Cell-suspension cultures of two chickpea (Cicer arietinum L.) cultivars, resistant (ILC 3279) and susceptible (ILC 1929) to the fungus Ascochyta rabiei (Pass.) Lab., showed differential accumulation of the phytoalexins medicarpin and maackiain, and transient induction of related enzyme activities after application of an A. rabiei-derived elicitor. The chalcone-synthase (CHS) activity (EC 2.3.1.74) which is involved in the first part of phytoalexin biosynthesis exhibited a maximum 8–12 h after elicitation in the cells of both cultivars. Concomitant with the fivefold-higher phytoalexin accumulation, CHS activity increased twofold in the cells of the resistant cultivar. The maximum of the elicitor-induced CHS-mRNA activity was determined 4 h after onset of induction in the cultures of both cultivars, although in cells of cultivar ILC 3279 this mRNA activity was induced at a level twofold higher than that in cells of the susceptible race ILC 1929. Investigations of CHS isoenzymes by two-dimensional gel electrophoresis of immunoprecipitated in-vitro-translated protein indicated the presence of five proteins. In the cells of both cultivars only two of the isoenzymes were induced after elicitor treatment. Analysis of the total in-vitro-translated proteins by two-dimensional gel electrophoresis showed that the constitutively expressed patterns of mRNA activities in the cell cultures of the two cultivars were identical. After elicitation, considerably more translatable mRNAs were induced in the cells of cultivar ILC 3279. The few induced proteins, and their respective mRNA activities, which could be detected in the cells of the susceptible cultivar, all existed in the cells of the resistant cultivar, too. One highly induced protein (Mr 18 kDa) found in the cells of cultivar ILC 3279 reached its maximum mRNA activity 6 h after elicitor application. The amount of this protein was hardly increased in the cells of the susceptible cultivar. This protein appears to be excreted from the cells into the growth medium.Abbreviations CHS

Summary Empirical results routinely demonstrate that the reduced Additive Main effects and Multiplicative Interaction (AMMI) model achieves better predictive accuracy for yield trials than does the full treatment means model. It may seem mysterious that treatment means are not the most accurate estimates, but rather that the AMMI model is often more accurate than its data. The statistical explanation involves the Stein effect, whereby a small sacrifice in bias can produce a large gain in accuracy. The corresponding agricultural explanation is somewhat complex, beginning with a yield trial's design and ending with its research purposes and applications. In essence, AMMI selectively recovers pattern related to the treatment design in its model, while selectively relegating noise related to the experimental design in its discarded residual. For estimating the yield of a particular genotype in a particular environment, the AMMI model uses the entire yield trial, rather than only the several replications of this particular trial, as in the treatment means model. Abstract. Mixed convection (forced convection plus free convection) in the leaf boundary layer was examined by air flow visualization and by evaluation of the boundary layer conductance at different leaf-air temperature differences ( T L - T A ) under low wind velocities. The visualized air flow was found to become more unstable and buoyant at higher T L - T A . An ascending longitudinal plume was induced along the upper surface, and the air flow along the lower surface ascended after passing the trailing leaf edge. The air flow modified by buoyancy was considered to result in an increase in boundary layer conductance ( G A ) for mixed convection, which became higher with higher T L - T A as compared with the conductance for pure forced convection without buoyancy. This increase in G A appeared larger at larger Grashof number (Gr) and at smaller Reynolds number (Re). The dependences of buoyancy effect on Gr and Re were related to 'edge-effects'. 相似文献

The activity of phosphoglucose isomerase, its kinetic properties, and the effect of 6-phosphogluconate on its activity in the forward (glucose 6-phosphate----fructose 6-phosphate) and the reverse (fructose 6-phosphate----glucose 6-phosphate) reactions were determined in adult rat brain in vitro. The activity of phosphoglucose isomerase (in nmol/min/mg of whole brain protein) was 1,865 +/- 20 in the forward reaction and 1,756 +/- 32 in the reverse reaction at pH 7.5. It was 1,992 +/- 28 and 2,620 +/- 46, respectively, at pH 8.5. The apparent Km and Vmax of phosphoglucose isomerase were 0.593 +/- 0.031 mM and 2,291 +/- 61 nmol/min/mg of protein, respectively, for glucose 6-phosphate and 0.095 +/- 0.013 mM and 2,035 +/- 98 nmol/min/mg of protein, respectively, for fructose 6-phosphate. The activity of phosphoglucose isomerase was inhibited intensely and competitively by 6-phosphogluconate, with an apparent Ki of 0.048 +/- 0.005 mM for glucose 6-phosphate and 0.042 +/- 0.004 mM for fructose 6-phosphate as the substrate. With glucose 6-phosphate as the substrate, at concentrations from 0.05 to 0.5 mM, the activity of the enzyme was inhibited completely in the presence of 0.5-2.0 mM 6-phosphogluconate. With 0.05-0.2 mM fructose 6-phosphate as the substrate, it was inhibited greater than or equal to 85% at the same concentrations of the inhibitor. The decrease in the electron flow of the aerobic respiratory chain of the bacterium Paracoccus denitrificans, owing to either the drop in the saturation of terminal oxidases by oxygen or to the inhibition of the rate of respiration by azide or nitrite, resulted in the synthesis of dissimilatory nitrate reductase and nitrite reductase. The dependence of the resulting activities of the two enzymes (after a three-hour adaptation) on the initial value of the parameter Vmax/kLa (oxidase activity of the volume unit of the culture divided by the volumetric oxygen transfer coefficient) or on the concentrations of the inhibitors had a similar form, characterized by the appearance of a maximum. The increasing parts of the obtained curves reflect the synthesis of enzymes, probably initiated by the increase in the intracellular degree of reduction, the subsequent drop being evidently in connection with the lack of metabolic energy for biosynthesis. The possible mechanisms of the effect of nitrogenous terminal acceptors (NO-3 and NO-2) on the formation of the denitrification pathway are discussed. 相似文献

Hyperpolarization-activated K channels (K
H
channels) in the plasmalemma of guard cells operate at apoplastic pH range of 5 to over 7. Using patch clamp in a whole-cell
mode, we characterized the effect of varying the external pH between 4.4–8.1 on the activity of the K
H
channels in isolated guard cell protoplasts from Vicia faba leaves.
Acidification from pH 5.5 to 4.4 increased the macroscopic conductance of the K
H
channels by 30–150% while alkalinization from pH 5.5 to 8.1 decreased it only by roughly 15%. The voltage-independent maximum cell conductance, increased by ∼60% between pH 8.1 and 4.4 with an apparent pK
a
of 5.3, most likely owing to the increased availability of channels. Voltage-dependent gating was affected only between pH 5.5 and 4.4. Acidification in this range shifted the voltage-dependent open probability by over 10 mV. We interpret this shift as an increase of the electrical field sensed by the gating subunits
caused by the protonation of external negative surface charges. Within the framework of a surface charge model the mean spacing
of these charges was ∼30 ? and their apparent dissociation constant was 10−4.6. The overall voltage sensitivity of gating was not altered by pH changes. In a subgroup of protoplasts analyzed within the
framework of a Closed-Closed-Open model, the effect of protons on gating was limited to shifting of the voltage-dependence
of all four transition rate constants.

We investigated the influence of mesohabitats on fish communities and on attributes of a multimetric index of fish integrity in the River Meuse basin. Three consecutive 150 m sectors in a Meuse tributary (Ourthe, Belgium), each divided in two or three sub-sections presenting various percentages of mesohabitats (riffles, runs and pools), were sampled by electrofishing. In each sub-section, relative and absolute biomasses of each fish species were estimated. The presence of limnophilic cyprinids was inversely correlated (r
2 = 0.70 and 0.56 for absolute and relative biomass, respectively) with the percentage of riffles. Salmonids preferred runs and their absolute biomass was highly dependent (r
2 = 0.71) on the proportion of this mesohabitat, whereas biomass of limnophilic cyprinids was highly correlated (r
2 = 0.75 and r
2 = 0.82 for absolute and relative biomass, respectively) with pools. A positive correlation (r
2 = 0.58) was established between relative biomass of predators and the percentage of this mesohabitat. An IBI was calculated for the three sectors on the basis of results from the entire Meuse catchment. Scores of most metrics showed low variation among sectors but values of two metrics (% of individuals as tolerant, % of individuals as ubiquitous spawners) were greatest in sector 2, where pools predominated. Further, the lowest IBI score (51/65, integrity class: fair to good) was recorded in sector 2 where pools dominated, while sector 1 (where runs dominated) obtained the highest score (63/65, integrity class: excellent). Sector 3 which has a balanced proportion of riffles and runs obtained an intermediate score (57/65). Considering the response of IBI to the natural variation of mesohabitat proportions, it appears that an accurate sampling requires the prospection of a variety of mesohabitats (with a majority of runs) for the evaluation of river quality. 相似文献
