Summary The apical cell coat of the olfactory epithelium proper and the vomeronasal neuroepithelium of the rat was investigated electronmicroscopically by means of the Ruthenium-red reaction. In the olfactory epithelium proper, the cilia of receptor cells and microvilli of supporting cells possess a cell coat measuring approximately 10 nm in thickness. In the vomeronasal neuroepithelium, the apical cell coat is thicker than in the olfactory epithelium proper. On microvilli of vomeronasal receptor cells the cell coat varies in thickness from 15 to 20 nm, and on microvilli of supporting cells it measures approximately 75 nm. Red clover (Trifolium pratense L.) seedlings were screened for the ability to regenerate plantlets from hypocotyl-derived callus tissue. Media sequences described by Beach and Smith (1979) and Collins and Phillips (1982) and a variation using media from both sequences were tested. Plantlets were regenerated from three out of 642 genotypes. In all three cases, callus was initiated on B5C medium and regeneration was accomplished on SPL medium. Attempts to regenerate plants from petiole-derived callus tissue have so far been successful only with regenerants of clone F49. Petiole callus from epicotyl-derived F49 plants proved to be non-regenerative. Pollen viability varied significantly among individuals regenerated from callus cultures of clone F49. Root tip squashes from F49 regenerants revealed the normal diploid chromosome number (2n=14). Summary Hydroxylamine, a potent oxidizing agent used to reverse carbethoxylation of histidine by diethylpyrocarbonate, activated Cl-dependent K flux (KCl cotransport) of low K sheep red blood cells almost sixfold. When KCl cotransport was already stimulated by N-ethylmaleimide, hydroxylamine caused an additional twofold activation suggesting modification of sites different from those thiol alkylated. This conclusion was supported by the finding that hydroxylamine additively augmented also the diamide-induced KCl flux (Lauf, P.K. 1988.J. Membrane Biol. 101:179–188) with dithiothreitol fully reversing the diamide but not the hydroxylamine effect. Stimulation of KCl cotransport by hydroxylamine was completely inhibited by treatment with diethylpyrocarbonate also known to prevent KCl cotransport stimulation by N-ethylmaleimide, both effects being independent of the order of addition. Hence, although the effect of carbethoxy modification on KCl flux cannot be reversed by hydroxylamine and thus excludes histidine as the target for diethylpyrocarbonate, our finding reveals an important chemical determinant of KCl cotransport stimulation by both hydroxylamine oxidation and thiol group alkylation.  相似文献

The interactions of Heterodera glycines at four egg inoculum levels (0, 100, 1,000, and 10,000 per pot) and three cyst levels (0, 100, and 200 per pot) and Calonectria crotalariae at 500, 5,000, and 50,000 microsclerotia per pot were evaluated on soybean. At the two lowest nematode egg levels, the presence of C. crotalariae did not affect nematode reproduction. At 10,000 eggs per pot, however, nematode reproduction was increased significantly at each microsclerotial level. The increase in nematode reproduction was stepwise at 500 and 5,000 microsclerotia per pot but declined at 50,000 microsclerotia per pot. Similar results were obtained when cysts rather than eggs were used as nematode inoculum. The nematode x fungus interaction significantly affected 60-day plant growth parameters of both Lee 74 and Centennial soybean. The nematode x fungus interaction was antagonistic to plant roots and significantly influenced root injury ratings. The presence of C. crotalariae in tissues of stock plants or plants used as race differentials did not alter the analysis of this population as race 3.  相似文献

The palm weevil, Rhynchophorus palmarum (L.), was collected in cocoons from red ring-diseased coconut palms (Cocos nucifera L.) in Trinidad and Tobago. Juveniles of five species of nematodes were extracted from the genitalia and macerated bodies of newly emerged adults of the palm weevil: Rhadinaphelenchus cocophilus (Cobb) Goodey (the red ring nematode), Teratorhabditis sp., Diplogasteritus sp., Mononchoides sp., and Bursaphelenchus sp. Over 90% of newly emerged weevil females and males were infested internally with red ring nematode juveniles, and over 47% of the weevils contained more than 1,000 red ring nematodes each. There was no significant correlation between weevil body length and the number of red ring nematodes carried internally by each weevil. Teratorhabditis sp. and Diplogasteritus sp. were extracted from over 50% of the palm weevils, and Monochoides sp. and Bursaphelenchus sp. were found in a small proportion of the weevils. Field-collected adult weevils were also internally and externally infested with a Rhabditis sp., which was not observed in or on weevils allowed to emerge from field-collected cocoons.  相似文献

At the developmental stage at which the apical hook passed the 3rd and 4th nodes, dark-grown seedlings of pea ( Pisum sativum L. cv. Progress No.9) opened the hook upright and then formed a new hook above the node nearly in the opposite direction to the previous one. In cv. Alaska, in contrast, many (about 84%) seedlings closed the hook in the original direction after they partially (up to about 110°) opened it at the 3rd node, thus doing a wagging movement, while a small percentage (about 16%) of the seedlings reversed the hook direction. Exposure to red light of cv. Alaska seedlings for 10 min increased the percentage of the hook reversion up to 71% or more. The hook reversion was never observed except when the hook part passed the nodes, suggesting the involvement of the nodes in the phenomenon.  相似文献

A comparative analysis of the gastrointestinal tracts of wild-caught marmosets, Callithrix emiliae, and tamarins, Saguinus fuscicollis weddelli, was undertaken in order to evaluate the degree of specialisation for digestion of plant exudates. Compared to S.f. weddelli, C. emiliae exhibits a reduced small intestine and a relatively large, compartmentalised caecum in which gum is probably fermented. The apparent specialisation of the digestive tract in C. emiliae correlates with that of its dentition, which is adapted for gouging the bark of gum-producing plants. A similar degree of specialisation of the caecum is predicted for other marmosets (Callithrix spp. and Cebuella pygmaea).  相似文献

We studied the specificities of human red cell membrane bindings of three long chain fatty acids, palmitic- arachidonic- and oleic acid, using resealed membranes, ghosts. Previously estimated binding capacities, affinities and inside/outside distributions [6, 10, 11, 12], suggest separated binding sites. This possibility is explored by estimating the binding properties of one fatty acid in the presence of one or two of the others. Binding capacities, nmol g−1 ghosts, of palmitic and arachidonic acid estimated simultaneously vs. separately are 27.4 ± 2.7 vs. 29.0 ± 2.1 (P < 0.6) and 6.5 ± 0.6 vs. 5.5 ± 0.5 (P < 0.2) respectively. The corresponding estimates for oleic- and palmitic acid are 36.5 ± 2.0 vs. 34.0 ± 2.2 (P < 0.4) and 28.4 ± 1.8 versus 29.1 ± 2.1 (P < 0.8). The binding sites are therefore independent. For each of the three fatty acids in the absence or in the presence of one or two of the others, the inside/outside distributions of the binding sites and the membrane transfer rate constants are elucidated by exchange efflux kinetics at 0°C from ghosts with and without enclosed albumin. Packed ghosts loaded with radioactive acids are injected rapidly into a large volume of vigorously stirred buffer with albumin. With a resolution time of about 1-sec serial filtered ghost-free aliquots are collected and counted. The analyses show that palmitic- and oleic acid sites of transport are entirely independent but do not exclude that palmitic- and/or oleic acid binding may diminish the arachidonic acid affinity a little. The diversity combined with specificity suggests that the transport sites for long chain fatty acids are protein-determined microdomains of phospholipids. In a previous study large-subunit ribosomal RNA gene (LSU rDNA) sequences from the marine dinoflagellates Alexandrium tamarense (Lebour) Balech, A. catenella (Whedon et Kofoid) Balech, A. fundyense Balech, A. affine (Fukuyo et Inoue) Balech, A. minutum Halim, A. lusitanicum Balech, and A. andersoni Balech were compared to assess inter- and intraspecific relationships. Many cultures compared in that study contained more than one class of LSU rDNA. Sequencing pooled clones of rDNA from single cultures revealed length heterogeneities and sequence ambiguities. This complicated sequence comparisons because multiple rDNA clones from a single culture had to be sequenced individually to document the different classes of molecules present in that culture. A further complication remained as to whether or not the observed intraculture sequence variations were reliable genetic markers or were instead artifacts of the polymerase chain reaction (PCR) amplification, cloning, and/or sequencing methods employed. The goals of the present study were to test the accuracy of Alexandrium LSU rDNA sequences using restriction fragment-length polymorphism (RFLP) analysis and to devise RFLP-based assays for discriminating among representatives of that group. Computer-assisted examination of the sequences allowed us to identify a set of restriction enzymes that were predicted to reveal species, strain, and intraculture LSU rDNA heterogeneities. All groups identified by sequencing were revealed independently and repeatedly by RFLP analysis of PCR-amplified material. Five ambiguities and one length heterogeneity, each of which ascribes a unique group of Alexandrium species or strains, were confirmed by restriction digests. Observed intraculture LSU rDNA heterogeneities were not artifacts of cloning and sequencing but were instead a good representation of the spectrum of molecules amplified during PCR reactions. Intraculture LSU rDNA heterogeneities thus serve as unique genetic markers for particular strains of Alexandrium, particularly those of A. tamarense, A. catenella, and A. fundyense. However, some of these “signature heterogeneities” represented a smaller portion of PCR product than was expected given acquired sequences. Other deviations from predicted RFLP patterns included incomplete digestions and appearance of spurious products. These observations indicate that the diversity of sequences in PCR product pools were greater than that observed by cloning and sequencing. The RFLP tests described here are useful tools for characterizing Alexandrium LSU rDNA to define the evolutionary lineage of cultures and are applicable at a fraction of the time, cost, and labor required for sequencing.  相似文献
