毛木耳种质资源的酯酶同工酶分析 总被引:4,自引:0,他引:4
采用垂直平板聚丙烯酰胺凝胶电泳技术对毛木耳56个菌株酯酶同工酶的酶谱多样性进行了研究。结果表明,56份材料有一定的遗传变异,共检测到迁移率不同的10条谱带,各菌株分别具有1~6条酶带,共有26种酶谱类型。菌株被分成了8大群:第一群包括黄耳zh等34个菌株;第二群包括白背木耳等8个菌株;第三、第四群分别为黑木耳和915两个单独的菌株;第五群包括99等4个菌株;第六群包括43等4个菌株;第七群包括50385和AP067两个菌株;第八群包括小上3和杂交34两个菌株。酯酶同工酶分析技术是鉴别种及品种以下菌株的有效手段。 相似文献

The origin of echinoderms is one of the most crucial questions within the evolutionary history of deuterostomes. An ancestral position was suggested byGarstang, Romer andNichols. They also assumed that hemichordates and chordates are sistergroups. In all other hypotheses the echinoderms took a more derived position.Gislén, Jefferies andHolland viewed the hemichordates as basal to the deuterostomes and postulated that echinoderms and chordates are sistergroups. According toJollie, Peterson et al. andMooi &; David echinoderms and hemichordates are sistergroups.Gudo andGutmann adopted the view ofMetschnikoff who combined the hemichordates and echinoderms in the Ambulacraria; they supposed that echinoderms were derived from pterobranchs. This variety of views is linked with different approaches to phylogenetic reconstruction utilized by each of the authors.Garstang, Romer, Jefferies andGislén compared morphological features, in the case ofGislén andJefferies with some attention to fossil evidence, whereasJollie, Holland andGislén also considerd embryological aspects.Mooi &;David as well asPeterson et al. used modern embryological (epigenetical) approaches.Nichols combined functional morphology and comparative anatomy. 分别从显微和超微结构观察木耳菌种老化过程中菌丝细胞的形态变化。结果显示:接种后30d时,光镜下观察到菌丝结构均匀紧凑,细胞壁光滑;电镜下观察到细胞结构完整,内含物丰富,各种细胞器形态规整,没有老化现象。接种后60d时,光镜下菌丝部分肿胀,色泽加深;电镜下细胞壁疏松,线粒体和液泡肿大,细胞核不规则肿胀,核仁消失,脂肪滴和囊泡增多,并有少量电子致密度高的嗜锇性黑色颗粒状物质出现,表明菌丝开始老化。90d时,光镜下部分菌丝严重肿胀,且色泽更深;电镜下线粒体和液泡肿胀明显,部分细胞核破裂,脂肪滴、囊泡和电子致密度高的嗜锇性黑色颗粒状物质显著增多,细胞壁更加疏松。120d时,光镜下许多菌丝开始断裂,色泽进一步加深;电镜下细胞壁塌陷,膜系统也随之解体,线粒体等细胞器部分溶解消失。150d时,光镜下大部分菌丝完全断裂,并失去菌丝形态;电镜下细胞膜及其内含物已基本消失,只剩部分严重塌陷的细胞壁残骸。由此表明,木耳菌丝的老化是一个由个别向整体逐渐过渡的不可逆的过程。 相似文献

针对目前黑木耳代用料栽培中存在的问题,结合吉林省东部地区资源及气候特点,系统地对黑木耳栽培技术进行研究。结果表明:黑木耳最适培养料装量为800g;在接种时间为3月10日、置于相同培养条件下,最适栽培时间为4月下旬至5月初;菌丝体培养最适温度20~25℃,初期略高;最适水分条件为采用间歇式供水,干湿交替;最适光照条件为全日光。 相似文献

‘昊阳黄背1号’是黄背木耳新品种,适宜工厂化液体菌种扩繁和农法固体菌种生产。该品种是从上海引进的SH0072号菌株,经组织分离纯化、系统选育而成。成熟新鲜子实体为单片簇生型,有明显耳基,耳片有明显荷叶边;鲜耳子实体背面和腹面灰褐色,背面有短绒毛。菌丝生长适宜温度16-25℃,出耳适宜温度15-32℃;原基分化至第一潮耳采摘41-55d,一般可采3潮耳,出耳转化率达20.6%;抗毛木耳柱霉。适宜四川省成都市、德阳市和简阳市等地及相似生态区棚栽,冬季制袋,春夏季出耳。 相似文献

A simple and efficient procedure for the fluorescent labeling of saccharides is a prerequisite step for imaging the transport of polysaccharides in living cells. We report a one-pot strategy for the fluorescent labeling of saccharides with fluorescein-5-thiosemicarbazide (FTSC), which introduces the thiosemicarbazide group of FTSC to the aldehyde group at the reducing end of saccharides to form stable amino derivatives via reductive amination. The Glc-FTSC derivative was characterized by HPLC–MS, HRESIMS and NMR spectroscopy. Saccharides were quantitatively labeled with FTSC at 75 °C for 1 h under optimum reaction conditions. Fluorescence studies illustrated that the conjugation of FTSC to saccharides did not change its florescence properties (λex = 495 nm, λem = 517 nm), presenting desirable compatibility with commonly used fluorescence equipment. Polysaccharide AAG-FTSC derivatives exhibited rather low levels of cytotoxicity against rat thymus cells, and the fluorescent labeling procedure had slight impact on their anti-tumor activity. Results indicate that the assay neither introduces discernible cytotoxicity against living cells nor obviously alters the functional activities of polysaccharides, and provides a convenient, highly efficient fluorescent labeling approach for imaging the transport of polysaccharides in living cells. 相似文献

利用聚丙烯酰胺凝胶等电聚焦圆盘电泳,分析比较了黑木耳(Auricularia auricula)16个菌株和光帽鳞伞(Pholiota nameko)9个菌株的胞外漆酶同工酶酶谱。经多批次重复,得到了所有菌株的恒定酶谱。依照这些酶谱及各酶带等电点(pI)值的不同,确立了它们的漆酶标志位点相似系谱。16株黑木耳被区分成4种类型9个组。9株光帽鳞伞被分成两种类型3个组,但各组内之菌株间仍有异同。试验结果表明,把漆酶同工酶谱应用于菌株鉴别将是一种有意义的方法。 相似文献
