Infections by Sarcocystis in cattle are ubiquitous worldwide. There is considerable debate concerning the identity of Sarcocystis spp. in cattle. Proper diagnosis of Sarcocystis spp. is important to assess their economic and public health importance. Currently there are seven named species: Sarcocystis hirsuta, Sarcocystis cruzi, Sarcocystis hominis, Sarcocystis bovifelis, arcocystis heydorni, Sarcocystis bovini and Sarcocystis rommeli. Additionally, there are unnamed Sarcocystis spp. Two species, S. hominis and S. heydorni, are zoonotic. One out of seven species (S. hirsuta, contracted from cats) forms macroscopic cysts which can be visible during carcass inspection. Current molecular characterization is based on DNA extracted from sarcocysts from naturally infected cattle because DNA was not characterized from tissues of experimentally infected cattle or feces of experimentally infected definitive hosts. Sarcocystis cruzi (transmitted via canids) is recognized as the most pathogenic species and it causes abortion, low milk yield, poor body growth, and outbreaks of clinical sarcocystosis and death. Additionally, Sarcocystis infections have been linked to an inflammatory condition of striated muscles termed bovine eosinophilic myositis (BEM). Cattle affected by BEM appear clinically normal. Diagnosis of BEM at slaughter occurs when inspecting the carcass surface, or once the carcass has been divided into prime cuts or quarters. Sex and breed have no apparent influence on prevalence of BEM. The condition evidently occurs with equal frequency in steers, cows, and heifers. Virtually all striated muscles can be affected including skeletal muscles, the muscles of the eye, larynx, and the heart. In the USA, regulations require condemnation of BEM-affected parts, or (in severe cases) the entire carcass. These aesthetic considerations result in economic losses. Cattle experimentally infected with Sarcocystis did not have BEM at slaughter. Here, we review the status of Sarcocystis spp. and BEM in cattle including prevalence, lesions, epidemiology, and association of BEM with different species of Sarcocystis. 相似文献

Regulated exocytosis involves calcium-dependent fusion of secretory vesicles with the plasma membrane with three SNARE proteins playing a central role: the vesicular synaptobrevin and the plasma membrane syntaxin1 and SNAP-25. Cultured bovine chromaffin cells possess defined plasma membrane microdomains that are specifically enriched in both syntaxin1 and SNAP-25. We now show that in both isolated cells and adrenal medulla slices these target SNARE (t-SNARE) patches quantitatively coincide with single vesicle secretory spots as detected by exposure of the intravesicular dopamine beta-hydroxylase onto the plasmalemma. During exocytosis, neither area nor density of the syntaxin1/SNAP-25 microdomains changes on the plasma membrane of both preparations confirming that preexisting clusters act as the sites for vesicle fusion. Our analysis reveals a high level of colocalization of L, N and P/Q type calcium channel clusters with SNAREs in adrenal slices; this close association is altered in individual cultured cells. Therefore, microdomains carrying syntaxin1/SNAP-25 and different types of calcium channels act as the sites for physiological granule fusion in "in situ" chromaffin cells. Assessing the agronomic and environmental effects of the application of cattle manure compost on soil by multivariate methods 总被引:1,自引:0,他引:1
Multivariate analysis was used for interpreting data from a pot experiment using samples of three Spanish soils. Samples of soil fertilized with compost were compared with untreated control samples. We also compared the effect of adding the compost to soil with a controlled moisture content of 50% of its water holding capacity (WHC), and to a near-saturated soil (95% WHC). Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were used; they perfectly differentiated sample groups both as a function of the treatment applied and by sampling date. The compost samples were characterized by higher pH, electrical conductivity (EC), organic matter (OM) content and cation exchange capacity (CEC), together with nutrient concentrations than the control pots. The pots with a soil–compost mixture at 95% WHC presented lower values of EC, CEC, inorganic N, K, Na and B than the mixtures at 50% WHC. Multivariate methods may therefore be useful for the analysis and interpretation of a large number of data in soil research. 相似文献

目的探索建立实验性兔主动脉粥样硬化易损斑块模型的新方法。方法24只雄性日本大耳白家兔随机分为对照组8只,实验组16只,对照组给予普通饲料;实验组给予高胆固醇饲料、注射牛血清白蛋白及进行腹主动脉球囊拉伤术,分别于0周、3周、6周、10周检测血脂、ox-LDL、TNF-α、IL-1、IL-10,实验结束时取腹主动脉进行病理形态学分析及NF-κBp65亚基免疫组化染色分析。结果实验组斑块内膜面积比为53.6%,脂核面积与斑块面积比为54.9%,斑块纤维帽厚度与内膜中膜厚度(IMT)比约为8.5%;除甘油三酯变化不大外,实验组TC、LDL-C、HDL-C、LDL-C/HDL-C、TC/HDL-C、ox-LDL、TNF-α、IL-1、IL-10均有明显升高趋势,在3周、6周、10周时与对照组比较差异有统计学意义;且NF-κBp65亚基阳性染色面积较对照组高13.5倍。结论高脂喂养、免疫损伤加球囊拉伤可以成功建立家兔主动脉粥样硬化易损斑块模型。 相似文献

Abstract: Basic knowledge of white-tailed deer (Odocoileus virginianus) hearing can improve understanding of deer behavior and may assist in the development of effective deterrent strategies. Using auditory brainstem response testing, we determined that white-tailed deer hear within the range of frequencies we tested, between 0.25–30 kilohertz (kHz), with best sensitivity between 4–8 kHz. The upper limit of human hearing lies at about 20 kHz, whereas we demonstrated that white-tailed deer detected frequencies to at least 30 kHz. This difference suggests that research on the use of ultrasonic (frequencies >20 kHz) auditory deterrents is justified as a possible means of reducing deer—human conflicts. 相似文献

Summary The subcellular distribution of noradrenaline (NA), neuropeptide Y (NPY), Met and Leu-enkephalin (ENK), substance P (SP), somatostatin (SOM), and vasoactive intestinal polypeptide (VIP) was investigated in homogenates of bovine splenic nerve. The distribution of noradrenergic peptide-containing nerves in the bovine celiac ganglion, splenic nerve and terminal areas in spleen was studied by indirect immunofluorescence histochemistry using antisera to tyrosine hydroxylase (TH), dopamine--hydroxylase (DBH), NPY, enkephalin peptides, SP, SOM, VIP and peptide HI (PHI).After density gradient centrifugation, high levels of NPY and ENK-like immunoreactivity (LI) were found in high-density gradient fractions, coinciding with the main NA peak. SP, SOM and VIP were found in fractions with a lower density, VIP being also enriched in a heavy fraction; the latter three peptides were present in low concentrations.Immunohistochemistry revealed that staining for NPYLI and ENK-LI partly overlapped that for TH and DBH in celiac ganglia, splenic nerve axons and terminal areas of spleen. Almost all principal ganglion cells were TH- and DBH-immunoreactive. Many were also NPY-immunoreactive, whereas a smaller number were ENK-positive. In the celiac ganglion patches of dense SP-positive networks and some VIP/PHI- and ENK-immunoreactive fibers were seen around cell bodies.The results indicate that NPY and ENK are stored with NA in large dense-cored vesicles in unmyelinated axons of bovine splenic nerve. Bovine seminal ribonuclease (BS-RNase), a dimeric homologue of RNase A, cleaves both single- and double-stranded RNA and inhibits the growth of tumor cells. Its catalytic activity against double-stranded RNA, either homopolymeric ([3H]polyA/polyU) or mixed sequence, is enhanced by bovine or human recombinant interferon-γ (IFN-γ). Activation is seen with as little as 4–10 interferon units per assay. The oligo-O-acetylation of sialic acids found in normal colonic mucins is greatly reduced in colorectal cancer. Mucins prepared from cancer tissue in adenocarcinoma showed this reduction, while normal O-acetylation was detected in resection margin and control cases and total mucin sialic acid content was significantly decreased in cancer vs control samples. A reduction of the O-acetyl transferase activity catalysing the O-acetylation reaction was also found. A series of cultured human colorectal cell lines derived from the same premalignant adenomatous line, and representative of the adenoma-carcinoma sequence were examined and revealed a depletion of oligo-O-acetylation in the original diploid premalignant line, re-expression in a further premalignant line and reduction in malignant mucinous and adenocarcinoma cell lines. Reduction of sialic acid O-acetylation appears as an early event in the process of malignant transformation in human colorectal cancer. 相似文献
