公园绿地作为城市重要的自然景观元素,其可达性是体现公园绿地所提供的自然服务能否被市民便捷、公平享用的一个重要指标.对可达性理论进行深入研究,从而更加准确、真实地对公园绿地可达性进行评价是目前亟需解决的问题.本文基于QuickBird卫星影像和GIS平台,以沈阳市铁西区为例,选取步行、非机动车、机动车和公共交通4种方式来定量评价公园绿地的可达性,并对4种交通方式下公园绿地可达区的空间分布、可达面积及可达人口的数量差异进行对比分析.结果表明: 沈阳市铁西区公园绿地数量较少且分布不均衡,表现为西南部及中心区域公园绿地的可达性较好,边缘区域可达性较差,东部和北部边缘可达性最差;各交通方式下公园绿地的可达性存在较大差异,机动车可达性最好,其次为非机动车和公共交通,步行可达性最差:近九成的居民可以采用步行30 min、非机动车和公共交通15 min、机动车10 min的方式到达最近的公园绿地.该研究对城市公园绿地可达性理论的深入、系统研究提供了一种新思路,对基于可达性的绿地空间布局优化具有重要的现实意义.  相似文献

冷蒿(Artemisia frigida)挥发性有机化合物(volatile organic compounds, VOCs)具有特殊气味, 在植物受损伤时, 此气味会更加浓烈。该文通过对未损伤与损伤冷蒿VOCs成分分析、地上部分结构观察, 初步揭示了冷蒿VOCs释放与结构之间的关系。结果表明, 未损伤冷蒿VOCs主要含有22种化合物, 其主要成分是莰烯(14.27%)、(E)-乙酸-3-己烯酯(10.85%)、对-伞花烃(9.05%)、桉树脑(39.80%)、α-萜品醇(10.04%)、β-萜品醇(2.48%)、樟脑(5.66%)和(R)-(-)-对薄荷-1-烯-4-醇(3.84%)。损伤较未损伤冷蒿VOCs增加了12种物质, 其中相对含量大于1%的化合物分别为顺-3-己烯醛(1.15%)、2-己烯醛(1.34%)、顺-牻牛儿醇(2.66%)、冰片(4.47%)、(1R,4R)(+)-对-薄荷-2,8-二烯(9.15%)、乙酸冰片酯(1.37%)和4(14), 11-桉叶双烯(1.30%)。冷蒿叶片中栅栏组织发达, 叶柄内具有2-3处栅栏组织, 并且栅栏组织中都具有发达的气室, 同时气室与气孔相连。因此, 损伤较未损伤冷蒿VOCs种类和浓度增多的原因可能为: 冷蒿VOCs合成后大量储存于气室中, 当叶片损伤时, VOCs大量释放出来, 同时合成释放一些新的VOCs, 致使损伤冷蒿VOCs种类和浓度增加。  相似文献

Three epitopes have been localized by immunoelectron microscopy on subunit Aa6 of the 4 x 6-meric hemocyanin of the scorpion Androctonus australis. Soluble immunocomplexes composed of monoclonal antibodies and of native hemocyanin were purified, negatively stained with uranyle acetate by the single-layer technique, and examined under the electron microscope (EM). The molecule images were digitized, aligned, and submitted to correspondence analysis according to the method of Van Heel and Frank (Ultramicroscopy 6:187-194, 1981). A high-precision localization of the attachment point of the Fab arm to the antigen was achieved through a careful analysis of the average images. This method easily allowed the discrimination of epitopes located in different domains (Mr 20 kDa) of the same subunit. Nonoverlapping epitopes located in the same structural domain of subunit Aa6 could be distinguished by the stain exclusion patterns of their Fab arms. The method is general and may be used for epitope mapping in any antigen producing definite EM views.  相似文献

Progress on reducing nutrient loss from annual croplands has been hampered by perceived conflicts between short‐term profitability and long‐term stewardship, but these may be overcome through strategic integration of perennial crops. Perennial biomass crops like switchgrass can mitigate nitrate‐nitrogen (NO3‐N) leaching, address bioenergy feedstock targets, and – as a lower‐cost management alternative to annual crops (i.e., corn, soybeans) – may also improve farm profitability. We analyzed publicly available environmental, agronomic, and economic data with two integrated models: a subfield agroecosystem management model, Landscape Environmental Assessment Framework (LEAF), and a process‐based biogeochemical model, DeNitrification‐DeComposition (DNDC). We constructed a factorial combination of profitability and NO3‐N leaching thresholds and simulated targeted switchgrass integration into corn/soybean cropland in the agricultural state of Iowa, USA. For each combination, we modeled (i) area converted to switchgrass, (ii) switchgrass biomass production, and (iii) NO3‐N leaching reduction. We spatially analyzed two scenarios: converting to switchgrass corn/soybean cropland losing >US$ 100 ha?1 and leaching >50 kg ha?1 (‘conservative’ scenario) or losing >US$ 0 ha?1 and leaching >20 kg ha?1 (‘nutrient reduction’ scenario). Compared to baseline, the ‘conservative’ scenario resulted in 12% of cropland converted to switchgrass, which produced 11 million Mg of biomass and reduced leached NO3‐N 18% statewide. The ‘nutrient reduction’ scenario converted 37% of cropland to switchgrass, producing 34 million Mg biomass and reducing leached NO3‐N 38% statewide. The opportunity to meet joint goals was greatest within watersheds with undulating topography and lower corn/soybean productivity. Our approach bridges the scales at which NO3‐N loss and profitability are usually considered, and is informed by both mechanistic and empirical understanding. Though approximated, our analysis supports development of farm‐level tools that can identify locations where both farm profitability and water quality improvement can be achieved through the strategic integration of perennial vegetation.  相似文献

Intention, Goal, Scope, Background  Input-Output Analysis (IOA) has recently been introduced to Life Cycle Assessment (LCA). In applying IOA to LCA studies, however, it is important to note that there are both advantages and disadvantages. Objectives  This paper aims to provide a better understanding of the advantages and disadvantages of adopting IOA in LCA, and introduces the methodology and principles of the Missing Inventory Estimation Tool (MIET) as one of the approaches to combine the strengths of process-specific LCA and IOA. Additionairy, we try to identify a number of possible errors in the use of IOA for LCA purposes, due to confusion between industry output and commodity, consumer’s price and producer’s price. Method  MIET utilises the 1996 US input-output table and various environmental statistics. It is based on an explicit distinction between commodity and industry output. Results and Discussion  MIET is a self-contained, publicly available database which can be applied directly in LCA studies to estimate missing processes. Conclusion  By adopting MILT results in existing, process-based, life-cycle inventory (LCI), LCA practitioners can fully utilise the process-specific information while expanding the system boundary. Requirements for mitigation of the continued increase in greenhouse gas (GHG ) emissions are much needed for the North China Plain (NCP ). We conducted a meta‐analysis of 76 published studies of 24 sites in the NCP to examine the effects of natural conditions and farming practices on GHG emissions in that region. We found that N2O was the main component of the area‐scaled total GHG balance, and the CH 4 contribution was <5%. Precipitation, temperature, soil pH , and texture had no significant impacts on annual GHG emissions, because of limited variation of these factors in the NCP . The N2O emissions increased exponentially with mineral fertilizer N application rate, with =  0.2389e0.0058x for wheat season and =  0.365e0.0071x for maize season. Emission factors were estimated at 0.37% for wheat and 0.90% for maize at conventional fertilizer N application rates. The agronomic optimal N rates (241 and 185 kg N ha?1 for wheat and maize, respectively) exhibited great potential for reducing N2O emissions, by 0.39 (29%) and 1.71 (56%) kg N2O‐N ha?1 season?1 for the wheat and maize seasons, respectively. Mixed application of organic manure with reduced mineral fertilizer N could reduce annual N2O emissions by 16% relative to mineral N application alone while maintaining a high crop yield. Compared with conventional tillage, no‐tillage significantly reduced N2O emissions by ~30% in the wheat season, whereas it increased those emissions by ~10% in the maize season. This may have resulted from the lower soil temperature in winter and increased soil moisture in summer under no‐tillage practice. Straw incorporation significantly increased annual N2O emissions, by 26% relative to straw removal. Our analysis indicates that these farming practices could be further tested to mitigate GHG emission and maintain high crop yields in the NCP .  相似文献

对双孢蘑菇CGMCC No. 0214及其基质降解能力退化菌株0214-3、0214-5进行了蛋白质双向电泳(2-DE或2D-PAGE)分析,发现了2个明显的、可重复的差异蛋白质,在退化菌株中分别为上调和下调表达,且2个退化菌株表现一致。根据质谱(MALDI-TOF/MS)分析和数据库检索结果,2个差异蛋白质初步鉴定为肌动蛋白和NADH脱氢酶铁硫蛋白3。  相似文献

对凤蝶科6属25种的COⅠ基因和20种Cyt b基因的部分序列进行测定和分析, 探讨它们之间的系统发育关系; 以茶小卷叶蛾Adoxophyes honmai为外群, 用邻接法(neighbor-joining, NJ)、 最大简约法(maximum parsimony, MP)和贝叶斯法(Bayesian inference, BI)重建了凤蝶科6属的分子系统树。结果表明: COⅠ基因部分序列长度为661 bp, 其中保守位点417个, 可变位点244个, 简约信息位点191个; A+T的平均含量为70.3%, 明显高于C+G的平均含量29.6%。Cyt b基因部分序列长度为433 bp, 其中保守位点239个, 可变位点194个, 简约信息位点135个; A+T的平均含量为74.2%, 明显高于C+G的平均含量25.7%。分子系统树表明, 凤蝶属Papilio、 斑凤蝶属Chilasa、 尾凤蝶属Bhutanitis、 珠凤蝶属Pachliopta和喙凤蝶属Teinopalpus为单系性, 与传统形态分类结果相一致。但青凤蝶属Graphium单系性不够明确, 需要进一步探讨。研究结果为我国凤蝶科分子系统学研究积累了资料。  相似文献

A major aim of landscape genetics is to understand how landscapes resist gene flow and thereby influence population genetic structure. An empirical understanding of this process provides a wealth of information that can be used to guide conservation and management of species in fragmented landscapes and also to predict how landscape change may affect population viability. Statistical approaches to infer the true model among competing alternatives are based on the strength of the relationship between pairwise genetic distances and landscape distances among sampled individuals in a population. A variety of methods have been devised to quantify individual genetic distances, but no study has yet compared their relative performance when used for model selection in landscape genetics. In this study, we used population genetic simulations to assess the accuracy of 16 individual‐based genetic distance metrics under varying sample sizes and degree of population genetic structure. We found most metrics performed well when sample size and genetic structure was high. However, it was much more challenging to infer the true model when sample size and genetic structure was low. Under these conditions, we found genetic distance metrics based on principal components analysis were the most accurate (although several other metrics performed similarly), but only when they were derived from multiple principal components axes (the optimal number varied depending on the degree of population genetic structure). Our results provide guidance for which genetic distance metrics maximize model selection accuracy and thereby better inform conservation and management decisions based upon landscape genetic analysis.  相似文献

为探索脑源性神经营养因子BDNF基因与中国汉族人群骨密度(bone mineral density, BMD)的关系,解析该基因调节骨质疏松症的功能机制,进而为中国汉族人群的骨质疏松症的防治提供依据。本研究从我国陕西地区征集了1300例汉族样本,并测量髋部/脊椎骨密度。选取BDNF基因上的14个标签SNPs进行基因分型,与1300例样本BMD进行关联分析,发现8个SNPs与髋部/脊椎BMD显著关联(P < 0.05)。其中,SNP rs16917237同时与髋部和脊椎骨密度关联,经Bonferroni校正后仍表现出显著性(0.05/14 = 0.0036)。整合连锁不平衡和单体型分析、表观功能注释、表达数量基因座分析、代谢通路分析进一步探索BDNF基因调节骨质疏松症的机制。构建小鼠前成骨细胞系(MC3T3-E1)人骨形态形成蛋白(rhBMP-2)诱导分化模型,利用小干扰RNA(siRNA)敲除BDNF。结果显示:14个SNPs位于同一单体型内;rs16917237在成骨细胞中表现出较强的激活型组蛋白H3K4me1、H3K4me3、H3K27ac修饰信号以及P300结合信号,表明其在成骨细胞中可能具有调控活性;rs16917237在11个组织中均能显著影响BDNF基因表达;BDNF基因位于与成骨细胞增殖分化相关的MAPK通路中;BDNF敲低能够显著降低MAPK通路中与成骨分化相关的CREB 基因mRNA和蛋白表达水平,提示其可能通过调控CREB表达进而影响成骨分化。生物信息学分析和功能实验结果一致,表明BDNF基因可能是影响骨质疏松症的重要功能基因。  相似文献
