Two strains of Drosophila melanogaster represent the extremes in resistance and sensitivity to the lethal effects of CdCl2. The strain containing the mutations vermilion and brown (v; bw) and the strain Austin had LC50's of 3.3 and 1.3mm CdCl2, respectively. The three major chromosomes from these two strains were assorted genetically into the six possible combinations. The measured LC50's for CdCl2 for these six genotypes fell into two groups according to the X chromosome; those containing the X chromosome from v; bw had LC50's 0.5–1.0mm greater than those in which the X chromosome was from Austin. Since the parent strains differed by 2mm, we suggest that the X chromosome is a major, but not the sole, site of genes to produce resistance to CdCl2. When 109Cd was in the diet the uptake by v; bw and Austin over 2 days was the same. After 4 days of uptake, the Austin strain excreted the 109Cd five times faster than v; bw but the six genotypes did not differ appreciably in excretion rate from one another and resembled the sensitive parent Austin more than the resistant one. Summary Field experiment was conducted to show the effect of one irrigation of 10 cm water applied to durum wheat (Triticum durum) at crown root initiation stage, active tillering stage, anthesis stage and compared them with non irrigated plot using A-206, A-28, A-1-8-8-42-1, G.W.-1 and A-23-10 genotypes under the conditions of salt affected areas having shallow saline aquifer, on quality, yield and mottling phenomena in wheat grain. Application of irrigation significantly increased the yield and decreased the per cent mottled grain and the maximum achievement was obtained when water was applied during the stage of anthesis. Non mottled grain contained higher protein than mottled grain and there were no considerable differences in other biochemical constituents. Genotype G.W.-1 stands first in its yield performance and low mottled grain.  相似文献

Summary The use of several S1 individuals to represent an S0 individual permits the use of a Design II mating scheme for plants with only one pistillate flower per plant. Estimates of additive (V A ) and dominance (V D ) variance from this mating scheme will be biased upwards, when a small number (10) of individuals of each S1 line are used. This bias can be computed, and the additive and dominance estimates can be corrected. Of particular interest is the observation that the additive genetic variance contributes to bias in estimates of V D . When S0 plants are non inbred and their selfedprogeny (S1 lines) are used to represent them in developing families for use in the Design II, where m1 is the number of individuals used to represent an S1 line in developing half sib-families and m2 is the number of individuals used to represent the S1 line in making up full sib-families. For example, in a 3×3 Design II, with about 10 individuals used to represent each S1 line in each cross, m2 = 10 and m1 = 30. Summary Two 30 g/ml BrdU-resistant sublines and two 60 g/ml BrdU-resistant sublines are induced from a Chinese hamster cell line Wg3h (HGPRT) by one-step and two-step selections, respectively. By inoculating the cells into BrdU-free medium or by adding more BrdU into the culture medium for 26–27 h, it was found that the two BrdU-resistant sublines analysed have very clear sister chromatid differential (SCD) staining patterns. This indicates that some of the nuclear DNA of the BrdU-resistant cells incorporate with BrdU to reach a kinetic balance. Frequencies of sister chromatid exchange (SCE) of the resistant cells are twice to four times as high as those of the Wg3h cells, depending on which BrdU-resistant subline is analysed. The SCE frequencies of the resistant cells also increase with the BrdU concentration in the medium. Analysis of silver-stained nucleolar organizer regions (NORs) indicates that the NOR activity of three out of the four BrdU-resistant sublines is significantly suppressed, i.e., averages of the Ag-NOR number and number of the chromosomes bearing Ag-NORs per cell decrease significantly. The degree of suppression for different BrdU-resistant sublines may be quite different. The suppressed NOR activity of the resistant cells can gradually be restored when the cells are inoculated into BrdU-free medium, but the recovery speed is far lower than that of the Wg3h cells. The suppression of the NOR activity of the BrdU-resistant sublines should be due to BrdU toxicity.  相似文献

Genetic variation is described at 15 loci in 2 neotenic and 12 nonneotenic populations of red-spotted newts. Though high levels of genetic similarity (I=0.990) were found among all populations, allele frequencies at six of the eight most polymorphic loci show significant heterogeneity across populations. Change in allele frequencies at two of these loci (Pep-2 and Ldh-1) is significantly correlated with latitude. Interspecific homologies are established for newt peptidases based on substrate specificities and lactate dehydrogenases based on tissue distribution, thermal stability, and kinetic properties. Nonneotenic populations are highly variable (H=0.157) and neotenic populations are only slightly, but significantly, less variable (H=0.120). The high levels of heterozygosity detected in nonneotenic populations may result from large effective population size and/or environmental heterogeneity. The unexpectedly high heterozygosity values obtained for the neotenic populations may indicate adult dispersal or the presence of some previously undetected red efts at these localities. In any case, a major change in life history has apparently had little effect on the genetic structure of these populations.This research was supported by grants from the Blakeslee Fund of Smith College.  相似文献

Genetic population structure was studied in two types of populations in the ants Formica exsecta and F. pressilabris: populations consisting of single-nest colonies (monodomy) and populations consisting of multi-nest colonies (polydomy). These characteristics seem to be associated with the number of egg-laying females (gynes) in a nest, mating structure of the population, sex ratio and male size variation. The monodomous populations are characterized by single-gyne nests, the population sex ratio is either I:1 or female-biased, males are mainly large-sized, and there is slight inbreeding in the population. The polydomous populations have multi-gyne nests with gynes related to each other, sex ratio is strongly male-biased, most males are small-sized, and there is slight genetic microdifferentiation within the populations. Diploid males found in a polydomous F. pressilabris population suggest that the population is inbred and isolated. Habitat localization is presented as a plausible explanation for the evolution of the polygynous and polydomous population structure.  相似文献

Summary The coculture of mouse PG19 cells with human MGC cells can significantly suppress nucleolar organizer region (NORs) activity of both PG19 and MGC cells. 5-bormodeoxyuridine (BrdU) can also significantly suppress the NOR activity of rat RC cells, human MGC and Hela cells, and mouse PG19 cells: i.e. the average number of Ag-NORs and the number of chromosomes bearing Ag-NORs per cell decrease significantly. The degree of the suppression increases with increase in both BrdU concentration in the culture medium and BrdU treatment time. The suppressed NOR activity of the PG19 cells can gradually be restored when the BrdU-treated cells are transferred into BrdU-free medium for 50 h. In PG19 cells deoxycytidine (dC) can reverse the suppression of NOR activity caused by BrdU. Coculture plus BrdU treatment suppress the NOR activity of PG19 cells more severely than BrdU treatment alone. In coculture medium containing 30 g BrdU/ml, dC can also reverse the suppression of the NOR activity of PG19 cells but not that of the MGC cells. The degree of the reversion in the coculture plus BrdU treatment is significantly lower than that found with BrdU-treatment alone.  相似文献

Analysis of genetic variation between the banana and the citrus races of Radopholus similis by starch gel eleclrophoresis demonstrated that 7 of 16 enzyme-encoding loci could be used for their diagnostic separation. The two races are closely related arid share approximately 75% of the enzymes evaluated. The level of dissimilarities o1 inherited bands indicates that no gene flow occurs between the races. Aldolase, α + β esterase, glucose-6-phosphate dehydrogenase, isocitrate dehydrogenase, lactate dehydrogenase, malate dehydrogenase, and phosphoglucose isomerase are diagnostic markers of the races.  相似文献

Specific gravity (sp. gr.) of cell-free hemolymph and gut and coxal fluids was determined at different states of the gonotrophic cycle (unfed + 15 days, engorgement day before and after coxal fluid emission, engorgement + 1 day, oviposition day, and oviposition completion + 1 day) of female Argas (Persicargas) persicus and A. (P.) aboreus (Argasidae). The patterns of hemolymph and gut fluid sp. gr. change differed from each other during the gonotrophic cycle, but both patterns were similar in the 2 Argas species. Hemolymph sp. gr. decreased to a minimum one day after feeding (1.0085 and 1.0081 for persicus and arboreus, respectively), and increased through oviposition to a maximum on oviposition + 1 day (1.0187 and 1.0221). Minimum gut fluid sp. gr. occurred on engorgement day before coxal fluid emission (1.0565 and 1.0697). Afterward, gut fluid sp. gr. increased to a maximum on engorgement day + 1 for persicus (1.1089) and on oviposition day for arboreus (1.0973), and then decreased during oviposition in both species. In each tested state of each species, the sp. gr. was consistently higher in gut fluid than in hemolymph. In each species, coxal fluid and hemolymph sp. gr. were the same on engorgement day.  相似文献

Allozyme and chromosomal polymorphism were analysed simultaneously in three population samples of Drosophila subobscura from Scotland, Germany and Italy. The study revealed significant genic divergence between the various chromosomal gene arrangements for the loci Acph, Lap and Pep, which are located inside inversions. No difference was found with the loci Me and Odh, which are both on the same chromosome but proximal to the structurally polymorphic region. Samples of a given gene arrangement taken from the different localities are genetically uniform. The data indicate that the observed non-random associations between enzyme genes and chromosomal inversions are the result of inhibited recombination in heterokaryotypes and gradual differentiation of the separated gene pools, rather than selection and coadaptation.  相似文献
