Complex I (CI) deficiency is the most common respiratory chain defect representing more than 30% of mitochondrial diseases. CI is an L-shaped multi-subunit complex with a peripheral arm protruding into the mitochondrial matrix and a membrane arm. CI sequentially assembled into main assembly intermediates: the P (pumping), Q (Quinone) and N (NADH dehydrogenase) modules. In this study, we analyzed 11 fibroblast cell lines derived from patients with inherited CI deficiency resulting from mutations in the nuclear or mitochondrial DNA and impacting these different modules. In patient cells carrying a mutation located in the matrix arm of CI, blue native-polyacrylamide gel electrophoresis (BN-PAGE) revealed a significant reduction of fully assembled CI enzyme and an accumulation of intermediates of the N module. In these cell lines with an assembly defect, NADH dehydrogenase activity was partly functional, even though CI was not fully assembled. We further demonstrated that this functional N module was responsible for ROS production through the reduced flavin mononucleotide. Due to the assembly defect, the FMN site was not re-oxidized leading to a significant oxidative stress in cell lines with an assembly defect. These findings not only highlight the relationship between CI assembly and oxidative stress, but also show the suitability of BN-PAGE analysis in evaluating the consequences of CI dysfunction. Moreover, these data suggest that the use of antioxidants may be particularly relevant for patients displaying a CI assembly defect.  相似文献

Aim  The study aimed to identify areas of endemism for aphids in the Qinghai-Tibetan Plateau and the Himalayas (QTPH), and to test congruence between patterns of endemism and patterns of overall species richness identified in a previous study.
Location  The QTPH.
Methods  A distribution data base of 326 endemic aphids in the QTPH was compiled. The study area was divided into a grid of 2°× 2° operative geographical units. Parsimony analysis of endemicity (PAE) was used to identify areas of endemism, and the diversity patterns of endemic species were then mapped using GIS.
Results  We identified 326 endemic species belonging to 138 genera within Adelgidae and 14 subfamilies of Aphididae. Five areas of endemism were found using PAE analysis: the eastern Himalayas, the western Himalayas, north-western Yunnan, southern Tibet and the eastern QTPH. Maps of patterns of endemism identified four major centres for endemic aphids, namely the western Himalayas, the eastern Himalayas (or Sikkim-Assam Himalayas), north-western Hengduan Mountains and the mountains of southern Gansu Province, and three minor centres, southern Tibet, south-eastern Tibet and the eastern Qinghai Province in the north-eastern QTPH.
Main conclusions  Our study identifies major centres of aphid endemism. Furthermore, there is a noticeable congruence between patterns of endemism and patterns of species richness. The patterns of endemism were most likely influenced by the recent uplift of the QTPH.  相似文献

Summary Simple correlations were calculated between nine different cms sources (cms-S, -R, -ML, -L, -CA, -EK, -C, -Rb, -T) on the basis of the weighted restoring reactions of 41 inbred lines. The Principal Component Analysis was applied to a 9 × 9 matrix which clearly grouped cytoplasms according to their similarities. The Principal Component I included S, R, ML, L, CA and EK cytoplasms; the Principal Component II contained C and Rb cytoplasms while T-cytoplasms was placed in Principal Component III. This corresponds to the main groupings indicated in the literature (Beckett 1971). However, after varimax rotation of the Principal Components, the S main group, including the 6 tested cytoplasms, fell into 3 subgroups: I.: S, R, ML; II.: L, CA; III.: EK.These data indicate that the Principal Component Analysis can be used to select a limited number of cms sources from the S group, representing the variability of the cytoplasmic gene pool of that group.  相似文献

通过中性景观模型和元胞自动机模型模拟了不同景观格局、不同竞争力和关键种群特征的外来入侵物种的传播动态,模拟结果表明:(1)竞争力强的外来入侵物种,可利用生境面积越大,集聚度越高,越有利于其入侵和传播,而对竞争力弱的外来入侵物种来说,可利用生境面积越小,越分散,越有利于其生存。竞争力强可以有效利用整块的高集聚度的资源,而弱竞争力的外来入侵物种为了逃避土著种的竞争往往趋向于分布在分散的小生境中;(2)可利用生境面积大于50%时,景观格局集聚度越小,竞争力弱的外来入侵物种适应环境的弛豫时间越长;(3)外来入侵物种的传播与种子产量呈正相关关系,与繁殖年龄呈负相关关系,拟合关系因景观格局和竞争而异;(4)平均迁移距离对外来入侵物种传播的影响最大,在管理过程中应根据不同的目标,不同的景观格局、竞争力和种群特征选择合适的管理措施。  相似文献

Butterfly pigmentation patterns are one of the most spectacular and vivid examples of pattern formation in biology. They have attracted much attention from experimentalists and theoreticians, who have tried to understand the underlying genetic, chemical and physical processes that lead to patterning. In this paper, we present a brief review of this field by first considering the generation of the localised, eyespot, patterns and then the formation of more globally controlled patterns. We present some new results applied to pattern formation on the wing of the mimetic butterfly Papilio dardanus.  相似文献

Modern theory predicts that relative parental investment of the sexes in their young is a key factor responsible for sexual selection. Seahorses and pipefishes (family Syngnathidae) are extraordinary among fishes in their remarkable adaptations for paternal care and frequent occurrences of sex-role reversals (i.e., female-female competition for mates), offering exceptional opportunities to test predictions of sexual selection theory. During mating, the female transfers eggs into or onto specialized egg-brooding structures that are located on either the male's abdomen or its tail, where they are osmoregulated, aerated, and nourished by specially adapted structures. All syngnathid males exhibit this form of parental care but the brooding structures vary, ranging from the simple ventral gluing areas of some pipefishes to the completely enclosed pouches found in seahorses. We present a molecular phylogeny that indicates that the diversification of pouch types is positively correlated with the major evolutionary radiation of the group, suggesting that this extreme development and diversification of paternal care may have been an important evolutionary innovation of the Syngnathidae. Based on recent studies that show that the complexity of brooding structures reflects the degree of paternal investment in several syngnathid species, we predicted sex-role reversals to be more common among species with more complex brooding structures. In contrast to this prediction, however, both parsimony- and likelihood-based reconstructions of the evolution of sex-role reversal in pipefishes and seahorses suggest multiple shifts in sex roles in the group, independent from the degree of brood pouch development. At the same time, our data demonstrate that sex-role reversal is positively associated with polygamous mating patterns, whereas most nonreversed species mate monogamously, suggesting that selection for polygamy or monogamy in pipefishes and seahorses may strongly influence sex roles in the wild.  相似文献

Mitochondria dynamically fuse and divide within cells, and the proper balance of fusion and fission is necessary for normal mitochondrial function, morphology, and distribution. Drp1 is a dynamin-related GTPase required for mitochondrial fission in mammalian cells. It harbors four distinct domains: GTP-binding, middle, insert B, and GTPase effector. A lethal mutation (A395D) within the Drp1 middle domain was reported in a neonate with microcephaly, abnormal brain development, optic atrophy, and lactic acidemia (Waterham, H. R., Koster, J., van Roermund, C. W., Mooyer, P. A., Wanders, R. J., and Leonard, J. V. (2007) N. Engl. J. Med. 356, 1736–1741). Mitochondria within patient-derived fibroblasts were markedly elongated, but the molecular mechanisms underlying these findings were not demonstrated. Because the middle domain is particularly important for the self-assembly of some dynamin superfamily proteins, we tested the hypothesis that this A395D mutation, and two other middle domain mutations (G350D, G363D) were important for Drp1 tetramerization, higher order assembly, and function. Although tetramerization appeared largely intact, each of these mutations compromised higher order assembly and assembly-dependent stimulation of Drp1 GTPase activity. Moreover, mutant Drp1 proteins exhibited impaired localization to mitochondria, indicating that this higher order assembly is important for mitochondrial recruitment, retention, or both. Overexpression of these middle domain mutants markedly inhibited mitochondrial division in cells. Thus, the Drp1 A395D lethal defect likely resulted in impaired higher order assembly of Drp1 at mitochondria, leading to decreased fission, elongated mitochondria, and altered cellular distribution of mitochondria.  相似文献

介绍近十几年来利用蜜蜂Apis mellifera L.、果蝇Drosophila melanogaster Meigen和寄生蜂等昆虫对学习和记忆行为研究的成果。这些研究表明昆虫的记忆形成是多阶段的。从短时记忆向长时记忆的形成过程中,cAMP反应元件结合蛋白(cAMP response element-bindingprotein,CREB)起重要的作用;而蘑菇体是学习、记忆形成的主要位点。已有的研究还表明昆虫记忆的动态是适应于不同物种的生态学需要。这些研究为探索昆虫和其它动物记忆的形成和生态适应性提供了理论基础。  相似文献
