在内循环气升式光生物反应器中 ,研究了钝顶螺旋藻 (SpirulinaplatensisGeitler)细胞的连续生长及其对碳源底物的利用特性。结果表明 :随着稀释率的增大 ,反应器中碳源浓度和细胞浓度分别呈上升和下降趋势 ,它们之间的关系可用Monod类型的方程很好地加以关联。细胞产率和碳消耗速率与稀释率的关系存在峰值现象 :在本实验条件下 ,最大细胞产率为 0 .36 2g/(L·d) ,最大碳消耗速率为 0 .177g/(L·d) ,此时稀释率为 0 .45 /d ,细胞浓度为OD560 =1.2 82 ,细胞对碳的得率系数为 2 .0 5 0g/g。所提出的连续培养动力学模型与实验数据拟合较好  相似文献

Photoinhibition in outdoor cultures of Spirulina platensis was studied by measuring the polyphasic rise of chlorophyll fluorescence transients, which provide information on the primary photochemistry of PSII. The maximum efficiency of PSII photochemustry (Fv/Fm) declined in response to daily increasing irradiance and recovered as daily irradiance decreased. The greatest inhibition (15%) in Fv/Fm was observed at 12:00 hr which responded to the highest irradiance. The absorption flux, the trapping flux, and the electron transport flux per PSII reaction center increased in response to daily increasing irradiance and decreased as irradiance decreased. The daily change in the concentration of PSII reaction centers followed the same pattern as Fv/Fm. However, no significant changes in the probability of electron transport beyond QAo) were observed during the day. The results suggest that the decrease in Fv/Fm induced by photoinhibition in outdoor Spirulina cultures was a result of the inactivation of PSII reaction centers. The results also suggest that the measurement of polyphasic fluorescence transients is a powerful tool to study the mechanism of photoinhibition in outdoor Spirulina cultures and to screen strains for photoinhibition tolerance. 该文应用原子力显微镜(AFM)纳米级的分辨率对钝顶螺旋藻(Spirulina platensis)表面微观形貌进行了研究,获得了扫描范围为5.000×5.000μm、1.000×1.000μm和400.0×400.0nm三组清晰、稳定的图像,并对其进行了线性分析。结果表明:螺旋藻表面由紧密且无序堆积的突起结构组成,其高度小于20nm;突起结构高度从3nm~15nm不等,平均高度约为8~9nm。此法用于生物体表面,操作简单、快速、灵敏度好且样品无损伤,结果令人满意。  相似文献

利用倍频 Nd:YAG激光 (波长 53 2 nm,功率 50 0 m W,功率密度 1 60 m W/cm2 )诱变钝顶螺旋藻 ,辐照时间为 1 5min、1 0 min、5min通过测定藻丝形态参数、叶绿素 a、β 胡萝卜素、生长速度 ,比较倍频 Nd:YAG激光对钝顶螺旋藻生长的影响。实验结果表明 :与出发株相比 ,经倍频 Nd:YAG激光辐照后 ,藻丝形态发生变化 ,藻丝长、螺旋数、螺旋长变小 ;1 5min,1 0 min辐照组出现螺旋变松驰 ;1 0 min,5min辐照组促进藻的生长和叶绿素 a含量提高 ,使生长速度提高。三个诱变时间剂量都有利于 β 胡萝卡素积累 ,含量增幅最高达 2 2 .3 %。  相似文献

体外观察螺旋藻及其水提物对肠道菌群生长的影响。结果表明 ,螺旋藻及其水提物对双歧杆菌和乳杆菌和生长具有显著的促进作用 (培养 48h,P<0 .0 0 1) ,对肠道其他菌亦有不同程度的促进作用 ,而对大肠埃希菌的促生长作用不显著 (P>0 .0 5 )。  相似文献

The growth behavior, photosynthetic productivity and carbon utilizing characteristics of filamentous cyanobacterium Spirulina platensis Geitler were investigated in continuous cultures in a 10 L internalloop airlift photobioreactor. It was found that the cell density and the amount of carbon in the reactor increased and decreased with dilution rate respectively, and the relationships among them could well be expressed by the modified Monod equation. When dilution rate was 0.45/d, the rate of cell growth and that of carbon assumption reached their maximum points 0.362 g/(L·d) and 0.177 g/(L·d) respectively. The cell growth yield against carbon was 2.050 g/g. The proposed model of continuous growth fitted the experimental data quite well.  相似文献

温度是影响节旋藻大规模培养中生物质产率的重要因素。筛选高产和耐受温度胁迫的优良藻株对提高节旋藻产量具有重要意义。从生产规模跑道池中分离到形态特征差异显著而亲缘关系很近的两株节旋藻Arthrospira sp.DICP-D(D)和Arthrospira sp.DICP-F(F),对其生物质积累性能和对低温、高温胁迫的耐受能力进行了评估。结果显示,尽管藻株D和藻株F在正常条件下拥有相似的生物质组成,在N胁迫和低温胁迫下拥有相同的碳水化合物积累能力,然而藻株D的生物质产率比相同条件下的藻株F高33%~230%。藻株D对高温胁迫的耐受能力是藻株F的2.1倍。藻株D在41℃和15℃下的生物质产率分别维持在正常温度下的73%和61%,显示其对温度胁迫的良好耐受能力。藻株D比藻株F拥有更有效的光保护机制,使其能够更好地适应胁迫环境。藻株D在生物质产率和胁迫耐受能力上的优良性状使其在户外大规模培养中具有良好的应用前景。  相似文献

Both freely suspended cells and immobilized cultures of Spirulina platensis, a blue-green alga, biotransformed exogenously fed codeine, an opium alkaloid, to morphine. The external addition of codeine to the culture medium did not affect the growth of S. platensis. Immobilization of Spirulina in a calcium alginate gel matrix was optimized by using 2% (w/v) sodium alginate and reducing the concentration of nutrients of Zarrouk's medium, which caused destabilization of the calcium alginate gel. The accumulation of morphine increased gradually and reached maxima of 330g 100ml–1 culture at 105h in freely suspended and 351g 100ml–1 at 96h in immobilized Spirulina cultures. Accumulation of morphine was detected only in the medium, whereas cells did not show accumulation. The immobilized Spirulina cultures showed marginally higher conversion of codeine to morphine over freely suspended cultures.  相似文献

Arthrospira platensis cultivation produces a saline spent medium that must be treated to allow its reuse, thus saving water and avoiding environmental pollution. This study evaluates the association of flocculation followed by adsorption to treat the spent medium by applying different concentrations of granular activated carbon (GAC) and ferric chloride (F), and using different residence times (T). The simultaneous optimization of the independent variables GAC, F, and T was performed using both a 23 central composite design and a response surface methodology. The cells cultivated in the medium obtained after the optimal conditions of treatment (GAC = 54.2 g L?1, F = 10.0 mg L?1, and T = 30.8 min) provided the highest maximum cell concentration, Xm = 3140 ± 77 mg L?1 in 0.5 L Erlenmeyer flasks with the highest protein biomass content (44.9%). The treated medium in such conditions was also used in a 3.5 L tubular photobioreactor (PBR), reaching Xm = 4033 ± 110 mg L?1 and biomass with high contents of both protein (47.3 ± 2.6%) and chlorophyll (9.7 ± 0.3 mg g dry cell?1). Therefore, this study can contribute to diminishing costs of A. platensis production by reusing its culture medium and improving its biomass quality in PBRs.  相似文献
