过敏性哮喘的发病率呈上升趋势。使用了几十年的主要治疗药物肾上腺糖皮质激素副作用较大,因此发现好的预防和治疗方法成为迫切要解决的问题。动物模型是研究人类疾病的重要手段,但不少疑难病的发病机理不明确,因而制备的动物模型和人类疾病的相似度有差异。但I型变态反应作为过敏性哮喘的发病机理是比较明确的,据此制备的动物模型和人类的哮喘就有很高的相近度,结果的可信度就较高。本文回顾了哮喘动物模型制备的基本方法和某些重要的细节。着重讨论了当今最常用的气道高反应性模型的优劣。如果综合运用不同特点的模型尤其是能观察记录哮喘发作全过程包括速发和迟发反应的模型,将可以更直接地探索哮喘发病过程和治疗药物。对气道重塑及基因敲除和转基因技术在动物模型中的研究和使用也做了一般性论述。动物模型将是一个有力的工具为最后有效地预防和治疗过敏性哮喘找到突破口。  相似文献

AIMS: To evaluate the growth of Listeria monocytogenes and shelf life of cooked and peeled shrimps in modified atmosphere packaging (MAP). METHODS AND RESULTS: Storage trials with naturally contaminated cooked and peeled MAP shrimps (Pandalus borealis) were carried out at 2, 5 and 8 degrees C. Challenge tests at the same conditions were performed after inoculation with Listeria monocytogenes. Both storage trials and challenge tests were repeated after 4 months of frozen storage (-22 degrees C). Brochothrix thermosphacta and Carnobacterium maltaromaticum were responsible for sensory spoilage of cooked and peeled MAP shrimps. In challenge tests, growth of L. monocytogenes was observed at all of the storage temperatures studied. At 5 and 8 degrees C the concentration of L. monocytogenes increased more than a 1000-fold before the product became sensory spoiled whereas this was not observed at 2 degrees C. Frozen storage had only a minor inhibiting effect on growth of L. monocytogenes in the thawed product. CONCLUSIONS: To prevent L. monocytogenes becoming a safety problem, cooked and peeled MAP shrimps should be distributed at 2 degrees C and with a maximum shelf life of 20-21 d. At higher temperatures shelf life is significantly reduced. SIGNIFICANCE AND IMPACT OF THE STUDY: Information is provided to establish shelf life of cooked and peeled MAP shrimps.  相似文献

Growth of a near‐isogenic line (NIL) for the purple leaf gene Pl of rice with a genetic background of Taichung 65 (T‐65) rice was significantly retarded by supplementary ultraviolet‐B radiation (UV‐B), despite the fact that the amounts of UV‐absorbing compounds and anthocyanins in NIL were significantly higher than those in T‐65. In order to understand the role of flavonoids in UV‐B induced damage protection in T‐65 and the NIL, both the (1) relationships between changes in the steady state of cyclobutane pyrimidine dimer (CPD) levels and changes in accumulation of anthocyanins and UV‐absorbing compounds in leaves with leaf age, and (2) the susceptibility to CPD induction by UV‐B radiation and the ability to photorepair CPD were examined. Although supplementary UV‐B elevated the steady state of CPD levels in leaves in both strains, the level in the leaf of the NIL was higher than that in T‐65 at any time. The susceptibility to CPD induction by short‐term (challenge) UV‐B exposure was lower in the NIL than in T‐65. On the other hand, the CPD photorepair was also lower in the leaves of the NIL than in those of T‐65. The decrease in CPD‐photorepair in the NIL was due to a lowering of the leaf‐penetrating blue/UV‐A radiation, which is effective for photoreactivation by photolyase, by anthocyanins. Thus, accumulation of anthocyanins and UV‐absorbing compounds did not effectively function as screening against damage caused by elevated UV‐B radiation in the NIL, and the retardation of growth in the NIL resulted from its lower ability to photorepair CPD by higher amounts of anthocyanins.  相似文献

Blood respiratory, acid-base, and ionic changes in response to hyperosmotic shock were studied in vivo and in vitro in the European flounder. One primary aim was to evaluate regulatory changes in red blood cell (RBC) volume and its interrelationship with blood O2 transporting properties. An acute increase in the ambient salinity from 10 to 30 ppt caused small but significant increases in extracellular osmolality (<20 mosM kg−1), [Na+], and [Cl], which were corrected within 48 h. RBC volume was not significantly changed 3 h after the in vivo exposure to elevated salinity. A small metabolic acidosis was fully developed within 3 h, and this acidosis seemed responsible for a modest decrease in blood O2 affinity (i.e., increased P50-O2 tension at 50% O2 saturation). RBC organic phosphates were unchanged. In vitro elevation of whole blood extracellular osmolality by 60 mosM kg−1 caused immediate RBC shrinkage. The subsequent regulatory volume increase (RVI) showed a graded dependency on blood O2 saturation (SO2). At SO2 values of 0% and 20%, there were full RBC volume recoveries within 120 min, RVI was partial at SO2 values of 45% and 55%, and RVI was absent at a SO2 of 100%. SO2 and P50 did not change significantly during RBC shrinkage and RVI. Thus, the up-concentration of cellular haemoglobin and organic phosphates in hyperosmotically shrunken RBCs had minimal influence on blood O2 transporting properties. The degree of cell shrinkage and time needed for RVI were positively correlated with the magnitude of the rise in extracellular osmolality. The RVI proceeded via elevation of cellular [Na+], [Cl], and to some extent also [K+]. Cell volume regulatory mechanisms are only needed to correct minor volume disturbances in vivo, because changes in extracellular osmolality were limited by an efficient osmotic regulation at the epithelial interface between extracellular compartment and environment.  相似文献

Nest-holder male Salaria pavo have lower circulating concentrations of 11-ketotestosterone (KT) at the beginning of the breeding season than at its peak. At that peak density of nesting males was higher as were the number of visits of sneaker males to nests and of agonistic interactions between nest-holders and sneaker males. There was no difference between the two dates either in the frequency of male-male interactions or in the frequency of courtship episodes. Thus, higher plasma levels in nest-holders might be explained by a more intense sneaking pressure at the peak of the breeding season. At that peak, nest-holders had higher plasma levels of KT and a higher testosterone (T) to KT metabolization index in the gonads than did floater males. Both nest-holders and floaters had higher levels of KT and T in the testicular gland than in the testis. The levels of both androgens in the testicular gland, but not in the testis, were correlated with circulating concentrations of KT. These results suggest that the testicular gland is the major source of circulating KT in blenniids. Nest-holders had higher metabolization indexes than floaters both in the testis and in the testicular gland, which suggests that nest holding status promotes the conversion of T into KT.  相似文献

To investigate the role played by urease during the Bordetella bronchiseptica infection process, the ability to colonise and persist in the mouse respiratory tract of a urease-negative B. bronchiseptica BB7865 and a BB7865 derivative constitutively expressing urease was compared with that of the wild-type strain. The results obtained showed that neither constitutive expression nor abolishment of urease activity had any significant effect on the course of B. bronchiseptica infection. Therefore, under our experimental conditions, urease is not essential for B. bronchiseptica to colonise and persist within the murine host.  相似文献

Recent reports indicate that exposure to some stressors, such as shipping or immune challenge with the bacterial endotoxin, lipopolysaccharide (LPS), during the peripubertal period reduces sexual receptivity in response to ovarian hormones in adulthood. We hypothesized that a peripubertal immune challenge would also disrupt the response of a non-reproductive behavior, anxiety-like behavior, to ovarian hormones in adulthood. Female C57Bl/6 mice were injected with LPS during the peripubertal period and tested for anxiety-like behavior in adulthood, following ovariectomy and ovarian hormone treatment. Treatment with estradiol followed by progesterone reduced anxiety-like behavior in control, but not LPS-treated females. We next determined if the disruptive effect of LPS on adult behavior were limited to the peripubertal period by treating mice with LPS either during this period or in adulthood. LPS treatment during the peripubertal period disrupted the anxiolytic effect of ovarian hormones, whereas treatment in adulthood did not. We further tested if this model of peripubertal immune challenge was applicable to an outbred strain of mice (CD-1). Similar to C57Bl/6 mice, LPS treatment during the peripubertal period, but not later, disrupted the anxiolytic effect of estradiol and progesterone. These data suggest that a peripubertal immune challenge disrupts the regulation of anxiety-like behavior by ovarian hormones in a manner that persists at least for weeks after the termination of the immune challenge.  相似文献

An increasing number of international initiatives aim to reconcile development with conservation. Crucial to successful implementation of these initiatives is a comprehensive understanding of the current ecological condition of landscapes and their spatial distributions. Here, we provide a cumulative measure of human modification of terrestrial lands based on modeling the physical extents of 13 anthropogenic stressors and their estimated impacts using spatially explicit global datasets with a median year of 2016. We quantified the degree of land modification and the amount and spatial configuration of low modified lands (i.e., natural areas relatively free from human alteration) across all ecoregions and biomes. We identified that fewer unmodified lands remain than previously reported and that most of the world is in a state of intermediate modification, with 52% of ecoregions classified as moderately modified. Given that these moderately modified ecoregions fall within critical land use thresholds, we propose that they warrant elevated attention and require proactive spatial planning to maintain biodiversity and ecosystem function before important environmental values are lost.  相似文献
