Summary The cloning, sequencing and mutational analysis of the Bradyrhizobium japonicum symbiotic nitrogen fixation genes fixL and fixJ are reported here. The two genes were adjacent and probably formed an operon, fixLJ. The predicted FixL and FixJ proteins, members of the two-component sensor/regulator family, were homologous over almost their entire lengths to the corresponding Rhizobium meliloti proteins (approx. 50% identity). Downstream of the B. japonicum fixJ gene was found an open reading frame with 138 codons (ORF138) whose product shared 36% homology with the N-terminal part of FixJ. Deletion and insertion mutations within fixL and fixJ led to a loss of approximately 90% wildtype symbiotic nitrogen fixation (Fix) activity, whereas an ORF138 mutant was Fix+. In fixL, fixJ and ORF138 mutant backgrounds, the aerobic expression of the fixR-nifA operon was not affected. NifA itself did not regulate the expression of the fixJ gene. Thus, the B. japonicum FixL and FixJ proteins were neither involved in the regulation of aerobic nifA gene expression nor in the anaerobic NifA-dependent autoregulation of the fixRnifA operon; rather they appeared to control symbiotically important genes other than those whose expression was dependent on the NifA protein. The fixL and fixJ mutant strains were unable to grow anaerobically with nitrate as the terminal electron acceptor. Therefore, some of the FixJ-dependent genes in B. japonicum may be concerned with anaerobic respiration.  相似文献

Summary Parameters estimated from a Gardner-Eberhart analysis of the F2 generation of a six-parent diallel in oats (Avena sativa L.) were used to compare methods for predicting the performance of F3 row plots. The prediction methods were: (1) individual F2 plant performance (F2I), (2) parent average plus F2 plot deviations (PF2), (3) parent average plus weighted F2 plot deviations (PF2P), (4) best linear unbiased prediction (BLUP) of parent average plus F2 plot deviations (BPF2), and (5) BLUP plus weighted F2 deviations (BF2). The F2 single-plant traits used for prediction were biological yield to predict F3 biological yield, whole plant and primary tiller grain yield for prediction of F3 grain yield, and whole plant and primary tiller harvest index (HI) to predict F3 HI. Prediction methods were evaluated by correlations between predicted and observed F3 performance. Prediction methods and traits for which correlations were greater than for F2I included: BF2 for biological yield, PF2, PF2P and BF2 for whole plant grain yield, PF2, BPF2, and BF2 for primary tiller grain yield. None had a correlation significantly greater than F2I for either measure of HI, where heritability was large. PF2 is the recommended method for traits with low heritability because of its simplicity and because it had the largest or nearly the largest correlation for each of the yield traits. Abstract. Studies of the isoprene emission rate in response to changes in photon-flux density and CO2 partial pressure were conducted using a recently developed on-line isoprene analyser combined with a gas exchange system and chlorophyll fluorometer. Upon darkening, the isoprene emission rate from leaves of aspen ( Populus tremuloides Michaux.) began to decline immediately, demonstrating that the internal pool of isoprene, or its precursors, is small and that the instantaneous emission rate is tightly coupled to the rate of synthesis. A post-illumination burst of isoprene was observed within 5 min after darkening and lasted for 15–20 min in four isoprene-emitting species that were examined. In leaves of eucalyptus ( Eucalyptus globulus Labill.), the magnitude of the post-illumination burst was dependent on the photon-flux density that existed before darkening, but not on ambient CO2 partial pressure. The dependence of the post-illumination burst on photon-flux density paralleled that for the steady-state rate of isoprene emission. A step-wise increase in intercellular CO2 partial pressure from 24.5 to 60 Pa resulted in an immediate decrease in isoprene emission rate and non-photochemical fluorescence quenching, but an increase in CO2 assimilation rate. Given the several recent studies that link isoprene emission to chloroplastic processes, the results of this study indicate that the linkage is not dependent on the rate of CO2 flux through the reductive pentose phosphate pathway, but rather on more complex relationships involving metabolites not appreciably influenced by CO2 partial pressure.  相似文献

Summary White pine (Pinus strobus L.) trees have shoot populations composed of subpopulations of terminal and lateral shoots. I tested whether the subpopulations would show compensatory (increased) growth when separated from each other. Ten-year-old white pine (Pinus strobus L.) trees growing under an oak (Quercus) overstory were untreated or treated in winter by removing either all terminal, or all lateral buds (10 trees per treatment). Growth was compared between control and treated shoot subpopulations. In the 1st year, shoot-length frequency distributions were similar between control and treated subpopulations. There was significant compensatory shoot elongation (mean of 1.5 cm per shoot) in both treated subpopulations. In the 2nd year each subpopulation produced both terminal and lateral shoots. Shoot-length frequency distributions were similar, but shifted toward longer shoots in treated populations. Shoot number, mean length and total shoot length were greater in treated populations. The increased growth in treated subpopulations was due both to differences in parent shoot length and to compensatory shoot production and elongation.  相似文献

The efficacy of 2‐furfuraldehyde for control of Sclerotium rolfsii was studied in laboratory and greenhouse experiments. Mycelial growth of the fungus was reduced proportionally with concentrations of 0.1–0.5 ml furfuraldehyde l‐1 agar medium, and viability of sclerotia diminished on exposure to 2‐furfuraldehyde vapours. Detectable populations of bacteria and fungi, including Trichoderma spp., were reduced significantly (9=0.05) when furfuraldehyde was added to the agar used for soil dilution plates of untreated soil. Repeated treatments of natural soil with the fumigant significantly increased populations of Trichoderma spp. and bacteria, but diminished numbers of actinomycetes. Increasing dosages applied to soil artificially infested with S. rolfsii caused a reduction of disease on lentil, Lens culinaris. Results indicate that the compound, when applied to field soil, changes the composition of soil microflora and has potential for integrated control of S. rolfsii.  相似文献

The weevil Sitona regensteinensis is being considered for biological control of Cytisus scoparius in New Zealand. Adult feeding tests demonstrated that while foliage of C scoparius was preferred, weevils fed also on Chamaecytisus palmensis and Lupinus arboreus. Agrotis segetum nuclear polyhedrosis virus (AsNPV) and granulosis virus (AsGV), propagated in laboratory cultures of A. segetum in England and A. ipsilon in Spain, respectively, were applied to plots of maize plants at the one‐ to four‐leaf stage of growth. Plots were arranged in a 6 x 6 Latin square design and infested with second‐instar A. segetum larvae (the common cutworm). Each virus was applied in separate treatments by two application methods; as an aqueous spray containing 0.1% Agral as a wetting agent, and as a bran bait. The NPV was applied at a rate of 4 X 1012 polyhedra/ha, and the GV at 4 X 1013 granules/ha. Soil and plants were sampled for larvae on three occasions following virus treatment: 24 h, 4 days and 11 days. The larvae were reared on diet in the laboratory, until death or pupation, to examine the rate and level of viral infection. Infection data showed 87.5% and 91% NPV infection and 12.5% and 55% GV infection in spray and bait treatments, respectively, in larvae sampled 24 h after treatment. In larvae sampled 4 days after treatment, the results were 78% and 100% NPV infection, and 13% and 6% GV infection. A total of only six larvae were retrieved on day 11. In both treatments larvae infected with AsNPV died significantly more rapidly and at an earlier instar than those infected with AsGV, indicating that AsNPV appears to have better potential as a control agent for A. segetum.  相似文献

本文利用青蛙(Rana nigromaculata Hallowell)蝌蚪红细胞微核试验,作为检测城镇污水诱变活性的一种新的监测技术。在16d生活污水处理的实验中,青蛙蝌蚪红细胞微核细胞率2d后就呈现统计上的显著增加,并随处理时间的延长而增高,第12d达到最大值。在不同浓度混合污水处理实验中,蝌蚪红细胞微核细胞率呈现明显的剂量依赖性增加。上述实验证明城镇生活污水和混合污水都具有较强的诱变活性。作者从遗传毒理学的角度评价了湖北黄州综合生物塘系统对污水诱变活性的净化效能。城镇混合污水经综合生物塘各级塘处理,蝌蚪微核细胞率逐级下降,由进水的7.54‰下降到最后出水的1.52‰,接近对照(1.07‰)水平。其中综合生物塘的藻菌单元比水生植物单元对污水诱变活性具有更强的净化效力。本文提出污水“诱变指数”可作为综合生物塘一项功能评价指标。  相似文献
