Neurexin-1 is required for synapse formation and larvae associative learning in Drosophila 总被引:2,自引:0,他引:2
Neurexins are highly polymorphic cell-surface adhesive molecules in neurons. In cultured mammalian cell system, they were found to be involved in synaptogenesis. Here, we report for the first time that Drosophila neurexin is required for synapse formation and associative learning in larvae. Drosophila genome encodes a single functional neurexin (CG7050; Neurexin-1 or Nrx-1), which is a homolog of vertebrate alpha-neurexin. Neurexin-1 is expressed in central nervous system and highly enriched in synaptic regions of the ventral ganglion and brain. Neurexin-1 null mutants are viable and fertile, but have shortened lifespan. The synapse number is decreased in central nervous system in Neurexin-1 null mutants. In addition, Neurexin-1 null mutants exhibit associative learning defect in larvae. 相似文献

Interactions among mycorrhizae, atmospheric CO2 and soil N impact plant community composition 总被引:1,自引:0,他引:1
We examined plant community responses to interactions between arbuscular mycorrhizal (AM) fungi and availability of atmospheric CO2 and soil N. Communities of 14 plant species were grown in mesocosms containing living or killed AM fungal inoculum, ambient or elevated atmospheric CO2 and low or enriched soil N. After one growing season, significantly different plant communities existed in the different treatments. Plant species richness was lowest in +N mesocosms and highest in +AM + CO2 mesocosms. At ambient CO2, AM fungi reduced richness but at elevated CO2 they increased it. This was caused by changes in mortality rates of several C3 forbs and may suggest that CO2 enrichment ameliorates the carbon cost of some AM symbioses. Soil moisture was higher in +CO2 mesocosms but +AM counteracted this effect. These results suggest that AM symbioses may be important mediators of plant community responses to anthropogenic CO2 and N enrichment. 相似文献

本文研究了兰科石斛属鼓槌石斛的地理分布与菌根真菌区系组成的相关性。采用克隆文库技术对云南省西双版纳和临沧市两个地区的8份野生鼓槌石斛根部样品进行了分析。结果表明,共从两个地区的鼓槌石斛根部获得了14个真菌的可操作分类单元(OTU),主要隶属于胶膜菌科Tulasnellaceae和小纺锤菌目Atractiellales。分析鼓槌石斛对菌根真菌的专一性和偏好性发现,鼓槌石斛对菌根真菌专一性较低,每个植株都能同时与多种菌根真菌共生;不同地理分布的鼓槌石斛菌根真菌类群存在明显差异,两个地区的鼓槌石斛的菌根真菌中只有1个OTU是相同的,其余均不同。说明菌根真菌可能和鼓槌石斛的生态适应存在一定的相关性。 相似文献

Piriformospora indica association has been reported to increase biotic as well as abiotic stress tolerance of its host plants. We analyzed the beneficial effect of P. indica association on rice seedlings during high salt stress conditions (200 and 300 mM NaCl). The growth parameters of rice seedlings such as root and shoot lengths or fresh and dry weights were found to be enhanced in P. indica-inoculated rice seedlings as compared with non-inoculated control seedlings, irrespective of whether they are exposed to salt stress or not. However, salt-stressed seedlings performed much better in the presence of the fungus compared with non-inoculated control seedlings. The photosynthetic pigment content [chlorophyll (Chl) a, Chl b, and carotenoids] was significantly higher in P. indica-inoculated rice seedlings under high salt stress conditions as compared with salt-treated non-inoculated rice seedlings, in which these pigments were found to be decreased. Proline accumulation was also observed during P. indica colonization, which may help the inoculated plants to become salt tolerant. Taken together, P. indica rescues growth diminution of rice seedlings under salt stress. 相似文献

Association between arbuscular mycorrhizal fungi (AMF) and majority of terrestrial plant species provides many benefits to plants that range from stress alleviation and bioremediation in soils polluted with heavy metals to plant growth promotion and yield quantity. Some non-arbuscular mycorrhizal fungi such as, Trichoderma harzianum, are known to enhance the AMF symbiosis with vascular plants. However, information about their role in AMF symbiosis is still limited. Shoots of (Avocet S) wheat seedlings were sprayed with the fungal culture filtrate and gene expression patterns were analyzed in the treated tissues. An increase in the level of mRNA of arbuscular mycorrhizal protein comparing with control was found. The over-expression of this protein in wheat tissues might contribute in initiation of AMF colonization in wheat tissues. The result of this study can spark future researches to elucidate possible role of this protein in the symbiotic interaction mechanisms between soil AMF and various plant roots. 相似文献

Arbuscular Mycorrhizal fungi (AMF, Glomeromycota) form obligate symbiotic associations with the roots of most terrestrial plants. Our understanding of the molecular mechanisms enabling AMF propagation and AMF-host interaction is currently incomplete. Analysis of AMF proteomes could yield important insights and generate hypotheses on the nature and mechanism of AMF-plant symbiosis. Here, we examined the extraradical mycelium proteomic profile of the arbuscular mycorrhizal fungus Rhizophagus irregularis grown on Ri T-DNA transformed Chicory roots in a root organ culture setting. Our analysis detected 529 different peptides that mapped to 474 translated proteins in the R. irregularis genome. R. irregularis proteome was characterized by a high proportion of proteins (9.9 % of total, 21.4 % of proteins with functional prediction) mediating a wide range of signal transduction processes, e.g. Rho1 and Bmh2, Ca-signaling (calmodulin, and Ca channel protein), mTOR signaling (MAP3K7, and MAPKAP1), and phosphatidate signaling (phospholipase D1/2) proteins, as well as members of the Ras signaling pathway. In addition, the proteome contained an unusually large proportion (53.6 %) of hypothetical proteins, the majority of which (85.8 %) were Glomeromycota-specific. Forty-eight proteins were predicted to be surface/membrane associated, including multiple hypothetical proteins of yet-unrecognized functions. However, no evidence for the overproduction of specific proteins, previously implicated in promoting soil health and aggregation was obtained. Finally, the comparison of R. irregularis proteome to previously published AMF proteomes identified a core set of pathways and processes involved in AMF growth. We conclude that R. irregularis growth on chicory roots requires the activation of a wide range of signal transduction pathways, the secretion of multiple novel hitherto unrecognized Glomeromycota-specific proteins, and the expression of a wide array of surface-membrane associated proteins for cross kingdom cell-to-cell communications. 相似文献

Abstract: A possible role for protein kinases in the regulation of free cytosolic Ca2+ levels in nerve endings was investigated by testing the effect of several kinase inhibitors on the increase in cytosolic Ca2+ (monitored with the Ca2+ -sensitive dye fura-2) induced by depolarization with 15 or 30 mM K+ . The ability of various drugs to inhibit the cytosolic Ca2+ response appeared to correlate with their reported mechanism of action in inhibiting protein kinases. W-7 and trifluoperazine, drugs reported to inhibit calmodulin-dependent events, were effective inhibitors of the increase in cytosolic Ca2+ induced by high K+ depolarization, as was sphingosine, a drug that inhibits protein kinase C by binding to the regulatory site, but which also inhibits calcium/calmodulin kinase. On the other hand, drugs that inhibit protein kinases by binding to the catalytic site, such as H-7 (1 m/W ), staurosporine (1μ M ), and K252a (1μ M ), were ineffective. Activation of protein kinase C, which is blocked by each of these drugs, does not appear to be essential to the maintenance of elevated cytosolic Ca2+ in depolarized synaptosomes. All of the drugs, including sphingosine, that functionally inhibit the depolarization-induced elevation in cytosolic Ca2+ have in common the ability to bind to calmodulin. Because the drugs that inhibit protein kinases by competing with ATP binding at the active catalytic site did not block the response in this system, we suggest that a calmodulin or a calmodulin-like binding site participates in the regulation of Ca2+ increases after depolarization. 相似文献

We assessed whether (1) arbuscular mycorrhizal colonization of roots (RC) and/or plant responses to arbuscular mycorrhizae (MR) vary with plant phylogeny and (2) MR and RC can be more accurately predicted with a phylogenetic predictor relative to a null model and models with plant trait and taxonomic predictors. In a previous study, MR and RC of 95 grassland species were measured. We constructed a phylogeny for these species and found it explained variation in MR and RC. Next, we used multiple regressions to identify the models that most accurately predicted plant MR. Models including either phylogenetic or phenotypic and taxonomic information similarly improved our ability to predict MR relative to a null model. Our study illustrates the complex evolutionary associations among species and constraints of using phylogenetic information, relative to plant traits, to predict how a plant species will interact with AMF. 相似文献

城市湿地(urban wetland)植物多样性水平高,并具有积极的净化和美化环境的作用。本研究以浙江绍兴镜湖城市湿地公园为研究样地,通过ICP-AES检测了人工园区和次生林土壤中几种金属元素(Al、Cr、Cu、K和Zn)的含量,比较不同类型的菌根植物——黑麦草(Lolium perenne L.)(AM)、六月霜(Monochasma sauatieri Franch)(AM)和乌饭树(Vaccinium bracteatum Thunb)(ERM)对金属元素的富集作用。通过高通量测序分析各宿主植物根部真菌群落的组成和结构,比较其对宿主植物金属富集能力的影响。结果表明:(1)人工园区土壤中5种金属元素的含量均显著高于次生林下的土壤;(2)乌饭树对Al的富集能力较强,黑麦草和六月霜富集Cr能力都较强;(3)3种植物根部真菌主要来自Ascomycota,而乌饭树根部Basidiomycota也是优势真菌,AM真菌在黑麦草和六月霜根部真菌群落结构中较少,而公认的ERM真菌Helotiales和Sebacinales在乌饭树根部真菌群落中的比例较高,且与所测金属元素无显著相关性。绍兴镜湖城市湿地公园次生林土壤环境保持良好。 相似文献

This study used a plant bioassay to investigate the vesicular-arbuscular mycorrhizal (VAM) inoculum potential of soil from three vegetation types (fern, secondary forest, and grass) in an abandoned pasture in the tropical humid lowlands at La Selva, in northeastern Costa Rica. Growth, measured as seedling height, number of leaves, and total (above- and belowground) biomass, of Stryphnodendron microstachyum Poepp. et Endl. (Synon. S. excelsum Harms) seedlings was significantly lower when grown in soil inoculum from the fern areas than in soil inoculum from the forest and grass areas. However, S. microstachyum seedlings grown in the fern inoculum had significantly greater VAM colonization than seedlings grown in the forest and grass inoculum. In addition, roots collected from a dominant plant species from each of the three vegetation types showed that the fern (Nephrolepsis biserrata) had significantly greater mycorrhizal colonization than the tree (Pentaclethra macroloba (Willd.) Kuntze or the grass (Brachiaria spp.). The results of this study suggest that differences in mycorrhizal inoculum potential among vegetation types and its effects on seedling growth may have important implications for the restoration and management of degraded lands. 相似文献
