From the Vero Beach strain of the mosquito Aedes (Stegomyia) aegypti (L.) (Diptera: Culicidae), substrains were selected for susceptibility (SS) and refractoriness (RR) to the dog heartworm Dirofilaria immitis (Leidy) (Filarioidea: Onchocercidae). These two lines and their reciprocal F1 hybrids were analysed for genetic variation at 14 enzyme loci, using polyacrylamide gel electrophoresis. Six of the enzyme loci showed variation (sample size 48 alleles/locus/line). Three of these were monomorphic in the refractory line but polymorphic in the susceptible, i.e. aconitase hydratase (Acoh), isocitrate dehydrogenase-1 (Idh-1) and phosphoglucomutase (Pgm). The other three loci, glucose-6-phosphate isomerase (Gpi), hexokinase-1 (Hk-1) and isocitrate dehydrogenase-2 (Idh-2), were polymorphic in both SS and RR lines and their hybrids. At two loci (Hk-1, Pgm) three alleles were detected, whereas the other polymorphic loci had only two alleles. For Hk-1, the most frequent allele was Hk-1(80) (0.563) in refractory and Hk-1(100) in the susceptible (0.521) and F1 hybrids. For Pgm the most frequent alleles were Pgm125 in the susceptible line (0.646) and Pgm100 in the F1 hybrids (0.563 and 0.604) and refractory line (1.000). The mean observed heterozygosity (Ho), the mean Hardy-Weinberg expected heterozygosity (He) and the mean number of alleles per locus in the refractory line were lower, but not significantly so, than in the susceptible line and their reciprocal F1 hybrids; the proportion of polymorphic loci was significantly lower in the refractory than in the susceptible line and their F1 hybrids. Within both lines all polymorphisms were in Hardy-Weinberg equilibrium, whereas significant departures from predicted frequencies were observed in SS x RR hybrids at four polymorphic loci (Acoh, Gpi, Hk-1, Pgm) and at three polymorphic loci (Acoh, Hk-1, Pgm) in RR x SS hybrids. The average Nei's and modified Rogers' genetic distances between the lines were 0.024 and 0.139, respectively. These electrophoretic data show that the refractory line (putatively lacking fi allele) can be distinguished from the susceptible line (fi/fi) and their hybrids (heterozygous fi) by isozyme marker frequencies, but it remains to be seen whether this difference is causal or chance linkage. In any case, this model system of Ae. aegypti/D. immitis provides opportunities to better understand and manipulate the molecular biology of filariasis transmission.  相似文献

Most harmful algal blooms (HAB) originate away from the shore and, for them to endanger human health, they must be first transported to shore after which they must enter the surf zone where they can be feed upon by filter feeders. The last step in this sequence, entrance into the surf zone, depends on surf zone hydrodynamics. During two 30-day periods, we sampled Pseudo-nitzschia and particulate domoic acid (pDA) in and offshore of a more dissipative surf zone at Sand City, California (2010) and sampled Pseudo-nitzschia in and out of reflective surf zones at a beach and rocky shores at Carmel River State Beach, California (2011). At Sand City, we measured domoic acid in sand crabs, Emerita analoga. In the more dissipative surf zone, concentrations of Pseudo-nitzschia and pDA were an order of magnitude higher in samples from a rip current than in samples collected just seaward of the surf zone and were 1000 times more abundant than in samples from the shoals separating rip currents. Domoic acid was present in all the Emerita samples and varied directly with the concentration of pDA and Pseudo-nitzschia in the rip current. In the more reflective surf zones, Pseudo-nitzschia concentrations were 1–2 orders of magnitude lower than in samples from 125 and 20 m from shore. Surf zone hydrodynamics affects the ingress of Pseudo-nitzschia into surf zones and the exposure of intertidal organisms to HABs on the inner shelf.  相似文献

Cholera enterotoxin (CT) increased the concentration of adenosine 3′-5′-cyclic monophosphate (cAMP) in monolayer cultures of adrenal tumor cells after a 60 min lag phase in contrast to the rapid effect of adrenocorticotropin (ACTH). The increasing anthropogenic disturbance on coastal ecosystems has threatened ecological interactions and ecosystems functioning. To investigate if human pressure affects the trophic structure of sandy beaches, mass-balanced models were applied on two Brazilian sandy beaches with distinct human impact degree. The food web models included detritus, phytoplankton, macroinvertebrates, fish and seabirds. Macroinvertebrates in non-urbanized sectors represented the highest production fraction consumed by predators. The energy transfer and the cycling indicator were more efficient in the non-urbanized sectors than urbanized ones. The results indicate that macroinvertebrates sensitive to direct human impact such as trampling are important to the trophic functioning of sandy beaches. Establishing a threshold for the number of beachgoers or dispersing recreational activities to avoid crowds may be tangible ways to mitigate the trampling impact on macroinvertebrates.  相似文献

Habitat use has important consequences for avian reproductive success and survival. In coastal areas with recreational activity, human disturbance may limit use of otherwise suitable habitat. Snowy plovers Charadrius nivosus have a patchy breeding distribution along the coastal areas on the Florida Panhandle, USA. Our goal was to determine the relative effects of seasonal human disturbance and habitat requirements on snowy plover habitat use. We surveyed 303 sites for snowy plovers, human disturbance, and habitat features between January and July 2009 and 2010. We made multiple visits during three different sampling periods that corresponded to snowy plover breeding: pre‐breeding, incubation, and brood‐rearing and used multi‐season occupancy models to examine whether human disturbance, habitat features, or both influenced site occupancy, colonization (probability of transition from an unoccupied site to an occupied site), and extinction (probability of transition from an occupied site to an unoccupied site). Snowy plover site occupancy and colonization was negatively associated with human disturbance and site extinction was positively associated with human disturbance. Interdune vegetation had a negative effect on occupancy and colonization, indicating that plovers were less likely to use areas with uniform, dense vegetation among dunes. Also, dune shape, beach debris, and access to low‐energy foraging areas influenced site occupancy, colonization, and extinction. Plovers used habitat based on beach characteristics that provided stage‐specific resource needs; however, human disturbance was the strongest predictor of site occupancy. In addition, vegetation plantings used to enhance dune rehabilitation may negatively impact plover site occupancy. Management actions that decrease human disturbance, such as symbolic fencing and signage, may increase the amount of breeding habitat available to snowy plovers on the Florida Panhandle and in other areas with high human activity. The specific areas that require this protection may vary across snowy plover life history stages.  相似文献

Enhanced anaerobic biodegradation of groundwater contaminated by fuel hydrocarbons has been evaluated at a field experiment conducted at the Naval Weapons Station, Seal Beach, California. This experiment included the establishment of three different remediation zones in situ: one zone was augmented with sulfate, one was augmented with sulfate and nitrate, and the third was unaugmented. This enables a comparison of hydrocarbon biodegradation under sulfate-reducing, sequential denitrifying/sulfate-reducing, and methanogenic conditions, respectively. In general, the results from the field experiment are: (1) Certain fuel hydrocarbons were removed preferentially over others, but the order of preference is dependent upon the geochemical conditions; and (2) In the zones that were augmented with sulfate and/or nitrate, the added electron acceptors were consumed quickly, indicating that enhancement via electron acceptor injection accelerates the biodegradation process. More specifically, in the sulfate-reducing zone, sulfate was utilized with an apparent first-order rate coefficient of approximately 0.1 day-1. In the combined denitrifying/sulfate-reducing zone, nitrate was utilized preferentially over sulfate, with an apparent first-order rate coefficient of 0.1–0.6 day-1. However, the data suggest that slow sulfate utilization does occur in the presence of nitrate, i.e., the two processes are not strictly sequential. With regard to the aromatic BTEX hydrocarbons, toluene was preferentially removed under intrinsic conditions; biodegradation of benzene was slow if it occurred at all; augmentation with sulfate preferentially stimulated biodegradation of o-xylene; and ethylbenzene appeared recalcitrant under sulfate-reducing conditions but readily degradable under denitrifying conditions.  相似文献

In order to assess the value of common climatic information given to vacationists, measurements of thermophysiologically relevant climatic parameters were carried out on a beach in Italy and compared to those of a nearby official meteorological station. As an integral bioclimatic index the Physiological Equivalent Temperature, PET, was calculated. The results indicate that both the diurnal courses and the absolute values differ essentially at the different locations on the beach and at a nearby met. station. On the beach the thermal stress during the hottest time of the day is significantly lower than at a location a few kilometres inland.  相似文献

Synopsis The introduction of the Nile perch into Lake Victoria has dramatically altered the fishery in that lake and contributed to the decline of the fishery for indigenous tilapias. One sector of the fishery in Lake Victoria has benefitted from the Nile perch introduction, although catches have declined in recent years. Inefficient enforcement of fisheries regulations has had a detrimental effect on indigenous species but may also have contributed to the recent decline in Nile perch catches. Fisheries development plans have tended to favour capital-intensive fisheries and to ignore small scale subsistence fisheries. A case study at Wichlum Beach on the Kenyan shores of Lake Victoria has revealed the efficiency of traditional fishing and fish drying methods as well as the high ecological costs of the practice of kiln-drying Nile perch. Forty-five tons of firewood are used per month at Wichlum Beach alone for kiln-drying perch. The increased economic viability of the fishery has attracted professionals into the industry and resulted in the development of an export-oriented trade. The Yala Swamp adjacent to Lake Victoria has been extensively drained as part of a large land reclamation scheme and more draining is planned. Increased environmental awareness in Kenya, and Kenya's membership in the Convention on Wetlands of International Importance, has resulted in a critical review of these plans.  相似文献

通过野外调查研究了鄱阳湖滩地水生植物物种多样性和群落多样性,并将遥感(RS)技术与地理信息系统(GIS)技术和全球定位系统(GPS)技术相结合,研究了鄱阳湖滩地植被的分布、面积和生物量,绘制了滩地植被生物量等级分布图.结果如下1)鄱阳湖滩地分布有水生植物28科、56属、95种3变种,其中灰化苔草(Carexcineras-cens)、阿及苔草(C.argi)、草(Phalaris arundinacea)、获(Miscanthus sacchariflorus)、芦苇(Phragmites communis)、单性苔草(C.unisexualis)和针蔺(Eleochari valleculosa)7种植物为主要优势种;2)鄱阳湖滩地植被可划分为12个群丛类型,其中灰化苔草群丛(C.cinerascens Ass.)和阿及苔草群丛(C.argiAss.)占优势,"荻+芦-灰化苔草群丛”(M.sacchariflorus+P.communis-C.cinerascens Ass.)次之;3)鄱阳湖滩地植被主要分布于该湖的南部和西部,2000年4月16日其总面积为1018.74km2,占鄱阳湖总面积的33.65%,其中单位面积生物量≥4000g·m-2的植被面积为444.53km2,分别占鄱阳湖总面积和滩地植被总面积的14.68%与43.63%;4)2000年4月16日全湖滩地植被总生物量为3.81×106t(鲜重),全湖滩地植被平均单位面积生物量为3735.93g·m-2(鲜重);5)滩地植被单位面积生物量与Landsat-ETM+4亮度值间的回归方程为y=-4897.11+87.68x(式中y为生物量g·  相似文献
