为了深入了解青海沙棘果粉的品质,助其实现规模化生产,从1,1-二苯基苦基苯肼自由基(DPPH·)体系、羟基自由基(·OH)体系、超氧阴离子自由基(O-2·)体系和亚硝酸盐(NO-2)的清除效果方面着手,研究了其体外清除自由基活性。结果表明,青海沙棘果粉对DPPH·、·OH、O-2·和NO-2均有清除作用,且对DPPH·、·OH的清除能力优于O-2·和NO-2。  相似文献

帕金森病是人类第二大神经退行性疾病,虽然通过对帕金森病遗传家族的研究已发现不少与之相关的致病基因,然而其病因尚不清楚。在已发现的与帕金森病相关基因中,α-synuclein、LRRK2、Parkin、PINK1以及DJ-1也在调节神经元突触前囊泡的神经递质释放以及突触前囊泡循环过程中发挥重要作用。最近的一些研究表明突触前囊泡循环障碍在帕金森病发病过程中扮演重要角色。本文对上述基因在突触囊泡循环以及其突变导致帕金森病病程中的作用作一概述,并推测囊泡循环障碍在帕金森病发病过程中的作用机理,最后指出目前在该研究领域需要解决的一些问题。  相似文献

Mutant N-terminal huntingtin (Htt) protein resulting from Huntington's disease (HD) with expanded polyglutamine accumulates and forms aggregates in vulnerable neurons. Both ubiquitin proteasomai and autophagic pathways con- tribute to the degradation of mutant Htt. Here, we focus on the involvement of chaperone-mediated autophagy (CMA), a selective form of autophagy in the clearance of Htt. Selective catabolism in CMA is conferred by the presence of a KFERQ-Iike targeting motif in the substrates, by which molecular chaperones recognize the hydrophobic surfaces of the misfolded substrates, and transfer them to the lysosomal membrane protein type-2A, LAMP-2A. The substrates are taken into the lysosomes through LAMP-2A and are rapidly degraded by the lysosomal enzymes. Taken together, we summarize the recent evidence to elucidate that Htt is also a potential substrate of CMA. We propose that the manipulation of CMA could be a therapeutic strat- egy for HD.  相似文献

Two primary patterns of body size variation have been recorded in ectotherms in relation to latitudinal/altitudinal shifts. In some, body size increases with increasing latitude/altitude whereas, in others, body size decreases with increasing latitude/altitude. This clinal variation is generally assumed to be caused by local adaptation to environmental conditions however the selective variable(s) (temperature, humidity, diet quality, etc.) is still heavily debated. Here we investigate geographic variation in body size of dark and pale color morphs of males of the bush-cricket lsophya rizeensis collected from 15 locations along an elevation gradient ranging from 350 to 2 500 m. Using an information theoretical approach we evaluate the relative support of four different hypotheses (the temperature size rule, the moisture gradient hypothesis, the seasonal constraint hypothesis, and the primary productivity hypothesis) explaining body size variation along the altitudinal gradient. Body size variation in pale color morphs showed a curvilinear relationship with altitude while dark color morphs showed no variation in body size. Body size variation in pale color morphs was highly correlated with precipitation and temperature seasonality values thus giving strong support for the moisture gradient and seasonal constraint hypothesis. Our results reinforce the importance of gradients in humidity and seasonality over temperature in the creation of altitudinal body size clines and the role of selection for resistance to stress factors in the establishment of these clines. Whether a body size cline is observed or not might also depend on the phenotypic properties of the individuals, like coloration.  相似文献

Neurotoxins and alterations in Ca2+ homeostasis have been associated with Parkinson's disease (PD), but the role of store-operated Ca2+ entry channels is not well understood. Previous studies have shown the neurotoxicity of salsolinol and 1-methyl-4-phenylpyridinium ion on SH-SY5Y cells and cytoprotection induced by transient receptor potential protein 1 (TRPC1). In the present study, N-methyl-(R)-salsolinol was tested for its cellular toxicity and effects on TRPC1 expression. MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-dipbenyl- tetrazolium bromide) assays, DAPI (4',6-diamidino-2-pheny- lindole), fluorescein isothiocyanate-Annexin-V/propidium iodide, western blot analysis, and JC-1 labeling revealed that the three indicated drugs could induce caspase-dependent, mitochondrial-mediated apoptosis. Exposure of SH-SY5Y cells to the indicated drugs resulted in a significant decrease in thapsigargin-mediated Ca2+ influx and TRPC1 expression. Immnnocytochemistry experiments revealed that neurotoxins treatment induced TRPC1 translocation to the cytoplasm. Taken together, our results indicate that treatment with neurotoxins may alter Ca2+ homeostasis and induce mitochondrial-mediated caspase-dependent cytotoxicity, an important characteristic of PD.  相似文献

Wnts comprise a large family of proteins that have shown to be part of a signaling cascade that regulates several aspects of develop- ment including organogenesis, mid brain development as welt as stem cell proliferation. Wnt signaling pathway plays different roles in the development of neuronal circuits and also in the adult brain, where it regulates synaptic transmission and plasticity. It has been also implicated in various diseases including cancer and neurodegenerative diseases, reflecting its relevance in fundamental biological pro- cesses. This review summarizes the progress about Wnts function in mature nervous system with a focus on Alzheimer's disease (AD). We discuss the prospects of modulating canonical and non-canonical Wnt signaling as a strategy for neuroprotection. This will include the potential of Wnts to: (i) act as potent regulators of hippocampai synapses and impact in learning and memory; (ii) regulate adult neurogenesis; and finally (iii) control AD pathogenesis.  相似文献

The molecular mechanisms that regulate synapse formation have been well documented. However, little is known about the factors that modulate synaptic stability. Synapse loss is an early and invariant feature of neurodegenerative diseases including Alzheimer's lAD) and Parkinson's disease. Notably, in AD the extent of synapse loss correlates with the severity of the disease. Hence, understanding the molecular mechanisms that underlie synaptic maintenance is crucial to reveal potential targets that will allow the development of ther- apies to protect synapses. Writs play a central role in the formation and function of neuronal circuits. Moreover, Wnt signaling compo- nents are expressed in the adult brain suggesting their role in synaptic maintenance in the adult. Indeed, blockade of Wnts with the Wnt antagonist Dickkopf-1 (Dkkl) causes synapse disassembly in mature hippocampal cells. Dkkl is elevated in brain biopsies from AD patients and animal models. Consistent with these findings, Amyloid-β (Aβ) oUgomers induce the rapid expression of Dkkl. Importantly, Dkkl neutralizing antibodies protect synapses against Aβ toxicity, indicating that Dkkl is required for Aβ-mediated synapse loss. In this review, we discuss the role of Wnt signaling in synapse maintenance in the adult brain, particularly in relation to synaptic loss in neurodegenerative diseases.  相似文献

The conservation behavior framework is useful to identify key linkages between behavior and conservation practice. We apply this framework to a novel host-parasite system on the Galapagos Islands and ask if there have been changes in parasite oviposition behavior and host mortality patterns across the first decade (2004-2013) of its known association. The Dipteran parasite Philornis downsi was first discovered in Darwin's finch nests in 1997 and is the biggest threat to the survival of Galapagos land birds. Host mortality has increased over the past decade. In Dipterans, pupation and pupae size are determined by access to host resources. Here, we test the hypothesis that P downsi flies are laying eggs in finch nests earlier in the nestling phase to maximize larval feeding time and therefore chance of pupation success before host death. The results show fewer 1st instar larvae later in the host nesting cycle in support of earlier egg laying behavior by female flies. Between 2004 and 2013, parasite intensity increased from -28 to -48 parasites per nest, host mortality increased from -50% to -90%, and host age at death decreased from -11 to -5 days. The earlier age at host death was correlated with fewer pupae (from -50% to -20%) and smaller pupae size (-10% decrease). Changes in parasite behavior reveal new fitness costs to both the parasite and Darwin's finches. These findings un- derscore the need for urgent conservation action to save Darwin's finches from extinction due to a novel, lethal and introduced parasite [Current Zoology 60 (4): 542-550, 2014].  相似文献

目的 建立小鼠腹腔水液体积的检测方法并评价肾阳虚模型小鼠的水负荷状况.方法选取正常小鼠12 只,根据"阿基米德原理"改进实验方法测量小鼠的腹腔水液体积,对其腹腔水液体积测量的稳定性进行评价;选取正常小鼠5 只,利用小鼠腹腔水液检测方法对其进行线性关系考察;将SPF 级昆明种雄性小鼠30 只,随机分为正常组和肾阳虚模型组,肾阳虚模型组上午按0.08 mg/10 g 腹腔注射苯甲酸雌二醇注射,正常组腹腔注射相同剂量的大豆油,下午两组均腹腔注射1 mL 的生理盐水造成水负荷,连续15 d.结果该测量方法的稳定性和线性关系考察均取得了良好的效果;与正常组相比,造模后体重显著性降低(P <0.01),肛温显著性降低(P <0.01),趾温差异无显著性(P >0.05);自主活动明显减少(P <0.05);游泳时间明显降低(P <0.01),综合评价说明肾阳虚造模成功;肾阳虚模型组的腹腔水液测量体积整体表现出下降的趋势,腹水指数显著高于正常组(P <0.05).结论该检测方法可以快速、准确的测量出小鼠腹腔水液测量体积和腹水指数的变化,可以能较客观地评价中医证候的特点,为中医证候症状描述进一步提供实验数据支持.  相似文献
