In order to aid the study of photoacclimation, a new programmable deviceis described which provides automatic on-line acquisition of in vivo cellabsorption in phytoplankton cultures. The system was used for a long-termstudy of Rhodomonas salina grown at constant photon flux density ina nitrate-limited continuous culture with different dilution rates. Particulate absorption measured at the red chlorophyll a (Chl a)maximum was not a good proxy of biomass, because of the large variabilityof cellular chlorophyll induced by nitrogen limitation. However, thedevice is well suited to automatic assessment of Chl a andphycoerythrin (PE) concentrations in phytoplankton cultures, if algal cellsize and concentration are measured in parallel to correct the packagingeffect. The effects of nitrogen limitation on Chl a and PE contentsand particle absorbance are discussed.  相似文献

噬菌斑平板计数是微生物学理论研究与实际应用中常用的方法之一。但由于平板上噬菌斑与背景反差小,而且往往出现几个噬菌斑相连,计算机识别时出现较大的误差,因此噬菌斑计数目前仍为人工方法。本文以λ噬菌体为材料,感染E.coli宿主细胞,获得噬菌斑。然后将噬菌斑制成电子图像。抽取图像中有代表性的区域,利用分水岭算法对图像进行分割处理,将相连的噬菌斑分割成单独的噬菌斑,然后利用基于区域生长法进行计数,结果与人工计数完全相同,表明我们建立的新方法可以用于噬菌斑计算机自动计数。  相似文献

【目的】建立同时检测副溶血性弧菌gyrase、tdh、trh基因的三重PCR快速检测方法。【方法】将已报道的这3种基因的引物加入一个PCR反应管中,对引物浓度和退火温度进行优化,找到最佳引物比例和扩增条件。通过特异性验证、灵敏度验证以及方法间对比进行方法确认,其PCR产物使用全自动毛细管电泳分析系统进行分析。【结果】仅在91、269、485 bp处分别出现预期DNA扩增条带;纯培养条件下,扩增gyrase、tdh、trh的菌浓度检测限分别为6.6×101、6.6×102和6.6×101 CFU/mL;本底干扰物存在时,扩增gyrase、tdh、trh的菌浓度检测限分别为6.6×103、6.6×104和6.6×103 CFU/mL;模板DNA浓度检测限为1.36μg/L。检测进境海产品时,检测结果和FDA 2004标准结果一致,且更易辨认和判断。【结论】此检测方法的成功建立,为副溶血性弧菌及携带tdh和/或trh基因的致病性副溶血性弧菌的检测提供了一种准确、高效、便捷的分子技术手段。  相似文献

PurposeAutomated treatment planning is a new frontier in radiotherapy. The Auto-Planning module of the Pinnacle3 treatment planning system (TPS) was evaluated for liver stereotactic body radiation therapy treatments.MethodsTen cases were included in the study. Six plans were generated for each case by four medical physics experts. The first two planned with Pinnacle TPS, both with manual module (MP) and Auto-Planning one (AP). The other two physicists generated two plans with Monaco TPS (VM). Treatment plan comparisons were then carried on the various dosimetric parameters of target and organs at risk, monitor units, number of segments, plan complexity metrics and human resource planning time. The user dependency of Auto-Planning was also tested and the plans were evaluated by a trained physician.ResultsStatistically significant differences (Anova test) were observed for spinal cord doses, plan average beam irregularity, number of segments, monitor units and human planning time. The Fisher-Hayter test applied to these parameters showed significant statistical differences between AP e MP for spinal cord doses and human planning time; between MP and VM for monitor units, number of segments and plan irregularity; for all those between AP and VM. The two plans created by different planners with AP were similar to each other.ConclusionsThe plans created with Auto-Planning were comparable to the manually generated plans. The time saved in planning enables the planner to commit more resources to more complex cases. The independence of the planner enables to standardize plan quality.  相似文献

Anthropogenic noise across the world's oceans threatens the ability of vocalizing marine species to communicate. Some species vocalize at key life stages or whilst foraging, and disruption to the acoustic habitat at these times could lead to adverse consequences at the population level. To investigate the risk of these impacts, we investigated the effect of vessel noise on the communication space of the Bryde's whale Balaenoptera edeni, an endangered species which vocalizes at low frequencies, and bigeye Pempheris adspersa, a nocturnal fish species which uses contact calls to maintain group cohesion while foraging. By combining long‐term acoustic monitoring data with AIS vessel‐tracking data and acoustic propagation modelling, the impact of vessel noise on their communication space was determined. Routine vessel passages cut down communication space by up to 61.5% for bigeyes and 87.4% for Bryde's whales. This influence of vessel noise on communication space exceeded natural variability for between 3.9 and 18.9% of the monitoring period. Additionally, during the closest point of approach of a large commercial vessel, <10 km from the listening station, the communication space of both species was reduced by a maximum of 99% compared to the ambient soundscape. These results suggest that vessel noise reduces communication space beyond the evolutionary context of these species and may have chronic effects on these populations. To combat this risk, we propose the application or extension of ship speed restrictions in ecologically significant areas, since our results indicate a reduction in sound source levels for vessels transiting at lower speeds.  相似文献

The transition among hominids from quadrupedalism to bipedalism resulted in modifications in their musculoskeletal morphology. It is unclear, however, whether changes in the circuitry of the CNS were also necessary in order to accommodate the unique balance requirements of two-limb support. This study addresses the issue of modifications in control strategies by investigating the rapid, automatic postural responses of feline and human subjects to sudden disturbances of balance in the anteroposterior (AP) direction while they stand quadrupedally and bipedally on movable platforms. Postural responses are characterized in terms of segmental adjustments, generated AP shear forces, and electromyographic activity. Feline and human subjects correct posture similarly when standing quadrupedally. Furthermore, both species correct stance primarily with their hindlimbs and use their forelimbs as supportive struts. In contrast, both species use completely different correctional strategies when standing bipedally. Morphological restrictions, however, prevent cats from adopting the pillar-like plantigrade posture of human beings. Thus, the correctional strategies of bipedal cats are distinct from those of bipedal human subjects. It is concluded that 1) automatic postural response patterns of quadrupedal Felis and bipedal Homo reflect the different biomechanical characteristics of the initial postures rather than species differences in CNS circuitry controlling stance; 2) hindlimb-dominated posture control is probably a common and relatively ancient pattern; and 3) reorganization of hominid CNS circuitry was probably unnecessary because hindlimb control was already a feature of the system.  相似文献

The human cerebral cortex may be subdivided into architectonic fields according to variations within its laminar structure. Studies have shown correspondences between the locations of functional activation foci and architectonic regions. In order to perform accurate localization of functional activation foci to architectonic regions, a parcellation algorithm capable of segmenting architectonic regions on in vivo imaging datasets is required. This paper presents a novel 3D model-based approach to directly detect cortical layers and classify architectonic fields. The column-like structure of the cortex is modeled using a Laplace equation method which generates a collection of intensity profiles that span the cortical mantle. Bayesian evidence for intensity profile elements belonging to hyper- or hypo-intense bands, which represent cell or myelin poor or rich layers in imaging data, is gathered. A non-isotropic Markov Random Field model is used to encourage contiguous bands as well as a penalty term that completes bands across highly curved cortical regions where neighbouring evidence for banding is strong. This algorithm is validated on a 3D histological dataset of a macaque brain with visible layering at intermediate resolution between high-resolution MRI and histology. The algorithm detects the myelin-rich Stria of Gennari and uses this as the basis for finding the Brodmann Area 17/18 boundary.  相似文献

Analysis of natural host-parasite relationships reveals the evolutionary forces that shape the delicate and unique specificity characteristic of such interactions. The accessory long gland-reservoir complex of the wasp Leptopilina heterotoma (Figitidae) produces venom with virus-like particles. Upon delivery, venom components delay host larval development and completely block host immune responses. The host range of this Drosophila endoparasitoid notably includes the highly-studied model organism, Drosophila melanogaster. Categorization of 827 unigenes, using similarity as an indicator of putative homology, reveals that approximately 25% are novel or classified as hypothetical proteins. Most of the remaining unigenes are related to processes involved in signaling, cell cycle, and cell physiology including detoxification, protein biogenesis, and hormone production. Analysis of L. heterotoma's predicted venom gland proteins demonstrates conservation among endo- and ectoparasitoids within the Apocrita (e.g., this wasp and the jewel wasp Nasonia vitripennis) and stinging aculeates (e.g., the honey bee and ants). Enzyme and KEGG pathway profiling predicts that kinases, esterases, and hydrolases may contribute to venom activity in this unique wasp. To our knowledge, this investigation is among the first functional genomic studies for a natural parasitic wasp of Drosophila. Our findings will help explain how L. heterotoma shuts down its hosts' immunity and shed light on the molecular basis of a natural arms race between these insects.  相似文献

目的:探讨血浆取代血清检测乙肝标志物的检测结果以及临床意义。方法:选取我科检测乙肝标志物的血样30份,分别放在抗凝剂管和普通干燥试管,采用全自动酶免疫分析仪检测HBsAg,将检测结果进行OD及S/CO值统计处理,并进行比较分析。结果:血清与二种血浆检测结果相关系数均0.99,二种血浆与血清结果相关关系良好(P>0.0 5),无显著性差异。结论:血浆代替血清完全可以用于全自动酶标分析仪进行检测,既可以节省预处理时间,又可以减少标本在分离吸移血清过程中出现差错,值得临床推广和应用。  相似文献

There is increasing evidence that individuals with non-specific low back pain (LBP) have altered movement coordination. However, the relationship of this neuromotor impairment to recurrent pain episodes is unknown. To assess coordination while minimizing the confounding influences of pain we characterized automatic postural responses to multi-directional support surface translations in individuals with a history of LBP who were not in an active episode of their pain. Twenty subjects with and 21 subjects without non-specific LBP stood on a platform that was translated unexpectedly in 12 directions. Net joint torques of the ankles, knees, hips, and trunk in the frontal and sagittal planes as well as surface electromyographs of 12 lower leg and trunk muscles were compared across perturbation directions to determine if individuals with LBP responded using a trunk stiffening strategy. Individuals with LBP demonstrated reduced peak trunk torques, and enhanced activation of the trunk and ankle muscle responses following perturbations. These results suggest that individuals with LBP use a strategy of trunk stiffening achieved through co-activation of trunk musculature, aided by enhanced distal responses, to respond to unexpected support surface perturbations. Notably, these neuromotor alterations persisted between active pain periods and could represent either movement patterns that have developed in response to pain or could reflect underlying impairments that may contribute to recurrent episodes of LBP.  相似文献
