Barley Yellow Mosaic Virus disease caused by different strains of BaYMV and BaMMV is a major threat to winter barley cultivation in Europe. Pyramiding of resistance genes may be considered as a promising strategy to avoid the selection of new virus strains and to create more durable resistances. However, this goal cannot be achieved by phenotypic selection due to the lack of differentiating virus strains. For pyramiding of resistance genes rym4, rym5, rym9 and rym11, located on chromosomes 3H and 4H of barley two different strategies have been developed. These strategies are based on doubled haploid lines (DHs) and marker assisted selection procedures. On the one hand F1 derived DH-plants of single crosses were screened by molecular markers for genotypes being homozygous recessive for both resistance genes. These genotypes were crossed to lines carrying one resistance gene in common and an additional third gene, leading to a DH-population of which 25% carry three resistance genes, 50% have two resistance genes and 25% possess a single resistance gene homozygous recessively. Alternatively, F1 plants having one resistance gene in common were directly inter-crossed [e.g. (rym4 × rym9) × (rym4 × rym11)] and about 100 seeds were produced per combination. Within these complex cross progenies plants were identified by markers being homozygous at the common resistance locus and heterozygous at the others. From such plants, theoretically present at a frequency of 6.25%, DH-lines were produced, which were screened for the presence of genotypes carrying three or two recessive resistance genes in a homozygous state. Besides DH-plants carrying all possible two-gene combinations, 20 DH-plants out of 107 analysed carrying rym4, rym9, and rym11 and 27 out of 187 tested carrying rym5, rym9, and rym11 homozygously have been detected using the second strategy which is faster but needs co-dominant markers, because in contrast to the first strategy marker selection is carried out on heterozygous genotypes. 相似文献

Passport data for Mexico’s Guanajuato State were used to locate the sites where maize was collected in the 1940s and 1950s
in an effort to document and conserve diversity. A map presenting survey points illustrates that collections have occurred
repeatedly in the same locations. Observations of these locations reveal that urbanization and industrialization, not high
yielding varieties, are displacing traditional varieties. Non-linear principal components analysis was used to assess associations
between variables in areas where maize persists. Landraces appear to be associated with mountains and mesas, mixed cropping,
little or no access to irrigation and areas classified as having low agricultural capacity; conversely, landraces have more
commonly been replaced in areas of high agricultural capacity. The areas of high agriculture capacity, located in the riparian
areas and plains, also have been the easiest to develop for urban and industrial use. Increasingly high rates of urbanization

Summary Electrophoretic assays of 1506 accessions of domestic (Hordeum vulgare L.) and wild (H. spontaneum Koch.) barley, maintained in the USDA World Barley Collection, led to the following conclusions: (1) worldwide the four esterase loci, Est 1, Est 2, Est 3, and Est 4, have a minimum of 7, 12, 6, and 7 alleles, respectively; (2) little or no genetic differentation has developed between H. vulgare and H. spontaneum at these four esterase loci; (3) substantial genetic polymorphism and heterozygosity occur within many of the accessions despite the heavy inbreeding which results from the mating system of predominant self fertilization and from genetic drift associated with maintenance in small populations; (4) patterns of geographical distribution of alleles at these four loci are not at random over both small and large geographical areas, including differences on a continental scale; (5) four among 16 four-locus combinations of alleles are found in excess and all other combinations occur in deficiency on a worldwide basis.This work was supported in part by National Science Foundation Grant DEB 78-02046 相似文献

have been combined to predict the amount of material lost by barley roots during a season, and to predict the resulting microbial
biomass in the rhizosphere. The predicted microbial biomass C in the rhizosphere ranged from 10–34% of the total plant biomass
C depending mainly upon the value used for rate of loss of root material. Total loss of root material predicted during a season
in England constituted 7.7–25.4 percent of C fixed by photosynthesis. The major assumptions made in these calculations are

Barley (Hordeum vulgare L.) is one of the most Aluminum (Al) sensitive cereal species. In this study, the physiological, biochemical, and molecular response of barley seedlings to Al treatment was examined to gain insight into Al response and tolerance mechanisms. The results showed that superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activity were inhibited to different degrees following Al exposure. The MDA content also significantly increased with increasing Al concentrations. SRAP results indicated significant differences between Al treatments and controls in terms of SRAP profile, and the genomic template stability (GTS) decreased with increasing Al concentration and duration. These integrative results help to elucidate the underlying mechanisms that the barley response to Al toxicity. 相似文献

Summary Post-mitotic epidermal cells of barley leaves were found to contain, in addition to cortical microtubules (CMTs), distinct arrays of endoplasmic microtubules (EMTs). These encircle nuclei and continuously merge into the CMT arrays that underly the plasmalemma. Detailed three-dimensional reconstruction of both types of MTs during fungal infection showed that profound and very rapid MT rearrangements occurred especially in the case of incompatible (resistant) barley-powdery mildew genotype combination. The most early MT responses, followed by their subsequent complete disintegration, were recorded around nuclei. These events might be relevant for the induction of such nuclear processes as onset of DNA synthesis and nuclear chromatin condensation. Observed pattern of early infection events, as well as less prominent responses in the case of compatible (susceptible) barley-powdery mildew genotype combination, both findings suggest that rapid reorganization of the MT cytoskeleton could be involved in recognition of the fungus by host cells and in the initiation of resistance responses in barley leaves. We hypothesize that the integrity and dynamics of the MT cytoskeleton, especially of its perinuclear part, might participate in control mechanisms involved in activation of resistance genes.Abbreviations CMTs

The solid waste obtained in malting industries when dehulling barley grains, which was mainly made up of barley husks, spent grains and grain fragments, was subjected to a double hydrothermal processing under selected conditions. The liquor from the second stage (containing xylooligosaccharides, XOS) was refined by membrane and ion exchange processing (with or without a previous endoxylanase treatment to reduce the XOS molecular weight). Three XOS concentrates with different purity and/or molecular weight distribution were fermented in vitro with faecal inocula to assess their prebiotic potential. Succinate, lactate, formiate, acetate, propionate and butyrate were generated in fermentations, confirming the prebiotic potential of the various products assayed. The purity of XOS concentrates did not play a significant role in fermentation, whereas the sample with shorter average degree of polymerization presented a faster fermentation kinetics and led to the highest concentration of lactic acid. 相似文献

Molecular details underlying regulation of starch mobilization in cereal seed endosperm remain unknown despite the paramount role of this process in plant growth. The structure of the complex between the starch debranching enzyme barley limit dextrinase (LD), hydrolyzing α-1,6-glucosidic linkages, and its endogenous inhibitor (LDI) was solved at 2.7 Å. The structure reveals an entirely new and unexpected binding mode of LDI as compared with previously solved complex structures of related cereal type family inhibitors (CTIs) bound to glycoside hydrolases but is structurally analogous to binding of dual specificity CTIs to proteases. Site-directed mutagenesis establishes that a hydrophobic cluster flanked by ionic interactions in the protein-protein interface is vital for the picomolar affinity of LDI to LD as assessed by analysis of binding by using surface plasmon resonance and also supported by LDI inhibition of the enzyme activity. A phylogenetic analysis identified four LDI-like proteins in cereals among the 45 sequences from monocot databases that could be classified as unique CTI sequences. The unprecedented binding mechanism shown here for LDI has likely evolved in cereals from a need for effective inhibition of debranching enzymes having characteristic open active site architecture. The findings give a mechanistic rationale for the potency of LD activity regulation and provide a molecular understanding of the debranching events associated with optimal starch mobilization and utilization during germination.
