Efficient cryopreservation conditions for the edible alkalophilic cyanobacterium Arthrospira (Spirulina) platensis were investigated using a model strain A. platensis NIES-39. As a result, it was found that more than 60% of cells were viable upon thawing, when they had been frozen at a cooling rate of approximately ?1 °C min?1 in the presence of 10% (v/v) dimethyl sulfoxide. Further examination with other Arthrospira strains showed that many of them had strain-dependent optimal conditions for cryopreservation. For example, the best freezing conditions for A. platensis SAG 21.99 were snap-freezing in liquid nitrogen in the presence of 5% (v/v) dimethyl sulfoxide, while they were slow cooling at approximately ?1 °C min?1 in the presence of 10% (v/v) methanol for A. platensis NIES-46, NIES-2308 and UTEX 1926. The variety of successful cryopreservation conditions presented in this study is useful when attempting to cryopreserve various Arthrospira strains.  相似文献

Similar to other photosynthetic microorganisms, the cyanobacterium Arthrospira platensis can be used to produce pigments, single cell proteins, fatty acids (which can be used for bioenergy), food and feed supplements, and biofixation of CO2. Cultivation in a specifically designed tubular photobioreactor is suitable for photosynthetic biomass production, because the cultivation area can be reduced by distributing the microbial cells vertically, thus avoiding loss of ammonia and CO2. The aim of this study was to investigate the influence of light intensity and dilution rate on the photosynthetic efficiency and CO2 assimilation efficiency of A. platensis cultured in a tubular photobioreactor in a continuous process. Urea was used as a nitrogen source and CO2 as carbon source and for pH control. Steady‐state conditions were achieved in most of the runs, indicating that continuous cultivation of this cyanobacterium in a tubular photobioreactor could be an interesting alternative for the large‐scale fixation of CO2 to mitigate the greenhouse effect while producing high protein content biomass.  相似文献

为了研究细胞骨架蛋白FtsZ在螺旋藻形态建成中的作用,通过PCR克隆了ftsZ基因并进行原核表达,对表达的融合蛋白进行了纯化。通过免疫小鼠制备了FtsZ的多克隆抗体。分别用Western blot和免疫荧光技术检测螺旋藻不同形态藻丝体中ftsZ的表达和定位。结果表明,在两株不同螺旋藻Spirulina platensisFACHB869和FACHB882中,ftsZ在直线形藻丝体中的表达量都高于螺旋形藻丝体。免疫荧光定位结果显示,FtsZ蛋白在藻细胞中呈环状分布于细胞膜上,且这种环状结构在直线形藻丝体中排列较密而在螺旋形藻丝体中排列疏松。ftsZ在不同形态藻丝体中的表达量和细胞定位差异说明,细胞骨架蛋白FtsZ可能通过改变细胞刚性而参与螺旋藻形态建成。  相似文献

Regulatory properties of chorismate mutase from Corynebacterium glutamicum were studied using the dialyzed cell-free extract. The enzyme activity was strongly feedback inhibited by l-phenylalanine (90% inhibition at 0.1~1 mm) and almost completely by a pair of l-tyrosine and l-phenylalanine (each at 0.1~1 mm). The enzyme from phenylalanine auxotrophs was scarcely inhibited by l-tyrosine alone but the enzyme from a wild-type strain or a tyrosine auxotroph was weakly inhibited by l-tyrosine alone (40~50% inhibition, l-tyrosine at 1 mm). The enzyme activity was stimulated by l-tryptophan and the inhibition by l-phenylalanine alone or in the simultaneous presence of l-tyrosine was reversed by l-tryptophan. The Km value of the reaction for chorismate was 2.9 } 10?3 m. Formation of chorismate mutase was repressed by l-phenylalanine. A phenylalanine auxotrophic l-tyrosine producer, C. glutamicum 98–Tx–71, which is resistant to 3-amino-tyrosine, p-aminophenylanaine, p-fluorophenylalanine and tyrosine hydroxamate had chorismate mutase derepressed to two-fold level of the parent KY 10233. The enzyme in C. glutamicum seems to have two physiological roles; one is the control of the metabolic flow to l-phenylalanine and l-tyrosine biosynthesis and the other is the balanced partition of chorismate between l-phenylalanine-l-tyrosine biosynthesis and l-tryptophan biosynthesis.  相似文献

cpcHID操纵子序列用于钝顶节旋藻品系分类与鉴定的研究   总被引:4,自引:0,他引:4  
克隆并测定7株钝顶节旋藻品系的cpcHID操纵子序列,以及16SrRNA和16S-23SrRNA转录单元内间隔区(ITS)序列,进一步通过生物信息学和分子系统学等研究发现:(1)7株品系的cpcHID序列,以及16SrRNA和ITS序列具有很高的相似性。(2)基于7株品系cpcHID序列的GC含量绝对偏差平均值、碱基变异率和遗传距离系数普遍比基于16SrRNA和ITS序列的大。(3)基于cpcHID序列的分类结果与基于16SrRNA和ITS序列的十分相近。因此,cpcHID可作为节旋藻等蓝细菌分类与鉴定的一种新的分子标记,特别是以其丰富的信息量而在品系水平的分类鉴定中占有优势。  相似文献

Nostoc muscorum and Spirulina platensis were grown under phosphate deficiency in order to investigate the role of internal phosphate pool and activity of alkaline phosphatase on poly-β-hydroxybutyrate (PHB) accumulation. PHB accumulation in N. muscorum increased to 22.7% of dry weight (dw) after 4 day of phosphate deficiency, while the internal phosphate pool reduced to the level of 0.02 μM mg dw−1 at a maximum APase activity of 2.57 nM PNP mg dw−1 h−1. In contrary, S. platensis depicted maxima of 1.39 nM PNP mg dw−1 h−1 on day 30 of incubation, which was about 2 fold lower than the observed value of N. muscorum. PHB content in S. platensis remained low even after prolonged phosphate starvation, and a rise only up to 3.5% of dw was recorded on day 60 of phosphate deficiency. Supplementation of NADPH exogenously to S. platensis cultures grown under phosphate deficiency favoured PHB accumulation in 10, 20 and 30 days old cultures, but not in the cultures grown under phosphate deficiency for 60 days. The possible role of phosphate limitation on PHB accumulation is discussed.  相似文献

从富硒螺旋藻(Se richSpirulina platensis,Se-SP)中分离纯化高纯度的含硒别藻蓝蛋白(Se-containingallophycocyanin,Se-APC)并观察其生化特性。羟基磷灰石和DEAE-52柱层析方法结合制备电泳技术纯化Se-APC;光谱扫描、Native-PAGE、SDS-PAGE和IEF方法鉴定Se-APC生化特性;2,3-DAN荧光光度法检测蛋白质中Se含量。结果发现3种高纯度Se-APC的光谱特征分别与APCI、APCII、APCIII吻合;电泳鉴定它们可能都是(αβ)3,α、β亚基分别为18.3和15.7 kDa,其pI值分别为:4.76、4.85和5.02;3种Se-APC中Se含量分别为316、273和408μg/g,Se-APC经0.5mol/L NaSCN解聚和β-巯基乙醇变性处理后,蛋白质中Se含量依次减低并趋于稳定。结果提示Se-SP中APC可结合Se,APC中Se含量与其分子聚态有关,亚基中含Se量稳定,可能是以共价键方式结合,Se-APC生物活性及硒在蛋白质中的结合位点值得深入研究。  相似文献

硒是人和动物必需的微量元素 ,补硒可以防治多种疾病。有机硒具有低毒、高生物利用度的优点 ,目前主要寄希望于生物转化的途径来获得有机硒[1 ] 。植物对硒的生物有机化作用已有综述[2 ] ,并开发有富硒酵母[3 ] 、富硒菇类[4] 、富硒大蒜、富硒黄芪、富硒西洋参、富硒麦芽、富硒茶以及富硒鸡蛋、富硒牛奶等[5] 。螺旋藻是一种很有开发利用前景的藻类 ,但其含硒量极微 ,实验报道富硒螺旋藻对60 Co γ射线胸部照射大鼠诱发肺炎和早期肺、肝纤维增生有防治作用[6] 。在培养液中添加亚硒酸钠可以实现藻类对硒的富集和转化 ,而且螺旋藻对无机硒…  相似文献

Photosynthetic activity and growth physiology of Spirulina platensis (Nordstedt) Geitler cultures maintained at ultrahigh cell densities (i.e. above 100 mg chlorophyll-L?1) in a newly designed photobioreactor were investigated. Nitrogen (NaNO3) in standard Zarouk medium was characterized as a major nutrient-limiting factor in such cultures. The effect of ultrahigh cell density on photoinhibition of photosynthesis, as reflected by chlorophyll fluorescence and photosynthetic oxygen evolution, was studied: elevating the population density may arrest photoinhibition induced by high photon flux density, as well as low temperature. The relationship between incident irradiance and oxygen production rate was linear in situ for cultures at the optimal cell density, indicating that light limitation rather than light saturation or photoinhibition is the dominant condition outdoors in cultures of ultrahigh cell densities. In contrast with other reports, the extent of biomass loss at night due mainly to dark respiration was found to be relatively small when cell density was optimal, exerting only a minor effect on overall net productivity. Measurements of oxygen consumption at night revealed low rates of respiration, which may be explained by the low value of the volumetric mass transfer coefficient (KLa) of oxygen. Hence, reduced oxygen tension may play a role in preventing full expression of the respiratory potential in ultrahigh cell density cultures in which photoadaptive strategy may explain cell composition. Ultrahigh cell densities optimized with respect to the intensity of the light source, the length of the light path, and the extent of stirring represent the key for obtaining high output rates of cell mass and some natural products.  相似文献

The spiral structure of the cyanobacterium Arthrospira (Spirulina) platensis (Nordst.) Gomont was previously found to be altered by solar ultraviolet radiation (UVR, 280–400 nm). However, how photosynthetic active radiation (PAR, 400–700 nm) and UVR interact in regulating this morphological change remains unknown. Here, we show that the spiral structure of A. platensis (D-0083) was compressed under PAR alone at 30°C, but that at 20°C, the spirals compressed only when exposed to PAR with added UVR, and that UVR alone (the PAR was filtered out) did not tighten the spiral structure, although its presence accelerated morphological regulation by PAR. Their helix pitch decreased linearly as the cells received increased PAR doses, and was reversible when they were transferred back to low PAR levels. SDS-PAGE analysis showed that a 52.0 kDa periplasmic protein was more abundant in tighter filaments, which may have been responsible for the spiral compression. This spiral change together with the increased abundance of the protein made the cells more resistant to high PAR as well as UVR, resulting in a higher photochemical yield.  相似文献
