The role of cyclic 3′,5?AMP in modulating sarcoplasmic reticulum from fast skeletal muscle was studied. The rate of Ca2+ uptake was stimulated in the presence of protein kinase plus 1 μM cyclic AMP. The stimulation was absent when denatured protein kinase was used. When an adenylate cyclase inhibitor was added, the uptake rates fell to 55% of control. This decrease in rate was partially overcome by 1 μM cyclic AMP. A modulating role for cyclic AMP in fast skeletal muscle is proposed.  相似文献

Glow curves from spinach leaf discs infiltrated with o-phthalaldehyde (OPA) show significant similarity to those obtained by DCMU treatment which is known to block the electron flow from QA, the stable acceptor of Photosystem II (PS II). In both the cases, the thermoluminescence (TL) peak II (Q band) was intensified significantly, whereas peaks III and IV (B band) were suppressed. Total TL yield of the glow curve remained constant even when the leaf discs were infiltrated with high concentrations of OPA (4 mM) or with DCMU (100 M), indicating that even at these high concentrations no significant change in the number of species undergoing charge recombination in PS II occurred. However, studies with thylakoids revealed significant differences in the action of OPA and DCMU on PS II. Although OPA, at a certain concentration and time of incubation, reduced the B band intensity by about 50–70%, and completely abolished the detectable oxygen evolution, it still retained the TL flash yield pattern, and, thus, S state turnover. OPA is known to inhibit the oxidoreductase activity of in vitro Cyt b6/f (Bhagwat et al. (1993) Arch Biochem Biophys 304: 38–44). However, in the OPA treated thylakoids the extent of inhibition of O2 evolution was not reduced even in the presence of oxidized tetramethyl-p-phenylenediamine which accepts electrons from plastoquinol and feeds then directly to Photosystem I. This suggests that OPA inhibition is at a site prior to plastoquinone pool in the electron transport chain, in agreement with it being between QA and QB. However, an unusual feature of OPA inhibition is that even though all oxygen evolution was completely suppressed, a significant fraction of PS II centers were functional and turned over with the same periodicity of four in the absence of any added electron donor, an observation which appears to be similar to that reported by Wydrzynski (Wydrzynski et al. (1985) Biochim Biophys Acta 809: 125–136) with lauroylcholine chloride, a lipid analogue compound. The detailed chemistry of OPA inhibition remains to be studied. A bitter controversy had existed as to the minimum number of quanta required for the evolution of one molecule of oxygen in photosynthesis: Otto Warburg had insisted since 1923 that this value was 3–4, whereas Robert Emerson and others continued to obtain a value of 8–12 since the 1940s. It is shown in this letter that the 1931 Nobel-laureate of Physiology & Medicine Otto Warburg published, in his last and final paper, just before his death in 1970, a measured minimum quantum requirement of oxygen evolution of 12 at the lowest intensities of light he used. Although using his theory on photolyte, Warburg calculated a value of 3–4 for the quantum requirement, this is the first confirmation by Warburg of the higher measured quantum requirement. However, it has remained unknown to most investigators. It is expected that this information will be of general interest not only to those interested in the history and research on photosynthesis, but to the entire sci entific community, especially the writers of text books in biology, biochemistry and biophysics.  相似文献

We developed a method for the direct identification and quantification of carbohydrates in raw vegetable extracts using (13)C NMR spectroscopy without any preliminary step of precipitation or reduction of the components. This method has been validated (accuracy, precision and response linearity) using pure compounds and artificial mixtures before being applied to authentic ethanolic extracts of pine needles, pine wood and pine cones and fir twigs. We determined that carbohydrates represented from 15% to 35% of the crude extracts in which pinitol was the principal constituent accompanied by arabinitol, mannitol, glucose and fructose.  相似文献

In this article, the author examines the current arts education policies in the United States through the lens of Matthew Arnold's work and prose during his time as one of Her Majesty's School Inspectors in England and Wales in the nineteenth century. Arnold argued for government funding of public school systems, setting standards of quality for teacher education, and many other issues that still affect us today. His passion for a liberal education that included everyone, reinforced by his belief in the moral and humanistic goals that could be reached through an education in the arts, helped open the public schools to the humanities that we teach today. The author provides a valuable overview of what Arnold saw, not only in the English and Welsh schools but also in the French and German approach to education. Finally, the author leaves us with a concise list of what Arnold can teach us today about arts education.  相似文献

Abstract. A competitive effect hierarchy for 15 Namaqualand pioneer plant species was established by using the mean mass of the phytometer (Dimorphotheca sinuata) when grown in combination with itself and 14 other species. There were no clear groupings of species in the hierarchy. This competitive hierarchy (gradient) indicated which species are strong competitors (resulting in a low phytometer mass) with D. sinuata and which species are weak competitors (resulting in a high phytometer mass). Each plant species has a certain combination of plant traits which determines its life history strategy and competitive ability. Regressions of various plant traits (measured on plants grown singly) against phytometer biomass indicated which traits were significantly correlated. The traits, most being size-related, were: maximum shoot mass, total mass, stem mass, reproductive mass, leaf area, stem allocation, specific leaf area (SLA), vegetative height × diameter, leaf area ratio (LAR); and mean number of days to flower initiation. A forward stepwise multiple regression of the significant traits was used to determine an equation to predict competitive effect.  相似文献

The results were so far out of synch with the thinking of the times that it took many years for them to become a cozy part of the dogma of photosynthesis.
