The adaptor protein Shb has previously been shown to regulate apoptosis in response to cytokines and inhibitors of angiogenesis although the mechanisms governing these effects have remained obscure. We currently demonstrate interactions between Shb and c-Abl and that Shb regulates c-Abl kinase activity. The data suggest that c-Abl binds to tyrosine phosphorylated Shb via a concerted effort involving both the c-Abl SH3 and SH2 domains. The biological significance of the Shb/c-Abl interaction was presently tested in overexpression experiments and was found to promote hydrogen peroxide-induced cell death. We also show by Shb knockdown experiments that Shb regulates c-Abl activity and modulates cell death in response to the genotoxic agent cisplatin and the endoplasmic reticulum stress-inducer tunicamycin. The findings are in agreement with the notion of Shb playing a pivotal role in modulating c-Abl pro-apoptotic signaling in response to various stress stimuli.  相似文献

B细胞是体液免疫的重要执行细胞,其活化是机体产生保护性抗体的关键步骤.目前人们对B细胞早期活化的动态分子事件和信号起始机制等仍然未知.本文将重点总结超高清成像技术和高速高分辨率活体成像技术在B细胞领域的应用,这些研究将帮助人们理解B细胞早期活化的机制.本文系列总结了静息态下维持B细胞存活的B细胞受体(B cell receptor,BCR)滋养信号的研究进展,并提出了滋养信号来源的几种可能的模型.描述了抗原刺激导致的BCR活化的信号通路,并重点探讨了成像技术进步带来的关于BCR信号通路起始的机制探索这一免疫学领域的重大问题.结合高速高分辨率活细胞成像技术在免疫学领域的应用,抗原刺激后BCR活化过程中一系列动态变化过程和高级结构的形成能够被实时捕获.此外,还探讨了B细胞记忆性免疫发生的机制,重点阐述了亲和力成熟和BCR亚型转换,尤其是IgG(Immunoglobulin G)型BCR胞内尾巴对快速强烈的记忆性免疫反应的帮助.B细胞活化机制的调节过程发生异常会破坏正常的B细胞稳态平衡和免疫疾病的发生,本文总结抑制性调节受体FcγRIIB(Fcγreceptor IIB)突变与自身免疫病的关系,以及BCR信号通路信号分子突变与B细胞肿瘤的关系,这些研究将加深人们对B细胞免疫疾病的认识和相应医疗手段的改进.  相似文献

The present research was conducted to determine heavy metals in agricultural soils from Çanakkale, Turkey, using a sequential extraction procedure (acid soluble, reducible, oxidizable, and residual) as proposed by the Community Bureau of Reference (BCR) of the European Commission. Soil samples were taken from 12 different cultivated sites and analyzed for Cd, Co, Cr, Cu, Mn, Ni, Pb, and Zn concentrations. The results revealed an order of Mn > Cd > Pb > Co > Ni > Cu > Zn > Cr for the heavy metals based on the sum of the first three fractions (acid soluble + reducible + oxidizable). The relationships between soil properties and each metal fraction were identified through Pearsons's correlation analysis. Hierarchical cluster analysis was performed to determine the behaviors and similarities of metals in each fraction. While Mn, Pb, and Zn exhibited subjective behaviors in the acid-soluble fraction, Cd, Co, Cu, Cr, and Ni exhibited similar behaviors with each other.  相似文献

Protein adapters couple surface receptors to multiple intracellular signaling modules by acting as scaffolds for the assembly of multimolecular complexes responsible for the coordination and amplification of signals. Through the spatiotemporally controlled recruitment of mediators with opposite activities (e.g. protein tyrosine kinases and phosphatases), adapters are implicated not only in signal initiation and propagation, but also in feedback loops for signal extinction. Moreover, adaptors specialized in preventing or dampening signaling have been more recently discovered. Here we shall present of brief overview of the principal adaptors which act as negative regulators of TCR and BCR signaling, with a focus of the mechanisms underlying this function. We shall then discuss our recent findings implicating p66Shc and Rai, two members of the Shc family of cytosolic protein adapters, in the negative control of antigen receptor signaling, and their role as gatekeepers of autoimmunity and leukemia.  相似文献

Leukaemic transformation is frequently associated with the aberrant activity of a protein tyrosine kinase (PTK). As such it is of clinical relevance to be able to map the effects of these leukaemogenic PTKs on haemopoietic cells at the level of phosphorylation modulation. In this paradigm study we have employed a range of proteomic approaches to analyse the effects of one such PTK, BCR/ABL. We have employed phosphoproteome enrichment techniques allied to peptide and protein quantification to identify proteins and pathways involved in cellular transformation. Amongst the proteins shown to be regulated at the post‐translational level were cofilin, an actin‐severing protein thus linked to altered motility and Cbl an E3 ubiquitin ligase integrally linked to the control of tyrosine kinase signalling (regulated by 5 and 6 PTKs respectively). The major class of proteins identified however were molecular chaperones. We also showed that HSP90 phosphorylation is altered by BCR/ABL action and that HSP90 plays a crucial role in oncogene stability. Further investigation with another six leukaemogenic PTKs demonstrates that this HSP90 role in oncogene stability appears to be a common phenomenon in a range of leukaemias. This opens up the potential opportunity to treat different leukaemias with HSP90 inhibitors.  相似文献

Complement receptor type 2 (CR2, CD21) is a cell surface protein that links the innate and adaptive immune response during the activation of B cells. The extracellular portion of CR2 comprises 15 or 16 short complement regulator (SCR) domains, for which the overall arrangement in solution is unknown. This was determined by constrained scattering and ultracentrifugation modelling. The radius of gyration of CR2 SCR 1-15 was determined to be 11.5 nm by both X-ray and neutron scattering, and that of its cross-section was 1.8 nm. The distance distribution function P(r) showed that the overall length of CR2 SCR 1-15 was 38 nm. Sedimentation equilibrium curve fits gave a mean molecular weight of 135,000 (+/- 13,000) Da, in agreement with a fully glycosylated structure. Velocity experiments using the g*(s) derivative method gave a sedimentation coefficient of 4.2 (+/- 0.1) S. In order to construct a model of CR2 SCR 1-15 for constrained fitting, homology models for the 15 SCR domains were combined with randomised linker peptides generated by molecular dynamics simulations. Using an automated procedure, the analysis of 15,000 possible CR2 SCR 1-15 models showed that only those models in which the 15 SCR domains were flexible but partially folded back accounted for the scattering and sedimentation data. The best-fit CR2 models provided a visual explanation for the versatile interaction of CR2 with four ligands C3d, CD23, gp350 and IFN-alpha. The flexible location of CR2 SCR 1-2 is likely to facilitate interactions of C3d-antigen complexes with the B cell receptor.  相似文献
