Ultraviolet inactivation of transforming Bacillus subtilis markers was studied with the aid of an eightfold auxotrophic recipient and its excision-repair-deficient derivative. The results allow the following conclusions. (i) Wild-type B. subtilis cells are able to repair approx. 80% of the UV-induced lesions causing inactivation of transforming activity in UV-sensitive recipients; (ii) Saturating amounts of donor DNA increase the apparent marker sensitivities. This phenomenon is most pronounced in transformation of UV-sensitive recipients; (iii) various markers are inactivated to different degrees, both when assayed on the wild-type as well as on the UV-sensitive strain; (iv) Various markers are repaired to different degrees in the wild-type recipient.  相似文献

Growth of Escherichia coli on melibiose requires the induced synthesis of α-galactoside permease and α-galactosidase. Hydrolysis of the chromogenic substrate p-nitrophenyl-σ-galactoside by whole bacteria is dependent on intact oxidative metabolism. The α-galactosidase from E. coli was isolated for the first time as a soluble enzyme. In cell-free extracts p-nitrophenyl-α-galactoside hydrolisis was observed only at high protein concentrations and the activity decreased exponentially with the square of the dilution. The reason for this behaviour was shown to be that, unlike other known α-galactosidases, the enzyme of E. coli requires NAD. For optimal activity the enzyme also requires Mn2+, a high concentration of 2-mercaptoethanol, and a pH of 8.1. The approximate molecular weight of the active from of α-galactosidase as determined by sedimentation in a sucrose gradient is 200 000. Due to the instability of the enzyme, its purification has not been achieved.  相似文献

Ecological diversification through divergent selection is thought to be a major force during the process of adaptive radiations. However, the large sizes and complexity of most radiations such as those of the cichlids in the African Great Lakes make it impossible to infer the exact evolutionary history of any population divergence event. The genus Alcolapia, a small cichlid lineage endemic to Lakes Magadi and Natron in East Africa, exhibits phenotypes similar to some of those found in cichlids of the radiations of the African Great Lakes. The simplicity within Alcolapia makes it an excellent model system to investigate ecological diversification and speciation. We used an integrated approach including population genomics based on RAD‐seq data, geometric morphometrics and stable isotope analyses to investigate the eco‐morphological diversification of tilapia in Lake Magadi and its satellite lake Little Magadi. Additionally, we reconstructed the demographic history of the species using coalescent simulations based on the joint site frequency spectrum. The population in Little Magadi has a characteristically upturned mouth—possibly an adaptation to feeding on prey from the water surface. Eco‐morphological differences between populations within Lake Magadi are more subtle, but are consistent with known ecological differences between its lagoons such as high concentrations of nitrogen attributable to extensive guano deposits in Rest of Magadi relative to Fish Springs Lagoon. All populations diverged simultaneously only about 1100 generations ago. Differences in levels of gene flow between populations and the effective population sizes have likely resulted in the inferred heterogeneous patterns of genome‐wide differentiation.  相似文献

目的:探讨红细胞分布宽度血小板比率(RDW)和中性粒细胞淋巴细胞比率(NLR)预测急诊经皮冠状动脉介入(PCI)术后冠脉无复流的应用价值。方法:选取2016年1月~2016年12月于我院行急诊PCI手术治疗的患者66例为研究对象,检测其治疗前的RDW和NLR,统计其急诊PCI术后冠脉无复流现象的发生情况,比较冠脉无复流和正常血流患者的RDW和NLR,分析患者RDW和NLR与其冠脉无复流的关系及其对冠脉无复流的预测价值。结果:66例患者急诊PCI术后冠脉无复流发生率为30.30%(20/66)。与正常血流患者比较,冠脉无复流患者RDW和NLR均升高(P0.05)。Spearman相关分析结果显示患者RDW和NLR与其冠脉无复流均呈正相关(r=0.826,0.878,P0.05)。进一步ROC曲线分析结果显示急诊PCI术前RDW和NLR联合预测其术后冠脉无复流的的ROC曲线下面积高于二者单独预测。结论:急诊PCI患者RDW和NLR与其术后冠脉无复流密切相关且两者联合预测患者术后冠脉无复流的价值良好,可能作为急诊PCI术后冠脉无复流评估的参考指标。  相似文献

Aims A lack of explicit information on differential controls on net primary productivity (NPP) across regions and ecosystem types is largely responsible for uncertainties in global trajectories of terrestrial carbon balance with changing environment. The objectives of this study were to determine how NPP of different forest types would respond to inter-annual variability of climate and to examine the responses of NPP to future climate change scenarios across contrasting forest types in northern China.Methods We investigated inter-annual variations of NPP in relation to climate variability across three forest types in northern China, including a boreal forest dominated by Larix gmelinii Rupr., and two temperate forests dominated by Pinus tabulaeformis Carr. and Quercus wutaishanica Mayr., respectively, and studied the responses of NPP in these forests to predicted changes in climate for the periods 2011–40, 2041–70 and 2070–100 under carbon emission scenarios A2 and B2 of Intergovernmental Panel on Climate Change. We simulated the responses of NPP to predicted changes in future climate as well as inter-annual variability of the present climate with the Biome-BGC version 4.2 based on site- and species-specific parameters. The modeled forest NPP data were validated against values in literature for similar types of forests and compared with inter-annual growth variations reflected by tree-ring width index (RWI) at the study sites.Important findings Inter-annual variations in modeled NPP during the period 1960–06 were mostly consistent with the temporal patterns in RWI. There were contrasting responses of modeled NPP among the three forest types to inter-annual variability of the present climate as well as to predicted changes in future climate. The modeled NPP was positively related to annual mean air temperature in the L. gmelinii forest (P < 0.001), but negatively in the P. tabulaeformis forest (P = 0.05) and the Q. wutaishanica forest (P = 0.03), while the relationships of modeled NPP with annual precipitation for the three forest types were all positive. Multiple stepwise regression analyses showed that temperature was a more important constraint of NPP than precipitation in the L. gmelinii forest, whereas precipitation appeared to be a prominent factor limiting the growth in P. tabulaeformis and Q. wutaishanica. Model simulations suggest marked, but differential increases in NPP across the three forest types with predicted changes in future climate.  相似文献

The solubility of maltitol in pure water and industrial syrup was measured in a temperature range from 10 to 90 °C. Maltitol is highly soluble in water, and this yields high viscosity values for the saturated aqueous solutions at different temperatures. In addition, solubility of maltitol in ethanol/water mixtures was followed at 30, 35, 45, and 55 °C. Results show that maltitol solubility is highly dependent on water content in the solvent mixture. Moreover, it increases monotonically with temperature. The logarithm of viscosity changes linearly against the mole fraction of maltitol in the aqueous solutions up to saturation. The saturated solutions showed a Newtonian behavior in a temperature range from 20 to 90 °C. Maltitol is also characterized in supersaturated solutions by a narrow metastable zone, which slightly increases as temperature is raised. The density of aqueous solutions of maltitol was measured as a function of molality up to saturation at 20 °C, and results show that density can be correlated with concentration according to a linear relation. The obtained results were used to explain maltitol crystallization, which exhibits a high nucleation rate and a slow growth leading to small size crystals.  相似文献

Eukaryotic cells rely on a surveillance mechanism, the "Spindle Assembly Checkpoint"SACM in order to ensure accurate chromosome segregation by preventing anaphase initiation until all chromosomes are correctly attached to the mitotic spindle. In different organisms, a mitotic checkpoint complex (MCC) composed of Mad2, Bub3, BubR1/Mad3, and Cdc20 inhibits the anaphase promoting complex (APC/C) to initiate promotion into anaphase. The mechanism of MCC formation and its regulation by the kinetochore are unclear. Here, we constructed dynamical models of MCC formation involving different kinetochore control mechanisms including amplification as well as inhibition effects, and analysed their quantitative properties. In particular, in this system, fast and stable metaphase to anaphase transition can only be triggered when the kinetochore controls the Bub3:BubR1-related reactions; signal amplification and inhibition play a subordinate role. Furthermore, when introducing experimentally determined parameter values into the models analysed here, we found that effective MCC formation is not combined with complete Cdc20 sequestering. Instead, the MCC might bind and completely block the APC/C. The SACM might function by an MCC:APC/C complex rearrangement.  相似文献

We recorded the in vivo emission and time-resolved spectra of the firefly Luciola praeusta Kiesenwetter 1874 (Coleoptera: Lampyridae: Luciolinae). The emission spectrum shows that the full width at half maximum (FWHM) value for this particular species is 55 nm, which is significantly narrower than the in vivo half-widths reported till now. The time-resolved spectrum reveals that a flash of about 100 ms duration is, in fact, composed of a number of microsecond pulses. This suggests that the speed of the enzyme-catalysed chemiluminescence reaction in the firefly for the emission of light is much faster than was previously believed.
