The imperfect fungus Metarhizium anisopliae infects and kills larvae of many insect species, including the mosquito Culex pipiens. Mutants of M. anisopliae selected for enhanced production of amylase have been found to have simultaneously acquired hypervirulence against C. pipiens larvae. In the present work, wild-type and some mutant strains of M. anisopliae were found to excrete an acid or acids which alter the pH of fungal cultures below that permissible for amylase activity. The amylase-enhanced hypervirulence mutants did not excrete acid. Detection of protease and lipase activities was complicated by the acid excretion. When this was taken into account, mutants having altered lipase or protease production were found to have no alteration in virulence against mosquito larvae. The link between acid excretion, amylase activity, and hypervirulence is discussed.  相似文献

Summary UV irradiation of competent cells of Escherichia coli K12 produced an increase in the efficiency of transformation with plasmid DNA. This phenomenon has been called IPTE (increase in plasmid transformation efficiency) and is dependent on the activated state of the RecA protein. IPTE is independent of the lexA, recB recC, and recF genes. It is not related to the size or the replicon type of the plasmid. Furthermore, it is also induced in cells which have been previously treated with other SOS system-inducing agents such as bleomycin, mitomycin C, or nalidixic acid. IPTE is therefore similar to other repair (SOS) functions inducible by DNA damage since all of them are dependent upon activation of the RecA protein. IPTE differs from other SOS functions in the absence of a direct control by the LexA repressor.  相似文献

Abstract The rate of fimbrial phase variation in Escherichia coli strain 3040 was determined. The strain has type-1 and S fimbriae. The bacterial culture was fractionated into homogeneous subpopulations expressing either one of the fimbrial types only; the subpopulations were inoculated into broth and the fimbriation of individual cells was assayed by immunofluorescence as a function of time. The rate of the shift from S- or type-1-fimbriate cells to non-fimbriate ones was of the order of 10−2 per cell generation and more rapid than a direct shift from one fimbrial phase to another, although both types of phase variations were observed.  相似文献

The adhesion of cells of Salmonella typhimurium to albite, biotite, felspar, magnetite and quartz was correlated to the presence of fimbriae and degree of hydrophobicity and charge of the bacterial surface. It was found that the presence of fimbriae resulted in a higher degree of adhesion compared to adhesion of nonfimbriated cells. The significance of the physico-chemical characteristics of fimbriae was shown by a direct linearity between high hydrophobicity of fimbriated cells and degree of adhesion to the mineral particles. Fimbriated cells exhibited higher negative as well as positive surface charge as compared to nonfimbriated cells. Adhesion to several of the minerals was shown to be independent of the extent of negative charges on the bacterial surfaces. A high degree of adhesion to biotite, possibly due to a combination of characteristics of the particles, was not related to either bacterial fimbriation or a physico-chemical characteristic of the bacterial surface. The results of the nonspecific adhesion observed are discussed in terms of available binding sites and distribution of physico-chemical characteristics on the bacterial cell surface structures.  相似文献

We conducted a comprehensive analysis of virulence in the fungal wheat pathogen Zymoseptoria tritici using quantitative trait locus (QTL) mapping. High‐throughput phenotyping based on automated image analysis allowed the measurement of pathogen virulence on a scale and with a precision that was not previously possible. Across two mapping populations encompassing more than 520 progeny, 540 710 pycnidia were counted and their sizes and grey values were measured. A significant correlation was found between pycnidia size and both spore size and number. Precise measurements of percentage leaf area covered by lesions provided a quantitative measure of host damage. Combining these large and accurate phenotypic datasets with a dense panel of restriction site‐associated DNA sequencing (RADseq) genetic markers enabled us to genetically dissect pathogen virulence into components related to host damage and those related to pathogen reproduction. We showed that different components of virulence can be under separate genetic control. Large‐ and small‐effect QTLs were identified for all traits, with some QTLs specific to mapping populations, cultivars and traits and other QTLs shared among traits within the same mapping population. We associated the presence of four accessory chromosomes with small, but significant, increases in several virulence traits, providing the first evidence for a meaningful function associated with accessory chromosomes in this organism. A large‐effect QTL involved in host specialization was identified on chromosome 7, leading to the identification of candidate genes having a large effect on virulence.  相似文献
