The formation of pores by membrane-inserted diphtheria toxin is closely linked to the translocation of its catalytic chain across membranes. In this report a number of aromatic polyanionic molecules were identified that inhibit toxin-induced leakage of molecules from model membrane vesicles. One inhibitor, Cibacron blue, totally blocked pore formation. Aniline blue and Fast Green decreased the size of the molecule released by a given concentration of toxin. Amaranth appeared to reduce the maximal amount of leakage, without greatly affecting the size of the molecule released at a given toxin concentration. Finally, Ponceau S and Cibacron brilliant red appeared to exhibit a mixture of these various types of inhibition. The inhibitors neither prevented the conformational transition of the toxin to form a hydrophobic state at low pH, nor (with the exception of Cibacron Brilliant Red) appeared to strongly inhibit toxin binding to model membranes. Additional experiments showed release of trapped materials from model membranes by isolated T domain of the toxin was similar to that by whole toxin. The effects of inhibitors on T domain induced release was also similar to that they have on whole toxin. Therefore, it is likely that the inhibition of pore formation by whole toxin involves inhibitor interaction with the T domain. The inhibitors identified in this study may be helpful for development of agents that interfere with toxin action in vivo. Received: 10 March 1999/Revised: 22 June 1999  相似文献

  • 1 Plants produce natural enemy‐attracting semiochemicals known as herbivore‐induced plant volatiles (HIPV) in response to herbivore damage. Deployment of synthetic HIPV in crops could enhance the biological control of pests. To test this, six HIPV [methyl salicylate (MeSA), methyl anthranilate (MeA), methyl jasmonate (MeJA), benzaldehyde (Be), cis‐3‐hexenyl acetate (HA), cis‐hexen‐1‐ol (He)] in three concentrations (0.5%, 1.0% and 2.0% v/v) mixed with a vegetable oil adjuvant, Synertrol® (Organic Crop Protectants Pty Ltd, Australia), were sprayed onto winegrape, broccoli and sweet corn plants.
  • 2 The relative abundance of insects within treated plots was assessed with non‐attracting, transparent sticky traps at varying time intervals up to 22 days after spraying.
  • 3 In the vineyard experiment, Trichogrammatidae responded to Be and MeA (0.5%) and Be (1.0%); Encyrtidae and Bethylidae responded to MeA (1.0%); Scelionidae responded to all compounds at 1.0% and 2.0%; and predatory insects responded to MeA. In sweet corn, parasitoids as a group and Encyrtidae responded to MeA (0.5%); Braconidae responded to all compounds at 0.5% and Synertrol‐only; thrips responded to all compounds at 0.5% and 1.0%; while all parasitoids responded to all compounds at 0.5% and 1.0% and Synertrol‐only. In broccoli, parasitoids as a group and Scelionidae responded to Be, HA, He and Synertrol‐only; Trichogrammatidae responded to Be (0.5%), He (0.5% and 1.0%), MeJA (1.0%) and MeSA (0.5%); and thrips responded to all compounds at to 0.5% and 1.0%.
  • 4 Significant attraction of insects occurred up to 6 days after the HIPV application, suggesting that plants may have been induced to produce endogenous volatiles that attracted insects over an extended period.
  • 5 The results obtained are discussed in relation to the potential utility of synthetic HIPV to enhance the biological control of pests.

Myrtaceae family (121 genera, 3800-5800 spp.) is one of the most important families in tropical forests. They are aromatic trees or shrubs, which frequently produce edible fruits. In the neotropics, ca. 1000 species were found. Several members of this family are used in folk medicine, mainly as an antidiarrheal, antimicrobial, antioxidant, cleanser, antirheumatic, and anti-inflammatory agent and to decrease the blood cholesterol. In addition, some fruits are eaten fresh or used to make juices, liqueurs, and sweets very much appreciated by people. The flavor composition of some fruits belonging to the Myrtaceae family has been extensively studied due to their pleasant and intense aromas. Most of the essential oils of neotropical Myrtaceae analyzed so far are characterized by predominance of sesquiterpenes, some with important biological properties. In the present work, chemical and pharmacological studies carried out on neotropical Myrtaceae species are reviewed, based on original articles published since 1980. The uses in folk medicine and chemotaxonomic importance of secondary metabolites are also briefly discussed.  相似文献

The success of biological control is partly mediated by the longevity and reproductive success of beneficial insects. Availability of nectar and honeydew can improve the nutrition of parasitic insects, and thereby increase their longevity and realized fecundity. The egg parasitoid, Anaphes iole, showed strong gustatory perception of trehalulose, a carbohydrate found in homopteran honeydew. Chromatographic analysis demonstrated that enzymatic hydrolysis of sucrose, a common nectar sugar, proceeded at a faster rate than that of melezitose, a sugar common in aphid honeydew. A long-term bioassay showed that longevity was greater at 20 °C than at 27 °C, and at both temperatures survival was generally greatest for wasps provisioned with the three major nectar sugars, sucrose, glucose, and fructose. Patterns of food acceptance and utilization showed that A. iole accepted and utilized a broad range of sugars found in nature, including those found in nectar as well as honeydew. Glucose, fructose, and several oligosaccharides composed of these monosaccharide units appear to be more suitable for A. iole than other sugars tested. Evidence suggests that individual fitness benefits afforded by food sources are important for a time-limited parasitoid, and that continued investigations on the interface between nutrition and biological control are warranted for A. iole.  相似文献

St Lucia Lake on the north coast of Natal, South Africa, has an area of 325 km2 and is the largest estuarine complex in Africa. It consists of a 20 km tidal channel, averagingca. 400 m in width, linking the sea with the non-tidal lake which is H-shaped with a maximum length ofca. 40 km and width ofca. 20 km. Except during flood periods the depth of the lake does not exceed 2 m. The salinity gradient depends on evaporation, the configuration of the mouth and on the input of fresh water from four rivers which discharge into the northern and western areas of the lake. If fresh water input is high, the lake and much of the channel may be fresh. An intermediate stage features a normal salinity gradient while a third stage shows a reversed salinity gradient with salinities in excess of 100‰ in the upper reaches of the system. Changing salinities have marked effects on the biota. Aquatic macrophytes show cycles of appearance and disappearance depending on salinity tolerance and the presence of dormant stages. The resident benthic faunal species go through cycles of range expansion and contraction depending on prevailing salinities and recolonisation by dispersal phases. To date salinities in the southern part of the lake have approached, but not exceeded, lethal levels and this has therefore acted as a reservoir area. Catchment degradation and water abstraction are anticipated to exacerbate future salinity extremes. This has resulted in concern for the long term viability of this Ramsar site which has major southern African populations of hippopotamus and crocodile, provides breeding sites for South African Red Data water bird species and plays an important nursery role for marine fish and penaeid prawns.  相似文献

The retroviral Gag polyprotein mediates viral assembly. The Gag protein has been shown to interact with other Gag proteins, with the viral RNA, and with the cell membrane during the assembly process. Intrinsically disordered regions linking ordered domains make characterization of the protein structure difficult. Through small-angle scattering and molecular modeling, we have previously shown that monomeric human immunodeficiency virus type 1 (HIV-1) Gag protein in solution adopts compact conformations. However, cryo-electron microscopic analysis of immature virions shows that in these particles, HIV-1 Gag protein molecules are rod shaped. These differing results imply that large changes in Gag conformation are possible and may be required for viral formation. By recapitulating key interactions in the assembly process and characterizing the Gag protein using neutron scattering, we have identified interactions capable of reversibly extending the Gag protein. In addition, we demonstrate advanced applications of neutron reflectivity in resolving Gag conformations on a membrane. Several kinds of evidence show that basic residues found on the distal N- and C-terminal domains enable both ends of Gag to bind to either membranes or nucleic acid. These results, together with other published observations, suggest that simultaneous interactions of an HIV-1 Gag molecule with all three components (protein, nucleic acid, and membrane) are required for full extension of the protein.  相似文献

Summary Freeze-etch preparations of mesothelial cells taken from the peritoneum of mouse reveal the presence of vesicles invaginating the apical and the basal cell surfaces. These vesicles are scarcely seen within the cytoplasm. Long tortuous tubular profiles extend for considerable distance within the cytoplasm and are frequently associated with the vesicles. The possible nature and role of the vesicles and the tubules in transport phenomena across the mesothelial barrier, are discussed in relation to the pore theory advanced by physiologists and the stomata concept observed by early German and contemporary anatomists. Occludens junctions of the leaky type are seen though their macular or zonular nature is yet to be established.  相似文献

Prior research indicated the ability of concentrated metabolites from Xenorhabdus spp. and Photorhabdus spp. to suppress a variety of peach and pecan diseases in vitro, and on detached pecan leaves or terminals. In the current study, our objectives were to (1) determine if bacterial broths (in addition to concentrated metabolites tested previously) have suppressive ability and (2) determine if metabolites or bacterial broths are active in a soil medium. In laboratory studies, two pathogens of pecan (Fusicladium effusum and Phytophthora cactorum) and one peach pathogen (Armillaria tabescens) were tested for susceptibility to Xenorhabdus bovienii (SN) and Photorhabdus luminescens (VS) bacterial broths or concentrated metabolites on three different substrates. Treatments were applied to lesions of F. effusum on terminals to ascertain any suppressive effect on sporulation, to A. tabescens in soil to determine effect on survival of mycelia, and to lesions caused by P. cactorum on pecan leaf surfaces to assess any reduction in lesion development. Acetone (the metabolite solvent), un-inoculated media (tryptic soy broth) and water were included as controls. The X. bovienii metabolite treatment was as efficacious as a commercial fungicide (fenbuconazole) in reducing sporulation of F. effusum on pecan terminals. The P. luminescens metabolite treatment also caused reduced sporulation relative to water and acetone controls but bacterial broths had no effect. In contrast, all bacterial broth and metabolite treatments suppressed lesion growth caused by P. cactorum (measured on detached leaves maintained on agar). However, in soil, only the P. luminescens metabolite treatment was suppressive to A. tabescens (this is the first report of Photorhabdus or Xenorhabdus toxicity to Armillaria spp.). This study provides a basis for further research on the use of Xenorhabdus and Photorhabdus metabolites or bacterial broth for suppression of pecan and peach diseases.  相似文献
