Chemical sprout inhibitors are applied to over 50% of the potato harvest to extend storage time. In the U.S.A., CIPC (1-methylethyl-3-chlorophenylcarbamate) is the only synthetic chemical registered for postharvest sprout control of stored potatoes, and it is the most widely used sprout inhibitor world-wide. Due to environmental and health safety concerns, the use of CIPC has become more restricted, and alternative sprout control methods are sought. Six bacteria strains, exhibiting superior dry rot suppressiveness in previous research, were grown in two different liquid culture media and sprayed on Russet Burbank potatoes. In growth chamber and pilot experiments repeated at two storage sites in two successive years, all six isolates demonstrated significant sprout control capabilities when applied after growth on at least one of the culture media supplied. Of the six strains tested, Pseudomonas fluorescens bv.V S11:P:12 and two strains of Enterobacter sp., S11:T:07 and S11:P:08, exhibited highest relative performance levels with sprout control being statistically similar to that of 16.6 ppm CIPC thermal fog after 4-5 months storage.  相似文献

Mechanical loading of the spine is a major causative factor of degenerative changes and causes molecular and structural changes in the intervertebral disc (IVD) and the vertebrae end plate (EP). Pleiotrophin (PTN) is a growth factor with a putative role in bone remodeling through its receptor protein tyrosine phosphatase beta/zeta (RPTPβ/ζ). The present study investigates the effects of strain on PTN and RPTPβ/ζ protein expression in vivo. Tails of eight weeks old Sprague-Dawley rats were subjected to mechanical loading using a mini Ilizarov external apparatus. Rat tails untreated (control) or after 0 degrees of compression and 10°, 30° and 50° of angulation (groups 0, I, II and III respectively) were studied. PTN and RPTPβ/ζ expression were evaluated using immunohistochemistry and Western blot analysis. In the control group, PTN was mostly expressed by the EP hypertrophic chondrocytes. In groups 0 to II, PTN expression was increased in the chondrocytes of hypertrophic and proliferating zones, as well as in osteocytes and osteoblast-like cells of the ossification zone. In group III, only limited PTN expression was observed in osteocytes. RPTPβ/ζ expression was increased mainly in group 0, but also in group I, in all types of cells. Low intensity RPTPβ/ζ immunostaining was observed in groups II and III. Collectively, PTN and RPTPβ/ζ are expressed in spinal deformities caused by mechanical loading, and their expression depends on the type and severity of the applied strain.  相似文献

Impaired renal function has been suggested to significantly impact plasma midregional proADM (MR-proADM) level. The aim of this study was to assess whether improvement of renal function after living kidney transplantation has an impact on plasma MR-proADM-like immunoreactive substance (IS) level. Eleven patients with end-stage renal disease (ESRD) who were scheduled to undergo the first living kidney allograft transplantation were enrolled. Plasma MR-proADM-IS levels were measured before and 3, 7, 10, 14, 21, 30, 60 and 90 days after kidney transplantation. Plasma MR-proADM-IS level decreased significantly from day 3 after kidney transplantation compared to before kidney transplantation. A significant negative correlation was observed between creatinine clearance and plasma MR-proADM-IS level from before to 90 days after kidney transplantation (rs = −0.70, p < 0.0001). These results suggest that recovery of kidney function after kidney transplantation may lead to decrease in plasma MR-proADM level in patients with ESRD, and that plasma MR-proADM level may depend largely on renal function.  相似文献

DNA molecular weight standards (DNA markers, nucleic acid ladders) are commonly used in molecular biology laboratories as references to estimate the size of various DNA samples in electrophoresis process. One method of DNA marker production is digestion of synthetic vectors harboring multiple DNA fragments of known sizes by restriction enzymes. In this article, we described three novel strategies—sequential DNA fragment ligation, screening of ligation products by polymerase chain reaction (PCR) with end primers, and “small fragment accumulation”—for constructing complex synthetic vectors and minimizing the mass differences between DNA fragments produced from restrictive digestion of synthetic vectors. The strategy could be applied to construct various complex synthetic vectors to produce any type of low-range DNA markers, usually available commercially. In addition, the strategy is useful for single-step ligation of multiple DNA fragments for construction of complex synthetic vectors and other applications in molecular biology field. The fluxes of lead in submerged freshwater angiosperms, mosses and benthic algae were investigated by laboratory and field observations. Laboratory experiments showed that the initial uptake of lead by angiosperm shoots and Navicula sp. was rapid (up to 0.07 mg Pb g?1 dry weight min ?1 from a solution containing 1.0 mg Pb l?1), and over-whelmingly passive. Mosses were especially efficient at sorbing lead from solutions containing less than 1.0 mg Pb l?1 . Rates of uptake increased with rising external concentrations, but decreased with time. Anatomical and morphological characteristics of plant tissues affected metal flux and an epiphytic covering of Navicula (i.e., an Aufwuchs community) sorbed metal and reduced uptake into tissues by a third. Up to 90% of the lead sorbed in the first hour by shoots of Elodea canadensis Michx from a solution containing 1.0 mg Pb l?1 was released within 14 days of transfer into a lead-free medium; 10% was irreversibly bound.Moss monitors placed in Ullswater, a lake with sediments containing up to 26 mg Pb g?1 dry weight, confirmed that the metal is brought into circulation during turbulence, but not in periods of calm. Shoots of Elodea collected from the lake contained up to 0.3 mg g?1 dry weight non-exchangeable lead. Young shoots tended to contain less than older shoots (0.12 compared with 0.3 mg Pb g?1), and leaf more than stem tissues. The fluxes of lead from the environment through Aufwuchs and submerged plants are discussed.  相似文献

ObjectiveThe main causes for morbidity and mortality in von Hippel-Lindau (VHL) disease are central nervous system hemangioblastoma and clear cell renal cell carcinoma, but the effect of VHL-related pancreatic neuroendocrine tumors (PNET) on patient outcome is unclear. We assessed the impact of PNET diagnosis in patients with VHL on all-cause mortality (ACM) risk.MethodsWe used the Surveillance, Epidemiology, and End Results database. Of 16 344 patients, 170 had VHL based on clinical diagnostic criteria, and 510 patients had PNET (91 VHL-related and 419 sporadic).ResultsSurvival analysis demonstrated a lower ACM among patients with VHL-related PNET compared to patients with sporadic PNET (log-rank test, P = .011). Among patients with VHL, ACM risk was higher with vs without PNET (P = .029). The subgroup analysis revealed a higher ACM risk with metastatic PNET (sporadic P = .0031 and VHL-related P = .08) and a similar trend for PNET diameter ≥3 cm (P = .06 and P = 0.1 in sporadic and VHL-related PNET, respectively). In a multivariable analysis of patients with VHL, diagnosis with PNET by itself was associated with a trend of lower risk for ACM, while presence of metastatic PNET was independently associated with increased ACM risk.ConclusionDiagnosis with PNET is not associated with a higher ACM risk in VHL by itself. The independent association of advanced PNET stage with higher mortality risk emphasizes the importance of active surveillance for detecting high-risk PNET at an early stage to allow timely intervention.  相似文献
