The stereospecificity of chloramphenicol isomers on the inhibition of several plant systems was investigated. l-Threo, d-erythro, l-erythro and the antibiotic d-threo-chloramphenicol were effective inhibitors of auxin-induced elongation, 14C-leucine uptake and 14C-leucine incorporation into the protein fraction of coleoptiles from Avena sativa and Triticum vulgare. The isomers also inhibited Avena coleoptile uptake of 14C-α-aminoisobutyric acid and the de novo synthesis of α-amylase by aleurone layers from Hordeum vulgare seeds. All four compounds inhibited these processes to about the same extent and over a similar high concentration range (5 × 10−4 to 5 × 10−3 M). Bioassay of extracts from Avena coleoptiles treated with the non-antibiotic isomers revealed no tissue conversion into d-threo-chloramphenicol.  相似文献

A microsomal fraction was prepared by differential centrifugation from the homogenate of dark grown shoots of oats ( Avena sativa L. cv. Sol II). Plasma membranes were prepared from the microsomal fraction by means of an aqueous polymer two phase partition method. The content of phytochrome in the microsomal fraction and the plasma membrane fraction, respectively, were studied after different irradiation treatments of the intact shoots. Red irradiation increased the content of phytochrome in both the microsomal and plasma membrane fraction, especially in the presence of Mg2+. The increase induced by red light was fully reversible by far-red light for the plasma membrane fraction both in the presence and absence of Mg2+, in contrast to the microsomal fraction where Mg2+ had to be omitted. KI treatment of the membranes destroyed the binding of phytochrome whereas agents such as KCI, EDTA, CaCl2 and Triton X-100, did not have this effect, indicating that the phytochrome attachment to the membrane is hydrophobic. This in vivo binding resembles to a large extent the one obtained in vitro by Sundqvist and Widell (Physiol. Plant. 59: 35–41, 1983) even though some differences between the phytochrome species and the membrane side exposed probably occur; so that the present interaction between phytochrome and the plasma membrane does not necessarily reflect the interaction that leads to physiological responses, and there could be more than one type of interaction.  相似文献

The effect of silicon on organ growth and its mechanisms of action were studied in rice (Oryza sativa L. cv. Koshihikari), oat (Avena sativa L. cv. Victory), and wheat (Triticum aestivum L. cv. Daichino-Minori) seedlings grown in the dark. Applying silicon in the form of silicic acid to these seedlings via culture solution resulted in growth promotion of third (rice) or second (oat and wheat) leaves. The optimal concentration of silicon was 5–10 mM. No growth promotion was observed in early organs, such as coleoptiles or first leaves. In silicon-treated rice third leaves, the epidermal cell length increased, especially in the basal regions, without any effect on the number of cells, showing that silicon promoted cell elongation but not cell division. Silicon also increased the cell wall extensibility significantly in the basal regions of rice third leaves. These results indicate that silicon stimulates growth of rice and some other Poaceae leaves by increasing cell wall extensibility. Received: July 31, 2001 / Accepted: September 18, 2001  相似文献

Binding of 3H-labeled cyclic GMP (3H-cGMP) to the structural components of subcellular fractions was studied in oat seedlings. The binding was found to depend on the conditions of incubation and illumination of growing plants. In the green seedlings, the binding activity was lower than in the etiolated seedlings. The highest binding was observed in soluble cytosolic fraction where two types of specific sites for cGMP binding (with high and low affinity to the cyclic mononucleotide) were detected. The binding activity was found to increase as a result of red light influence via phytochrome, as well as in the presence of calcium ions and calcium–calmodulin complex.  相似文献

Studies of root morphology or root architecture are limited by the problems of observing roots in soil. NMR imaging can preferentially detect the water in roots rather than soil and can therefore produce a map of the distribution of the water in roots, even though there is much more water in the soil than in the roots. The gradient echo imaging technique was shown to detect less signal from water in soil than the more usual spin‐echo technique, probably because the signal loss due to magnetic field inhomogeneities was not refocused in the gradient echo experiments. The gradient echo sequence produced images of a water‐filled capillary tube in soil and of the main roots of a 31‐day‐old oat plant, images which had better contrast than those from the spin‐echo sequence. The improvement was particularly noticeable in the wetter soils. The finer roots could only be observed in the driest of soil and with the spin‐echo sequence. Magnetic field inhomogeneities have a greater effect on thinner roots than on thicker ones, and because the initial signal from thin roots was so small, the additional signal loss due to magnetic field inhomogeneities made the fine roots undetectable with the gradient echo sequence. The problem of loss of perspective associated with projection images was overcome by taking a number of images with different projection planes. By comparing images, a set of (x, y, z) coordinates was obtained for the 4 main roots of a 31‐day‐old oat plant. Computer graphics methods made it possible to compare the digitised root coordinate data set with the original images, thereby confirming the accuracy of the digitised data.  相似文献

Germination and carbohydrate concentrations were determined in excised dormant and afterripened wild oat (Avena fatua L. line M73) embryos cultured on N6 medium with and without 88 mM fructose (Fru). Without Fru dormant embryos began to germinate after approximately 2 weeks, and the germination rate was greater at 12 than 16°C. With addition of Fru 80% of dormant embryos germinated in 3 days. More than 80% of afterripened embryos germinated within 1 day on N6 with or without additional sugars. Therefore, relative to afterripened embryos, true embryo dormancy exists in line M73. Concentrations of starch and soluble sugars were initially similar in dormant and afterripened embryos. Culturing dormant and afterripened embryos on medium with Fru resulted in concentrations of glucose (Glu), sucrose (Sue), Fru and maltose (Mal) that were the same or higher than the initial levels. The concentration of starch in embryos initially increased slightly then remained constant or declined, except in dormant embryos on Fru-amended medium, where starch accumulated to 34 μg Glu equivalents (mg fresh weight)-1 at 52 h. Raffinose (Raf) and stachyose (Stach) concentrations declined over time in all embryos. Carbohydrate concentrations in afterripened embryos on medium without Fru decreased to nearly undetectable levels by 52 h. Soluble sugar concentrations in dormant embryos on medium without Fru also declined by 52 h, but changes were not as extensive as those in afterripened embryos without Fru. In 52 h Raf and Stach were nearly depleted in all afterripened embryos, and in dormant embryos cultured on Fru-containing medium but not in dormant embryos without Fru. The concentration of Stach in dormant embryos without Fru declined 60% at 12 to 18 days coinciding with the potential for germination. The results demonstrate that a decline in Stach concentration is associated with the potential for germination of dormant (D) excised embryos. The mechanism of dormancy-breaking associated with the Raf family oligosaccharides remains to be determined.  相似文献

Phospholipase D (PLD) activity was found to be higher in etiolated oat seedlings than in green seedlings. White and red (R) light exposure inhibited PLD activity in etiolated seedlings. Far-red light eliminated R-light-induced decrease in PLD activity, indicating phytochrome participation in observed photomodulation. Inhibitor of electron transport in chloroplast 3-(3,4-dichlorophenyl)-1,1-dimethylurea stimulated and glucose suppressed PLD activity in green and etiolated oat seedlings, respectively. These results suggest that PLD activity in oat seedling is regulated by light with involvement of phytochrome photoreceptor, and associated with photosynthesis process.  相似文献

The chromosomal loci of 5S and 45S ribosomal DNAs (rDNAs) and the activity of nucleolar‐organizing regions (NORs) were analysed in perennial oats of the genera Ammophila, Amphibromus, Arrhenatherum, Avena, Deschampsia, and Helictotrichon s.l. (Poaceae: Aveneae) using fluorescence in situ hybridization, staining with chromomycin/4′,6‐diamidino‐2‐phenylindole (DAPI), and silver impregnation. All chromosomes with a secondary constriction were nucleolar active. In chromosomes without a secondary constriction, NORs corresponded exclusively to broad bands of 45S rDNA with chromomycin‐positive, DAPI‐negative, and silver‐positive stainability. Additional minor bands of 45S rDNA showed no nucleolar activity. 5S rDNA was localized mostly in loci different from the nucleolar‐active 45S rDNA. If both rDNAs occurred within the same chromosome, they were at largely corresponding distances from the centromere, irrespective of their particular localization in either the same chromosome arm or in opposite arms. In the latter case, 5S rDNA was never more distal to the centromere than 45S rDNA. A new model was devised to explain this non‐random distribution of both rDNAs in nucleolar‐organizing chromosomes, which identified the Rabl orientation of chromosomes as ensuring a spatial proximity of 5S to 45S rDNA in interphase nuclei, even if they were localized in opposite arms. The possible role of the Rabl orientation in determining the spread and accumulation of 5S rDNA sequences in further chromosomes of the genome was discussed. B chromosomes were devoid of 5S rDNA, but most contained 45S rDNA and were nucleolar active. In some large groups of species, the number and arrangement of 5S and 45S rDNA sites in the chromosomes were remarkably uniform, especially in Helictotrichon subgenus Helictotrichon and Helictotrichon subgenus Pratavenastrum.
