Summary St. John's wort (Hypericum perforatum cv. Anthos) is a medicinal plant with historical and anecdotal evidence of efficacy as an anti-depressant. Recent research has demonstrated an active biosynthetic pathway leading to the production of the mammalian neurohormone melatonin in St. John's wort plantlets. The objective of the current study was to assess the physiological role of melatonin and related indoleamines in plant morphogenesis. In the initial experiments, two of the indoleamines; serotonin and melatonin, were supplemented to the culture medium. In subsequent research, six inhibitors of auxin and indoleamine action or transport, 2,3,5-triiodobenzoic acid, p-chlorophenoxyisobutyric acid, p-chlorophenyl-alanine, d-amphetamine, fluoxetine (ProzacTM), and methylphenidate (RitalinTM), were included in a culture medium in the presence or absence of the auxin, indoleacetic acid (IAA). The rate of de novo root and shoot organogenesis and the endogenous concentrations of auxin and indoleamines were determined in cultured explants. Significant reductions in de novo root regeneration were found to correspond with decreases in the pool of both IAA and melatonin. An increase in the endogenous concentration of melatonin was correlated with an increase in de novo root formation, and increased serotonin levels corresponded to increased shoot formation on the explants. Our findings provide the first evidence that a balance of the endogenous concentration of serotonin and melatonin may modulate plant morphogenesis in vitro.  相似文献

Exploratory surveys were conducted in Europe to locate potential biological control agents of Vincetoxicum nigrum and Vincetoxicum rossicum and to collect known specialist herbivores of Vincetoxicum hirundinaria (Abrostola asclepiadis, Chrysolina a. asclepiadis, Eumolpus asclepiadeus and Euphranta connexa). Development of the most abundant herbivores found attacking leaves, roots, and developing seeds was evaluated in the laboratory on three Vincetoxicum spp. Field surveys revealed that the highest diversity of herbivores was associated with the abundant and widespread V. hirundinaria. No new herbivores were reported from V. hirundinaria or V. nigrum, but the leaf‐feeding noctuid Hypena opulenta was recorded for the first time attacking V. rossicum and V. scandens in Ukraine. Based upon larval survival and development and adult fecundity, the leaf feeders A. asclepiadis, C. a. asclepiadis, and H. opulenta, perform better on V. nigrum and V. rossicum than on V. hirundinaria. Larval performance of the root feeder E. asclepiadeus followed a similar pattern, but adult fecundity of this insect did not vary among host plant species. Immature development time of the pre‐dispersal seed feeder E. connexa is similar among hosts, but larvae grow larger on V. nigrum and V. hirundinaria than on V. rossicum. All herbivores are promising biological control agents of Vincetoxicum in North America and we have prioritized host range testing with H. opulenta and E. asclepiadeus.  相似文献

Aim: Technological properties of Lactobacillus fermentum isolates involved in spontaneous fermentation of dolo and pito wort were examined to select starter cultures. Methods and Results: 264 isolates were screened for antimicrobial activity, acidifying activity, exopolysaccharides (EPSs) and amylase production. An antimicrobial activity was detected for 33·3%, 31·8%, 22·7% and 15·9% of the isolates towards Staphylococcus aureus enterotoxin A producer, Staphylococcus aureus enterotoxin A and B producer, Escherichia coli and Listeria innocua, respectively. A similarity was found between the isolates which were clustered in four groups according to their rates of acidification of sorghum malt broth. The faster acidifying group of isolates (43·48%) had a rate of acidification evaluated as ΔpH of 1·14 ± 0·15 pH unit after 6 h of fermentation, followed by a second group of isolates (38·08%) with a similar rate of acidification after 9 h of fermentation. From the isolates endowed with an antimicrobial activity, 5·76% belonged to the faster acidifying group and 40·38% belonged to the second group. 88·7% of the isolates had the ability for producing EPSs but not amylase. Conclusion: Lactobacillus fermentum ferments dolo and pito wort by lowering the pH and providing organic acids, EPSs and antimicrobial compounds. Significance and Impact of the Study: Strains with a rapid rate of acidification, an antimicrobial activity and producing EPSs are suggested to have potential for starter cultures.  相似文献

Human-mediated species introductions offer opportunities to investigate when and how non-native species to adapt to novel environments, and whether evolution has the potential to contribute to colonization success. Many long-established introductions harbour high genetic diversity, raising the possibility that multiple introductions of genetic material catalyze adaptation and/or the evolution of invasiveness. Studies of nascent invasions are rare but crucial for understanding whether genetic diversity facilitates population expansion. We explore variation and evolution in founder populations of the invasive shrub Hypericum canariense . We find that these introductions have experienced large reductions in genetic diversity, but that increased growth and a latitudinal cline in flowering phenology have nevertheless evolved. These life history changes are consistent with predictions for invasive plants. Our results highlight the potential for even genetically depauperate founding populations to adapt and evolve invasive patters of spread.  相似文献

Two previously uncharacterized cDNAs encoding for polyketide synthases (PKSs), designated as HpPKS1 and HpPKS2, were isolated from Hypericum perforatum. The full-length HpPKS1 was 1573bp containing an open reading frame (ORF) of 1161bp encoding for a 386 amino acid protein. The full-length cDNA of HpPKS2 was 1559bp with an ORF of 1182bp encoding for a 393 amino acid protein. The highly conserved catalytic amino acid residues common to plant-specific PKSs were preserved in both genes. HpPKS1 and HpPKS2 exhibited distinct tissue-specific expression patterns in H. perforatum. The HpPKS1 expression was highest in flower buds and lowest in root tissues. The expression of HpPKS2 was found to be high in flower buds and leaf margins and low in leaf interior parts, stems and roots. The expression of the HpPKS1 was found to correlate with the concentrations of hyperforin and adhyperforin while the expression of HpPKS2 showed correlation with the concentrations of hypericins and pseudohypericins in H. perforatum tissues.  相似文献

Hypericum perforatum L. (St. John’s wort, SJW) is a traditional medicinal plant with a long history of pharmaceutical application. Today, besides formulations based on hydro-alcoholic extracts, lipophilic SJW preparations are widely used in phytomedicine e.g. for the symptomatic treatment of minor inflammations of the skin and subcutaneous tissue disorders. In the present study, SJW extracts were obtained with twelve different fatty oils according to a standardized protocol. In order to evaluate and compare the resulting macerates, a set of analytical parameters was chosen: the phytochemical profile of SJW oil extracts comprising acylphloroglucinol derivatives, flavonols, biflavones and naphthodianthrones was assessed using UHPLC-DAD and UV/VIS-spectroscopic methods Furthermore, overall appearance was monitored applying the CIE L*a*b* color system. Depending on the type of oil applied for extraction, a*-values, which are a measure of red color hues, ranged from 50.3 ± 0.2 (almond oil) to −3.8 ± 0.3 (macadamia nut oil). Considering total hypericin contents, extraction with almond oil also brought about highest amounts (5.5 ± 0.21 mg/100 g). In contrast, preparations with macadamia nut oil resulted in highest contents of hyperforin and adhyperforin, whereas a semi-synthetic extractant composed of medium-chain triglycerides (MCT) revealed highest yields of flavonoids, i.e. quercetin, kaempferol and I3, II8-biapigenin. Since the phytochemical composition and the corresponding specific characteristics of SJW oil preparations markedly differed, tailor-made SJW extracts may be prepared by systematic adaption of the manufacturing conditions, including the choice of a specific fatty oil. Additionally, SJW oil extracts were stored under different temperature (5 °C, 20 °C, 50 °C) and light (artificial light, darkness) regimes over a period of six months to evaluate their respective storage stability. For maximizing the retention of the investigated plant secondary metabolites in the corresponding oil extracts, cold storage (5 °C) under the exclusion of light is recommended.  相似文献

St. John's wort (Hypericum perforatum L.) is a medicinal plant traditionally used, both externally and internally, in all Europe for many pathologies. Paracelsus named it “arnica of the nerves” because of its empirical use in nervous diseases. In the last two decades many studies have proved the efficacy of some St. John's wort extracts in mild to moderate depression and it has been successful as an antidepressant both in Europe and the US. Its high efficacy and tolerability is unquestionable and from the clinical studies the activity is comparable to other antidepressants while lacking major side effects, making it a safe antidepressant.However, recently its potential to induce the metabolism of co-administered medications has been reported because it may potentate certain enzymes of the cytochrome P450 enzyme system. This resulted in a lowering of serum concentration of a number of concomitant drugs, including warfarin, digoxin, theophylline, cyclosporin, and indinavir. Many drugs and also several common foods and drinks can influence this enzyme system. So, even if its safety has been well established, physicians should be aware that St. John's wort administration might significantly affect other prescribed medicines.  相似文献

Summary When glucose and fructose are fermented separately, the uptake profiles indicate that both sugars are utilized at similar rates. However, when fermentations are conducted in media containing an equal concentration of glucose and fructose, glucose is utilized at approximately twice the rate of fructose. The preferential uptake of glucose also occurred when sucrose, which was first rapidly hydrolyzed into glucose and fructose by the action of the enzyme invertase, was employed as a substrate. Similar results were observed in the fermentation of brewer's wort and wort containing 30% sucrose and 30% glucose as adjuncts. In addition, the high levels of glucose in the wort exerted severe catabolite repression on maltose utilization in theSaccharmyces uvarum (carlsbergensis) brewing strain. Kinetic analysis of glucose and fructose uptake inSaccharomyces cerevisiae revealed aK m of 1.6 mM for glucose and 20 mM for fructose. Thus, the yeast strain has a higher affinity for glucose than fructose. Growth on glucose or fructose had no repressible effect on the uptake of either sugar. In addition, glucose inhibited fructose uptake by 60% and likewise fructose inhibited, glucose uptake by 40%. These results indicate that glucose and fructose share the same membrane transport components.  相似文献

Abstract. Hypericin, a photodynamic pigment, occurring in members of the Hypericaceae, can induce photosensitivity in grazing animals. The pigment has been isolated from the glandular trichomes located on the calyx of Hypericum hirsulum. Hypericin is shown to be capable of sensitizing the photo-oxidation of methyl linolenate. This activity is reduced in the presence of crocin, a carotenoid. Evidence for the generation of singlet molecular oxygen by hypericin is provided by the monitoring of oxygen consumption during the photosensitized oxidation of imidazole. Rates of oxygen consumption were modified by deuterium oxide and sodium azide. The photodynamic action of hypericin on pea leaf discs results in the promotion of photo-oxidative damage, measured by pigment loss and ethane production. These results are discussed in relation to the possible function of hypericin within the plant and the role of photo-dynamic reactions in nature.  相似文献
