一株高效广谱染料降解细菌的分离鉴定及其脱色特性初探

从土壤样品中分离到一株高效染料脱色菌株N-4,根据形态学特征及16S rDNA基因序列分析,该菌株初步鉴定为Leucobacter sp.。利用表面响应法(RSM)对菌株N-4脱色活性深蓝K-R的主要因素进行优化,实验结果表明,菌株N-4脱色K-R的最优条件为:湿菌量10 g/L,染料浓度222 mg/L,硫酸铵1.5 g/L,果糖3.5 g/L,最佳脱色率为100%。此外,实验证明其对多种染料均具有较高的脱色效率。同时,考察了金属离子对染料脱色效率的影响,其中K+、Ca2+、Mg2+、Ba2+、Mn2+等对脱色具有促进作用,而Ni2+、Cu2+、Hg2+对脱色具有明显的抑制作用。     

盐泽螺旋藻藻胆蛋白的分离和特性研究

盐泽螺旋藻(Spirulina subsalsa)的水溶性色素粗提物经过硫酸铵沉淀和羟基磷灰石(HA)柱层析后可以分出两种藻胆蛋白,即藻蓝蛋白(c-PC)和别藻蓝蛋白(APC)。它们的纯度(指其在可见光部分的最大吸收与280nm处吸收之比)可分别达到7．27(c-PC)和6．55(APC)。而一般认可的纯度标准,PC为5,APC为6。纯化后的c—PC和APC在聚丙烯酰胺凝胶电泳(PAGE)中仅见一条色带,其最大吸收峰分别在620nm和650nm,其室温荧光发射峰分别为642nm和657nm。     

Biochemical characterization of an ABC transporter LptBFGC complex required for the outer membrane sorting of lipopolysaccharides

Seven Lpt proteins (A through G) are thought to be involved in lipopolysaccharide transport from the inner to outer membrane of Escherichia coli. LptB belongs to the ATP-binding cassette transporter superfamily. Although the lptB gene lacks neighboring genes encoding membrane subunits, bioinformatic analyses recently indicated that two distantly located consecutive genes, lptF and lptG, could encode membrane subunits. To examine this possibility, LptB was expressed with LptF and LptG. We report here that both LptF and LptG formed a complex with LptB. Furthermore, an inner membrane protein, LptC, which had been implicated in lipopolysaccharide transport, was also included in this complex.

Gill microsomal (Na+,K+)-ATPase from the blue crab Callinectes danae: Interactions at cationic sites

Euryhaline crustaceans tolerate exposure to a wide range of dilute media, using compensatory, ion regulatory mechanisms. However, data on molecular interactions occurring at cationic sites on the crustacean gill (Na⁺,K⁺)-ATPase, a key enzyme in this hyperosmoregulatory process, are unavailable. We report that Na⁺ binding at the activating site leads to cooperative, heterotropic interactions that are insensitive to K⁺. The binding of K⁺ ions to their high affinity sites displaces Na⁺ ions from their sites. The increase in Na⁺ ion concentrations increases heterotropic interactions with the K⁺ ions, with no changes in K_0.5 for K⁺ ion activation at the extracellular sites. Differently from mammalian (Na⁺,K⁺)-ATPases, that from C. danae exhibits additional NH₄⁺ ion binding sites that synergistically activate the enzyme at saturating concentrations of Na⁺ and K⁺ ions. NH₄⁺ binding is cooperative, and heterotropic NH₄⁺ ion interactions are insensitive to Na⁺ ions, but Na⁺ ions displace NH₄⁺ ions from their sites. NH₄⁺ ions also displace Na⁺ ions from their sites. Mg²⁺ ions modulate enzyme stimulation by NH₄⁺ ions, displacing NH₄⁺ ion from its sites. These interactions may modulate NH₄⁺ ion excretion and Na⁺ ion uptake by the gill epithelium in euryhaline crustaceans that confront hyposmotic media.     

Synthesis and characterization of carboxymethyl-polyaminate chitosan and its adsorption behavior toward a reactive dye

Carboxymethyl-polyaminate chitosan (DETA-CMCHS), a novel kind of amphoteric chitosan derivative, was prepared and characterized by elemental analysis, and by IR and ¹H NMR spectroscopy. The adsorption behavior of Reactive Blue (RB2) on DETA-CMCHS was also studied. Results showed that the maximum value of adsorption capacity was 1185.71 mg/g at pH 3. The adsorption kinetic behavior was fitted with a second-order reaction rate equation. The adsorption was an exothermic, irreversible and entropy-reduced process according to thermodynamic parameters, such as standard Gibbs free-energy change, enthalpy change, and entropy change, all of which were calculated from adsorption equilibrium data.     

免疫毒素Luffin B-Ng76对人黑色素瘤细胞的体外抑制作用

用ＢｌｕｅＳｅｐｈａｒｏｓｅＣＬ－６Ｂ凝胶亲和层析法从丝瓜籽中分离纯化了单链致核糖体失活蛋白（ｒｉｂｏｓｏｍｅｉｎａｃ－ｔｉｖａｔｉｎｇｐｒｏｔｅｉｎ,ＲＩＰ）——ｌｕｆｉｎＢ。并将ｌｕｆｉｎＢ与抗人黑色素瘤细胞单抗Ｎｇ７６制成了免疫毒素,命名为ｌｕｆｉｎＢ－Ｎｇ７６,它对体外培养的黑色素瘤细胞Ｍ２１有很强的抑制作用,ＩＣ５０为２．５×１０－１１ｍｏｌ／Ｌ,毒性比游离的ｌｕｆｉｎＢ提高４０００倍,而它对非靶的ＨｅＬａ细胞的毒性较Ｍ２１细胞低１２００倍。结果提示ｌｕｆｉｎＢ用于制备免疫毒素具有良好的应用前景。     

西藏墨脱地区6种鸟类种和亚种的新分布记录

2018年10月至2021年10月,在西藏墨脱县开展鸟类多样性调查期间记录到6种鸟类新分布,分别是大蓝仙鹟 (Cyornis magnirostris)、黄嘴角鸮 (Otus spilocephalus)、金胸雀鹛指名亚种 (Lioparus chrysotis chrysotis)、细嘴钩嘴鹛Sintextus亚种 (Pomatorhinus superciliaris intextus)、白喉姬鹟指名亚种 (Anthipes monileger monileger)、棕胸雅鹛assamense亚种 (Trichastoma tickelli assamense)。     

A modified Coomassie Brilliant Blue staining method at nanogram sensitivity compatible with proteomic analysis

A more sensitive and convenient Coomassie Brilliant Blue (CBB) staining method for visualizing proteins was developed. Compared with the modifications include the supplement of 10% (v/v) methanol into the fixing solution, an increase of an additional sensitization step and CBB raised from 0.1 to 0.125%. The improved method can detect proteins at nanogram level. The improved method is more sensitive than Blue Silver and more convenient than the Silver protocol. Mass spectrometry results confirmed that it is suitable for subsequent proteomic research.     

贺兰山保护区冬季岩羊集群特征的初步分析

Group size and composition of blue sheep （Pseudois nayaur) were studied in the Helan Mountains, Ningxia Autonomous Region from November to December 2003. We scanned mountain slopes with binoculars and observed with 20 - 60 x spotting-scope. A total of 310 herds of blue sheep and 1 336 individuals were observed during the study period. Blue sheep were frequently seen in small herds of 2 to 8 individuals, which represent 94.8% of total herds observed. Herds consisting of 9 individuals or more represented 5.2%. The largest herd we observed numbered 51 individuals. Mean group size was 4.2 individuals. Blue sheep herds can be divided into three types: male herds （composed solely of males), female herds (consisting of females with or without juveniles of both sexes), and mixed herds （including adult males, females, and subadults). Among the 310 herds, female herds were counted 150 times (48.4%}, mixed herds 154 times (49.7%), and male berds 6 times (1.9%) . Of 1 336 blue sheep classified by sex and age, adults, subadults and juveniles composed 64.1%, 20.8%, and 15.1% respectively. The female: male ratio of adults was 1:0.73. The ratio of adult females to juveniles was 1:0.56, which is higher than the ratio recorded in spring （ 1:0.43) or summer （ 1:0.44). The results showed that the Helan Mountains State Nature Reserve has succeed in protecting blue sheep.     

Purification and specificity of RNase A3 from Vicia faba root cells

Vicia faba root ribonucleases are bound to Cibacron blue F3GA. A Blue dextran-Sepharose column was used to purify RNase A₃, the more abundant enzyme from V. faba root. Using dinucleoside monophosphate as substrates, it appears that this enzyme behaves as a cyclizing phosphotransferase. With high enzyme/substrate ratios on prolonged digestion a partial release of a nucleoside 3′ phosphate occurs. The specificity is relatively high since only the purine-purine phosphodiester linkages out of 16 types of possible links are easily cleaved. When a pyrimidine is involved in the phosphodiester bond, a much slower rate of attack (Py in 5′) or no attack (Py in 3′) was detected.
