却表现为抑制(PlateI8);CB的作用始终是抑制(PlateI6&9)。6h后,各组生发泡均告破裂(PlateI10,11,12)。正常情况下,生发泡周围被一环形纤维包围,其外侧有两个纤维化小体(PlateI2)。发育较快者,纤维化小体消失,植物极附近纤维逐渐加厚(PlateI2  相似文献

The specific activity of three characteristic enzymes, adenylate deaminase, adenylate kinase, and creatine kinase, in the skeletal muscles and heart of a variety of vertebrate land animals, including the human, are surveyed. Data from this study and available studies in the literature suggest that adenosine monophosphate deaminase in land vertebrates is quite high in white skeletal muscle, usually somewhat lower in red muscle, and 15-to 500-fold lower in cardiac muscle. Adenosine monophosphate deaminase is active primarily under ischemic or hypoxic conditions which occur frequently in white muscle, only occasionally in red muscle, and ought never occur in heart muscle, and this may therefore account for observed enzyme levels. The common North American toad, Bufo americanus, provides a striking exception to the rule with cardiac adenosine monophosphate deaminase as high as in mammalian skeletal muscle, whereas its skeletal muscle level of adenosine monophosphate deaminase is several times lower. The exceptional levels in the toad are not due to a change in substrate binding and are not accompanied by comparable change in the level of adenylate or creatine kinase. Nor do they signal any major change in isozyme composition, since a human muscle adenosine monophosphate deaminase-specific antiserum reacts with toad muscle adenosine monophosphate deaminase, but not with toad heart adenosine monophosphate deaminase. They do not represent any general anuran evolutionary strategy, since the bullfrog (Rana catesbeiana) and the giant tropic toad (Bufo marinus) have the usual vertebrate pattern of adenosine monophosphate deaminase distribution. The distribution and colocalization of neuropeptides and 5-hydroxytryptamine in the posterior portion of the large intestine of the toad was studied using single- and dual-label immunohistochemistry. Neurons containing colocalized galanin/somatostatin or vasoactive intestinal peptide alone were observed along intramural pelvic nerves. Some of the galanin/somatostatin neurons also contained 5-hydroxytryptamine. Synaptic boutons containing colocalized calcitonin gene-related peptide/vasoactive intestinal peptide were associated with the galanin/somatostatin neurons. The muscle of the large intestine was also innervated by axons containing galamin/somatostatin, vasoactive intestinal peptide/calcitonin gene-related peptide or vasoactive intestinal peptide alone. Nerve fibres containing calcitonin gene-related peptide/substance P, probably representing primary afferent nerves, were also associated with muscle bundles. Submucosal blood vessels carried dense plexuses of fibres containing vasoactive intestinal peptide alone or and calcitonin gene-related peptide/substance P. Adrenergic perivascular nerves also contained galanin and neuropeptide Y.  相似文献

Summary The pulmonary artery of Bufo marinus contains large numbers of bipolar cells situated in the tunica adventitia and in the outer layers of the media. These cells show a bright green-yellow fluorescence (emission spectra 485 nm) after formaldehyde pre-treatment suggesting that they contain a primary monoamine. The most characteristic fine-structural feature of these cells is the presence of numerous dense-cored vesicles (80—300 nm diameter) in their cytoplasm. The cells are in close contact (20 nm gap) with both agranular and granular nerve fibres. Both EM-cytochemical and formaldehyde-induced fluorescence tests indicate that the granule-containing nerve fibres are adrenergic. The agranular nerve fibres form discrete synaptic contacts with pre-and post-synaptic membrane thickenings on the cells. This was never observed with respect to the adrenergic fibres. Each process of the cells is about 45 m long. The processes do not bear any special relationship to either vessels of the arterial vasa vasorum or medial smooth muscle cells. Their location in the wall of the artery suggests that they are functionally significant with respect to activity of the arterial media.  相似文献

Relationships among 14 species of North American Bufo were studied using the quantitative immunological technique of micro-complement fixation. Separate antisera to serum albumins of five North American Bufo and one Central American Bufo were used as probes of phylogenetic relationships. A cladogram for these species constructed with the albumin data was compatible with phylogenetic interpretations based on morphological analyses and hybridization studies. Using albumin as a molecular dating device radiations within the B. americanus, B. cognatus and B. boreas species groups were each estimated to be Pleistocene events. Separation of North and Central American lineages is estimated to have occurred as early as the upper Oligocene.  相似文献

In an ongoing long-term study of a breeding population of common toads Bufo bufo at a pond in southern England, the dates of first spawning have been recorded since 1980, the dates of toadlet emergence since 1984, and the numbers of emergent toadlets estimated since 1988. The dates when spawn was first laid varied considerably between years from the earliest on 2 February 1993 (day 33) to the latest on 19 March 1996 (day 79). The duration of the tadpole stage was negatively correlated with the date of appearance of first spawn and was up to 30 days longer in early spawning years than in late ones. Despite this, toadlets still emerged from the natal pond up to 36 days earlier in early spawning years than in late ones. Significant positive correlations were found between the duration of the tadpole stage and both the proportion of days during the tadpole stage when the minimum ground temperature was at or below 0°C and the proportion of days when 10 mm or more rain fell. Tadpole mortality was positively correlated with the proportion of days during the tadpole stage when the minimum ground temperature was at or below 1.5°C. Evidence was also found to suggest that tadpole mortality was density-dependent, being proportionately higher when initial tadpole numbers were high than when they were low. We sampled litter frogs in an 1800-ha mid-elevation seasonal forest in southeastern Brazil. One hundred 8 * 8-m plots were sampled during the dry/cold season and wet/warm season (unburned areas); we also examined the effects of fire in recently burned areas. A total of 267 frogs (305 g), belonging to 16 species (4 families) were caught. A single species comprised 78.5 percent of the individuals in the dry/cold season and 54.3 percent in the wet/warm season. The density of individuals did not change significantly with season, biomass did. Density and biomass of frogs were positively correlated with altitude. A Mantel test indicated that biological data (species and their abundance) were significantly associated with environmental parameters. The burned areas showed low values in richness, density and biomass of frogs. Harsh seasonal climate and a history of human disturbance may produce the low observed diversity values. The greater densities of frogs in sites of higher elevation may primarily result from mist-generated humidity, which diminishes the harshness of the dry/cold season in relation to lower sites.  相似文献

A computer simulation is performed of allele frequency changes resulting from genetic drift at eleven loci during the probable course of colonization of Australia by populations of the Giant Toad, Bufo marinus. The history of twelve populations for which allele frequency data are available is modelled. Account is taken of the likely pattern of relationship among the populations, the effective size of the populations (as indicated by the observed variance of allele frequencies) and the probable isolation of the populations from each other following their separation. In all simulations, allele frequencies at some loci show a significant association with latitude while others do not. In six of ten simulations, there is a significant association between degree of variation at a locus and the presence of a latitudinal cline of allele frequencies. There are also indications of this kind of association in simulations of a uni-directional range expansion. These results demonstrate that such associations, which were also observed in the data from the actual populations, can result from genetic drift during a range expansion, and therefore cannot be taken as evidence of the action of natural selection.  相似文献

We have used small subunit rRNA gene sequences to determine the phylogenetic relationships of species in three genera of endosymbiotic ciliates. We have confirmed that the astome Anoplophrya marylandensis is related to ciliates in the Class Oligohymenophorea, supporting the view that astomes are derived from hymenostome-like ancestors. We confirmed that Plaglotoma lumbrici, formerly considered to be a heterotrich, is a stichotrich spirotrich ciliate most closely related to Paraurostyla weissei in this analysis. Thus, the somatic polykinetids of Plagiotoma can be concluded to be cirri. We report the details of our isolation of Nyctotheroides deslierresae and Nyctotheroides parvus and confirm previous reports that these clevelandellids are related to the metopid and caenomorphid ciliates, now placed in the Class Armophorea.  相似文献
