Suspension cultured oat (Avena sativa L. cv. Garry) cells, which secrete polysaccharides into the medium, were used as starting material for analyses of Golgi-derived vesicle membranes and plasma membranes isolated during cell fractionation. Vesicles collected by a procedure employing ultrafiltration followed by ultracentrifugation into a sucrose step gradient exhibited an equilibrium density of 1.27 g cm?3 when run on continuous sucrose gradients, a feature which is most likely attributable to the high concentration of enclosed polysaccharides. Brief sonication lowered the density of these vesicles to about 1.15 g cm?3, as judged from the coincidence of the protein peak and the marker enzymes for Golgi [Triton-stimulated UDPase, cold-storage IDPase (EC 3.6.1.6)] and plasma membrane [vanadate-inhibited K+, Mg2+-ATPase (EC 3.6.1.3)]. Sonication of these vesicles also greatly diminished the amount of detectable polysaccharide observed in a colorimetric assay for sugars. Fractionation of a plasma membrane-enriched preparation from these cells on continuous sucrose gradients showed the major protein peak and the peak activity for the plasma membrane marker at 1.17 g cm?3, however, there was also significant overlap with a mitochondrial [cytochrome c oxidase (EC 1.9.3.1)] peak at 1.18 g cm?3, Smaller peaks of the Golgi markers were seen at 1.14 g cm?3. Analyses of marker enzymes for ER and mitochondria (EC 1.6.99.3) showed little contamination of the membranes of presumptive secretory vesicles from these sources, and the lack of significant vanadate-insensitive ATPase activity in the density range from 1.13–1.18 g cm?3 in either fractionation scheme suggests that these membranes do not include material from the tonoplast. Phototransformation Pt to Pfr was investigated with 124-kDa phytochrome from etiolated oat seedlings ( Avena sativa L. cv. Pirol) using circular dichroism spectroscopy at -110°C to +30°C. Using absorption spectra of the intermediates formed at the respective temperatures, circular dichroism spectra (300–800 nm) of pure intermediates were calculated.
The sign of the circular dichroic absorption bands changed upon formation of lumi-R, the primary photoproduct of Pr. This would be compatible with a Z→E isomerization taking place at this reaction step. The subsequent intermediates (meta-Ra and meta-Rc) as well as Pfr showed only small circular dichroism. Their absorption spectra were drastically shifted, but had similar spectral shapes. The results are discussed in terms of conformational changes of the phytochrome chromophore presumably taking place at the early steps of phototransformation Pr to Pfr.  相似文献

Summary The Giemsa C-banding technique was used to identify individual meiotic and somatic chromosomes in 21 monosomic lines of Avena byzantina C. Koch cv Kanota (genome designation AACCDD). The hexaploid complement is composed of three sets of seven chromosome pairs. The heterochromatin in the putative diploid progenitors is located at the telomeres (genome A), at the centromeric and interstitial regions (genome C), or more evenly spread throughout the set (genome D). Comparisons based on C-banding between A. byzantina and its diploid progenitor species allowed us to allocate individual chromosomes into specific genomes. The C-banding technique may be useful for interspecific chromosome pairing analyses. Nucleolar activity and competition were studied using a silver-staining procedure. Only three chromosome pairs showed nucleolar organizer regions, thus indicating that nucleolar competition occurs naturally in hexaploid oats.  相似文献

The sensitivity to gibberellic acid (GA3) of aleurone protoplasts isolated from a single harvest of an inbred line of Avena fatua seed that had been after-ripened over anhydrous CaCl2 at 25±2°C and 4±2°C for three years was assessed. Protoplasts isolated from aleurones of seed stored at 25°C produced substantially more -amylase in response to 10–7 M GA3 than those isolated from aleurones of seed stored at 4°C. The apparent difference in responsiveness does not appear to be due to a change in the duration of the lag phase between addition of GA3 and the production of -amylase. The dose response of aleurone protoplasts to GA3, measured as -amylase production, is complex and appears to have three phases. Protoplasts from seed stored at both temperatures respond appreciably to 10–14 M GA3. With increasing concentrations of GA3, up to 10–9 M, -amylase production increases similarly in protoplasts from both lots of seed, reaching a level approximately 2.7–3.8 times greater than when no GA3 is applied. GA3-induced -amylase production increases markedly as the concentration is raised from 10–9 M to 10–6 M, and the response then appears to be saturated. Over this part of the response curve protoplasts from the two seed lots differ markedly in their responsiveness to GA3. Those from seed stored at 25°C produce considerably more -amylase, >130-fold higher than the minus GA3 control, than those from seed stored at 4°C, <35-fold higher than the minus GA3 control. This apparent difference in the responsiveness of aleurone protoplasts to GA3 could be correlated with the loss of embryo dormancy in seed stored at 25°C. Seed stored at 4°C retained the dormancy characteristics present immediately after harvesting.  相似文献

Imbibition and germination experiments were conducted on the caryopses of wild oats (Avena fatua L.). The embryo envelopes, pericarp and aleurone layer, which completely cover the embryo-endosperm, do not form barriers against water uptake. The initial uptake of water is passive and the water moves across the pericarp with ease as it contains cracks; it is, however, transported across the aleurone layer through its cell walls into the endosperm and embryo of the caryopsis. The starchy endosperm enlarges due to water uptake causing the pericarp to rupture, thus exposing the aleuronelayer-covered seed. The aleurone layer is structurally heterogenous consistings of radially compressed irregular cells and cuboidal or radiallys tretched cells; the latter contains thicker walls. The former type is present along the abaxial side of the embryo and in the crease on the adaxial side of the caryopsis; the latter type covers the endosperm. The physical distention of the endosperm due to water uptake causes the rupture of the pericarp and the aleurone layer, and facilitates the emergence of the radicle and coleorhiza of the embryo during caryopsis germination.  相似文献

Functional analysis of a gibberellin-regulated wheat -amylase promoter, -Amy2/54, has indicated that three regions were essential for expression. By studying the ability of mutant promoters, containing a randomly inserted 22 bp excision linker, to direct expression in oat aleurone protoplasts we have refined the positions and extents of these three cis elements and also demonstrated the presence of two additional elements. By converting the linker insertions to either single base point mutations or deletions using the class IIS restriction endonuclease Bsm I we have shown that nucleotides –119 and –109 within the GARE –121GTAACAGAGTCTGG–108 and nucleotide –152 within the proposed element –156GATTGACTTGACC–144 are essential for high level expression from this promoter.  相似文献

Mixtures and monocultures of wheat (Triticum aestivum) and wild oat (Avena fatua), a common weedy competitor of wheat, were exposed to enhanced solar UV-B radiation simulating a 20% reduction in stratospheric ozone to assess the timing and seasonal development of the UV-B effects on light competition in these species. Results from two years of field study revealed that UV-B enhancement had no detectable effect on the magnitude or timing of seedling emergence in either species. End-of-season measurements showed significant UV-B inhibition of leaf insertion height in wild oat in mixture and monoculture in the second year (irrigated year) but not in the first year (drought year). Leaf insertion height of wheat was not affected by UV-B in either year. The UV-B treatment had no detectable effect on monoculture or total (combined species) mixture LAI but did significantly increase (5–7%) the fractional contribution of wheat to the mixture LAI after four weeks of growth in both years. In addition, the UV-B treatment had subtle effects on LAI height profiles with early season mixtures showing significant reductions in wild oat LAI in lower canopy layers in both years while midseason Year 2 mixtures showed significant reductions in wild oat LAI in upper canopy layers. The changes in canopy structure were found to significantly increase (6–7%) the proportional simulated clear sky canopy photosynthesis and light interception of wheat in mixture. These findings, and others, indicate that the effects of UV-B enhancement on competition are realized very early in canopy development and provide additional support for the hypothesis that UV-B enhancement may shift the balance of competition between these species indirectly by altering competitive interactions for light.  相似文献

Shock loading of toxic substances into natural waterways is apervasive practice which has substantial impacts on their biota.This paper describes the effects of shock (pulse) pollution loadingfrom two major industries on a river and wetland system in southernOntario, Canada. The assessment of shock loading frequencyindicated that sporadic discharges of polluted water occurred onaverage once every other day during the 38 days of monitoring inthe period April, 1986 to November, 1987.To estimate the frequency and intensity of the shock loads, anautomatic pump sampler that was activated by a thresholdconductivity was constructed and deployed. Samples were withdrawnfrom the river when the specific conductivity of the streamexceeded a threshold value of two times background. Backgroundspecific conductivity ranged between 200–250 S cm-1The impact of these shock load discharges on a downstream aquaticplant community in a Class One (Provincially Significant) wetlandwas examined. The area of Thompsons Creek upstream of the CyanamidChemical Company displayed a normal complement of aquatic organismswhile downstream of the companys main discharges for a distance ofnearly 2 km, the stream was barren of nearly all aquatic life.A reef-like structure in which nickel, chromium, and lead reachedrespectively 2900, 2100 and 2210 mg kg-1 dry weight ofsediment was located in the Welland River near the outfall of theAtlas-Mansfield storm sewer adjacent to the Atlas Specialty SteelsCompany. The average flow rate at the outfall of theAtlas-Mansfield storm sewer into the Welland River wasapproximately 28400 m3 day-1 in 1986 (MOE 1987).
