The gene encoding an alkaline active cyclodextrin glycosyltransferase (CGTase) from the alkaliphilic B. agaradhaerens LS-3C was cloned and sequenced. It encodes a mature polypeptide of 679 amino acids with a molecular mass of 76488 Da. The deduced amino acid sequence of the mature CGTase revealed 99 and 95% identity to the CGTase sequences from the other B. agaradhaerens strains, DSM 8721T and 9948, respectively. The next closest identity was of 59% with B. clarkii enzyme. CGTases from B. agaradhaerens, B. clarkii, and B. firmus/lentus formed a phylogenetically separated cluster from the other CGTases of Bacillus spp. origin. A number of usually conserved residues in the CGTases were found to be replaced in the sequence of B. agaradhaerens enzyme. The sequence analysis indicated the enzyme to be close to the so-called `intermediary enzymes' in the -amylase family.  相似文献

One of the emerging biopolymers that are currently under active investigation is bacterial poly(γ‐glutamic acid) (γ‐PGA). However, before its full industrial exploitation, a substantial increase in microbial productivity is required. γ‐PGA obtained from the Bacillus subtilis laboratory strain 168 offers the advantage of a producer characterized by a well defined genetic framework and simple manipulation techniques. In this strain, the knockout of genes for the major γ‐PGA degrading enzymes, pgdS and ggt, leads to a considerable improvement in polymer yield, which attains levels analogous to the top wild γ‐PGA producer strains. This study highlights the convenience of using the laboratory strain of B. subtilis over wild isolates in designing strain improvement strategies aimed at increasing γ‐PGA productivity. Biotechnol. The effect of repeated conditioning procedures (25 runs), consisting of soiling (milk and meat products) and cleaning steps, on the hygienic status, physico-chemical properties and surface chemical composition of stainless steel (SS) surfaces, was investigated. Five SSs differing in grade and finish were used. Both soiling and surface cleaning/conditioning procedures resulted in a similar increase in the surface contamination with carbon, while the changes in the basic component of the surface free energy depended on the conditioning procedure. The passive film was also affected, the Fe/Cr ratio in particular. The hygienic status was also changed, especially with milk as shown by monitoring the number of residual adhering Bacillus cereus spores after contaminating the surface with spores followed by cleaning. The results show that in food environments, the presence and the nature of conditioning molecules play a major role in the hygienic status of SS surfaces.  相似文献

A novel Pb-resistant bacterium was isolated from aged lead-contaminated alkaline soils, and was identified as Bacillus megaterium via the MIDI protocol. The biosorption isotherms and kinetics of Pb(II) associated with B. megaterium in vivo in the alkaline environment were investigated at the first time. All the batch experiments of biosorption demonstrate that the B. megaterium uptake of lead is pH-dependent, exothermic (ΔH° = ?5224.86 KJ mol?1), spontaneous, and fits well with the Langmuir isotherm, resulting in different kinetics under different examination temperatures. The maximum biosorption capacity is 503.86 mg g?1 at optimum conditions, which is much better in comparison to the biosorbent reported at the acidic condition in the literature. The Fourier-transform Infrared spectroscopic analysis of lead-loaded biomass confirms that the biosorption between B. megaterium and lead is the chemical adsorption in vivo. A site test indicates that B. megaterium really increases mobility and bioavailability of lead in Pb-contaminated alkaline soil in terms of chemical fractionation in vivo, which will potentially increase its uptake by hyperaccumulated plants in alkaline soils in arid or semi-arid areas of NW, China. Therefore, the novel isolate of B. megaterium with the highest adsorption capacity is a new promising biosorbent for the lead removal in alkaline water and soil.  相似文献

目的 :观察枯草芽胞杆菌喷雾剂对家兔皮肤创伤愈合的影响。方法 :采用家兔皮肤创伤为模型 ,连续给药 6 d,第 11天测定创面面积 ,并于第 17天记录创面愈合数。结果 :枯草芽胞杆菌喷雾剂高剂量组和磺胺嘧啶银组创面面积显著小于空白对照组 ( P<0 .0 5 ) ;枯草杆菌喷雾剂的愈合率显著高于空白对照组( P<0 .0 5 )。结论 :枯草芽胞杆菌喷雾剂能够缩小家兔创面愈合 ,增加创面愈合率  相似文献

目的:生物法脱羧制备4-乙烯基衍生物具有诸多优势和良好的发展前景,研究解淀粉芽孢杆菌Q-426酚酸脱羧酶(BaPAD-Q-426)的酶学性质,为其进一步应用提供理论基础。方法:从解淀粉芽孢杆菌中克隆酚酸脱羧酶基因;以pET-28a(+)为载体,将重组质粒转化至E. coli BL21(DE3)中,实现酚酸脱羧酶BaPAD-Q-426的高效表达,利用Ni-NTA亲和层析进行纯化,并进行酶学性质鉴定。结果:酚酸脱羧酶BaPAD-Q-426在pH 7.0~9.0范围内保持良好的pH稳定性,最适pH为8.0;在25~40℃范围内保持着较高的酶活性,最适温度为35℃,在4℃时保持30 min后该酶依然保持95%以上的酶活性;K+对BaPAD-Q-426的酶活具有明显促进作用,酶活力提高60%;该酶在石油醚中具有良好的耐受能力,在40%石油醚存在下,仍保留50%以上的酶活力;BaPAD-Q-426的最适底物为阿魏酸,酶活力达到19.5 IU/mL。结论:与其他来源的酚酸脱羧酶相比,BaPAD-Q-426在低温时具有更好的稳定性,在弱碱性环境下对阿魏酸的催化脱羧能力最强。  相似文献

The bacteria Bacillus thuringiensis mutant is highly producing melanin pigment with increased ultra violet resistance and insecticidal activity against the potato tuber moth Phthorimaea operculella (Zeller). The results showed that the high decrease of crystal protein formation rate ranged from 100% (B.t.EMS-M2 and B.t.EMS-M6) to 91.82% (B.t.EMS-M9). The EMS–UV-induced mutants (B.t.EMS–UV-2h-1, B.t. EMS–UV-2h-2, B.t.EMS–UV-2h-3, B.t.EMS–UV-2h-5, B.t.EMS–UV-4h-1, B.t.EMS–UV-4h-3 and B.t.EMS–UV-6h-2) showed 100% decrease in the crystal protein formation. Results also showed that the growth rate of B. thuringiensis isolates was detected by measuring the light absorption of culture broth (BP media at pH 8) at the wavelength of 600 nm. The absorbance values of the standard melanin were 2.055 and 0.134 at wavelengths of 226.5 and 602 nm, respectively. This means that the maximum absorbance at wavelength was 226.5 nm, this result is similar to that of the synthetic melanin which has the absorbance of 226 nm. Our experiments detected that the pigment extracted from the mutant isolate B.t.EMS-M3 (EMS-induced mutant) gave the maximum value of absorbance (2.615) at wavelength of 227.5 nm that was similar to standard melanin which gave absorbance value about 2.055 at a wavelength of 226.5 nm. This may be due to the genetic alterations that happened to the mutant isolates due to the mutation by EMS or/and UV irradiation.  相似文献

The lack of data on the effect of Bacillus thuringiensis (B.t.) toxins on larval feeding behavior of the pest Spodoptera exigua (Hübner) (Noctuidae: Amphypyrini) prompted us to investigate the effect of three delivery systems of CryIC, a commercial formulation, inclusion bodies, and the activated CryIC toxin. The commercial formulation was the least and CryIC toxin the most lethal form to neonates of susceptible colonies. All but two of the treatments in choice tests with neonates and third instars showed significant avoidance of B.t. treated diet, with greater proportion of larvae from susceptible (UCR-S and AUBURN-S) and resistant (AUBURN-R) colonies on untreated diet than on diet treated with any of the CryIC forms and concentrations tested. Furthermore, third instars consumed significantly more control than treated diet for all CryIC forms, colonies and concentrations. The avoidance of CryIC toxin by neonates and third instars strongly suggests that CryIC, which also is present in the commercial formulation and in the inclusion bodies, is responsible for eliciting avoidance behavior by S. exigua larvae. Behavioral observations of third instars in a no-choice test on either treated or control diet indicated that questing behavior in susceptible larvae appears to be positively related with presence of CryIC toxin in the diet. Furthermore, resistant third instars were on the whole more active than susceptible thirds on both treated and control diet. Resistant thirds raised on CryIC treated diet (AUBURN-RC) spent more time eating treated diet than resistant larvae raised on control diet (AUBURN-R), suggesting that diet conditioning plays an important role on feeding behavior of S. exigua. The implications of these results are discussed.  相似文献

Two rapid and easy-to-scale-up methods for the purification of cyclodextrin glycosyltransferase (CGTase) from Bacillus circulans were developed: affinity precipitation with starch and aqueous two-phase partition. The first method, optimised by a factorial design, gave an 80% CGTase adsorption at 11% starch and 1.6% ammonium sulphate, and a 65% recovery after elution with 10 mM α-cyclodextrin. The purification factor was 17. Aqueous two-phase partition yielded a 72% CGTase recovery in a two-step procedure; CGTase was obtained in the bottom phase with a purification factor of 37.  相似文献

Bacillus subtilis fmbj can produce lipopeptide antimicrobial substance, whose main components were surfactin and fengycin. In the study, the antiviral activity of antimicrobial lipopeptides (AMLs) from B. subtilis fmbj (CGMCC No. 0934) against Pseudorabies Virus (PRV), Porcine Parvovirus (PPV), Newcastle Disease Virus (NDV) and Infectious Bursal Disease Virus (IBDV) was evaluated in vitro. The AMLs represented a direct inactivation effect on cell-free virus stocks of PRV, PPV, NDV and IBDV, and it could effectively inhibit infection and replication of the NDV and IBDV, but failed to affect PRV and PPV. The AMLs were represented higher toxicity for the Porcine Kidney (PK-15) cells (50% cytotoxic concentration (CC50) value was 32.87 μM) and lower for the Chicken Embryo Fibroblasts (CEF) cells (CC50 value was 89.16 μM). The Selectivity index of AMLs on PRV, PPV, NDV and IBDV was 1.44, 2.23, 8.40 and 12.19, respectively.  相似文献
