Improving reproductive performance is one of the most important factors affecting the profitability of dairy herds. This study investigated the effect of feeding a high starch (HS) diet and body condition score (BCS) at calving on blood metabolites, fertility and ovarian function and milk production in Holstein dairy cows. One hundred seventy-four multiparous cows were fed common close-up and early lactation diets during the first 15 days in milk (DIM). Cows were randomly assigned to 1 of 2 experimental diets from 16 until 50 DIM (n = 87 per group); normal starch (228 g/kg diet DM; NS) or HS (270 g/kg diet DM; HS) diets. Each treatment group was further subdivided based on BCS at calving as normal BCS (BCS ⩽ 3.5; normal BCS (NBCS); n = 45) or high BCS (HBCS) (BCS ⩾ 3.75; HBCS; n = 42). A significant difference was detected for increased milk production (47.24 v. 44.55 kg/day) and decreased milk fat (33.93 v. 36.33 g/kg) in cows fed HS or NS, respectively. Plasma glucose and insulin concentrations were significantly higher in cows fed the HS compared to the NS diet. Diets significantly affected DIM at first artificial insemination (AI, 79.51 ± 3.83 v. 90.40 ± 3.83 days for cows fed HS and NS diets, respectively). High BCS groups had greater milk fat content and elevated plasma nonesterified fatty acids (NEFA), β hydroxybutyrate (BHB) and bilirubin concentrations. In general, feeding higher starch diets to normal BCS cows during the first 50 DIM improved productive and reproductive performance of early-lactating dairy cows.  相似文献

Mycoendophytes have been a promising source of bioactive metabolites for new developments in pharmaceuticals and industrial relevance. During the screening of mycoendophytes diversity from C. bonplandianus, bioactive C. verrucosus CBF 22 was isolated from the leaf. The bioactive isolate was subjected to fermentation and the ethyl acetate fraction was evaluated for antimicrobial activity by disc diffusion and microbroth dilution methods, followed by cytotoxicity using MTT assay. The results revealed significant activity against Gram negative bacteria, Gram positive bacteria, and human pathogenic dermatophytic fungi, C. albicans. The TLC Bioautography-Guided approach employed for the detection of antimicrobial metabolite from bioactive fraction was depicted by a zone of inhibition. Antimicrobial screening of mycoendophytes C. bonplandianus could envision a unique potential niche for bioactive compounds of pharmaceutical importance. These findings suggest C. verrucosus as a potent candidate for producing natural bioactives, to combat multidrug-resistant pathogens. This work is the first report on the incidence of C. verrucosus as mycoendophyte and its bioactive potential of inhabiting C. bonplandianus Baill.  相似文献

Two new macrolide metabolites of the hygrolidin family, catenulisporidins A and B (1 and 2), together with a known compound hygrolidin (3), were isolated from the culture broth of the rare actinobacterium Catenulispora sp. KCB13F192. Their structures were elucidated on the basis of HRESIMS spectrometric and NMR spectroscopic analyses. Catenulisporidins A and B are the first example of natural hygrolidin and bafilomycin derivatives featuring a modified macrolide ring, and catenulisporidin A possesses a tetrahydrofuran ring through an ether linkage between C-7 and C-10. In cell-based fluorescent imaging and immunoblot assays, the three compounds were shown to inhibit autophagic flux in HeLa cells.  相似文献

The composition of the skin microbiota of amphibians is related to the biology of host species and environmental microbial communities. In this system, the environment serves as a microbial source and can modulate the hosted community. When habitats are fragmented and the environment disturbed, changes in the structure of this microbial community are expected. One important potential consequence of fragmentation is a compromised protective function of the microbiota against pathogenic microorganisms. In this study, the skin microbiota of the amphibian Proceratophrys boiei was characterized, evaluated for relationships with environmental variables and environmental sources of microbial communities, and its diversity evaluated for frog populations from fragmented and continuous forests. In addition, the antimicrobial activity of this skin community was studied in frogs from both forest types. Culture methods and 16S rRNA high‐throughput gene sequencing were used to characterize the microbial community and demonstrated that the skin microbiota of P. boiei is more closely related to the soil microbial communities than those inhabiting water bodies or fragment matrix, the unforested area around the forested fragment. The microbial diversity and abundance of Pboiei skin microbiota are different between continuous forests and fragments. This community is correlated with environmental variables, especially with temperature of microhabitat and distance to human dwelling. All individuals of P. boiei harbored bacteria capable of inhibiting the growth of pathogenic bacteria and different strains of the pathogenic fungus Batrachochytrium dendrobatidis, and a total of 27 bacterial genera were detected. The results of this study indicate that the persistence of populations of this species will need balanced and sustained interactions among host, microorganisms, and environment.  相似文献

Soil contamination by toxic trace metal elements, like barium (Ba), may stimulate various undesirable changes in the metabolic activity of plants. The plant responses are fast and with, direct or indirect, generation of reactive oxygen species (ROS). To cope with the stress imposed by the ROS production, plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS, and their by-products, into stable nontoxic molecules. To assess the Ba stress response of two Brassicaceae species (Brassica juncea, a glycophyte, and Cakile maritime, a halophyte), plants were exposure to different Ba concentrations (0, 100, 200, 300 and 500 μM). The plants response was evaluated through their morphology and development, the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary metabolites. Results indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba (even at 500 μM). The biomass production of C. maritima was slightly affected whereas an increase in biomass B. juncea was noticed. The stress imposed by Ba activated the antioxidant defense system in the two species, noticed by the changes in the leaves activity of catalase (CAT), ascorbate peroxidase (APX) and guaicol peroxidase (GPX), and of the secondary metabolites, through the production of total phenols and flavonoids. The enzymatic response was not similar within the two plant species: CAT and APX seem to have a more important role against the oxidative stress in C. maritima while in B. juncea is GPX. Overall, total phenols and flavonoids production was more significant in the plants aerial part than in the roots, of the both species. Although the two Brassicaceae species response was different, in both plants catalytic and non-catalytic transformation of ROS occurs, and both were able to overcome the Ba toxicity and prevent the cell damage.  相似文献

The tomato (Solanum lycopersicum L.) is one of the world’s most important vegetable crops. Still, phytopathogenic bacteria affect the yield and quality of tomato cultivation, like Agrobacterium tumefeciens (At), Clavibacter michiganensis subsp. michiganensis (Cmm), Pseudomonas syringae pv. tomato (Pst), Ralstonia solanacearum (Rs), and Xanthomonas axonopodis (Xa). Synthetic chemical products are used mostly on disease plant control, but overuse generates resistance to bacterial control. This study aimed to evaluate the in vitro antibacterial activity of the ethanolic extract of Moringa oleifera Lam. leaves against At, Cmm, Pst, Rs, and Xa, as well as information about this plant species’ chemical composition. Antibacterial activity against pathogens observed by microplate technique, phytochemical screening, and FTIR analysis revealed different bio-active compounds on ethanolic extracts with antibacterial activity. The growth inhibition rate ranged between 0.08% and 99.94%. The inhibitory concentration, IC50, required to inhibit 50% of At, Cmm, Pst, Rs, and Xa bacterial growth, was 276.67, 350.48, 277.85, 351.49, and 283.22 mg/L, respectively. Inhibition of phytopathogen bacteria’s growth increased as the concentrations of the extract also increased. Moringa oleifera extract can be recommended as a potent bio-bactericide.  相似文献

Lavandula stoechas L. (Lamiaceae) is an attractive shrub native to the Mediterranean regions used for ornamental, melliferous, aromatic and medicinal purposes. Furthermore, this species presents an increasing interest in cosmetics, perfumery and pharmaceutical industries. The variability of qualitative and quantitative metabolic traits among nine wild germplasms representing the distribution area of this species in Tunisia was undertaken. A total of 45 essential oil components were identified in the aerial parts of the studied germplasms. The main essential oil components were camphor (15.32–50.63%), fenchone (6.57–34.70%), 1,8-cineole (0.05–13.45%) and γ-gurjunene (1.10–12.15%). In addition to the well known chemotypes camphor/fenchone and camphor/1.8-cineole, a new chemotype camphor/γ-gurjunene was detected in Tunisian L. stoechas L. Six phenolic acids (quinic acid, gallic acid, p-coumaric acid, 4,5-di-O-caffeoyquinic acid, salviolinic acid and trans cinnamic acid) and five flavonoids (luteolin-7-o-glucoside, naringin, apegenin-7-o-glucoside, quercetin and kampherol) were identified in the ethanolic extracts. Salviolinic acid (46.30–615.18 μg/g) and luteolin-7-o-glucoside (5.98–38.54 μg/g) were the most abundant phenolic compounds. A high significant phytochemical variability (p ˂ 0.01) in the accumulation of volatile and phenolic secondary metabolites among the studied germplasms was recorded. The conducted multivariate (PCA) and clustering (HCA) analyses revealed different classification pattern for essential oil and phenolic compounds. The detected phytochemical polymorphism among the investigated lavender ecotypes didn't show accordance with bioclimatic and geographical areas which suggests genetic background as the main explaining factor. The detected secondary metabolites polymorphism valorises Tunisian L. stoechas L. genetic resources as valuable plant material in further breeding programs. Moreover, an urgent in situ and ex situ conservation measures are required for these wild germplasms threatened by human over-harvesting practices and the occurring dramatic changes in climatic conditions.  相似文献

【目的】挖掘柞蚕Antheraea pernyi微粒子病的潜在生物标志物,为开发该病害检测方法及研究柞蚕被微孢子虫Nosema pernyi侵染后体内代谢产物的差异及功能奠定基础。【方法】利用高效液相色谱和高分辨率质谱进行非靶向代谢组学分析,调查健康和患微粒子病柞蚕雌成虫血淋巴中代谢物差异。【结果】正离子模式下从健康和患微粒子病柞蚕雌成虫血淋巴中共获得8 870个代谢物,注释代谢物5 390个,筛选到差异表达代谢物472个(上调260个,下调212个),其中二级鉴定差异表达代谢物12个(上调8个,下调4个);负离子模式下获得6 716个代谢物,注释代谢物3 848个,筛选到差异表达代谢物301个(上调207个,下调94个),其中二级鉴定差异表达代谢物9个(上调8个,下调1个)。正离子模式下二级鉴定的差异表达代谢物包括缬氨酸(valine)、苯并噻唑(benzothiazole)、3-脱羟基肉碱(3-dehydroxycarnitine)、1 甲基鸟嘌呤(1-methylguanine)、2-乙氧基萘(2-ethoxynaphthalene)、N6-乙酰基-L-赖氨酸(N6-acetyl-L-lysine)、生物素(biotin)、桑色素(morin)、噻吗洛尔(timolol)、酰基肉碱15∶0(acylcarnitine 15∶0)、酰基肉碱18∶4(acylcarnitine 18∶4)和异槲皮苷(isoquercitrin);负离子模式下二级鉴定的差异表达代谢物包括二甲基丙二酸(dimethylmalonic acid)、戊二酸(glutaric acid)、2,5-二羟基苯甲酸(2,5-dihydroxybenzoic acid)、1,3-二乙酰基丙烷(1,3-diacetylpropane)、3-(4-羟基苯基)乳酸(DL-p-hydroxyphenyllactic acid)、泛酸(pantothenate)、荧光素(fluorescein)、飞燕草素-3-O-beta-吡喃葡萄糖苷(delphinidin-3-O-beta-glucopyranoside)和溶血磷酯酰肌醇16∶1 (lysoPI 16∶1)。【结论】健康与患柞蚕微粒子病的柞蚕雌成虫血淋巴内代谢物具有显著差异,通过代谢组学挖掘出21个二级鉴定的差异表达代谢物,这些代谢物可作为潜在生物标志物用于开发柞蚕微粒子病的检测方法。  相似文献
