Isolation of a novel stable peptide from cultivated Raphanus sativus with peroxidase activity has been achieved using zinc ion precipitation followed by aqueous two phase extraction with a polyethylene glycol (PEG)/phosphate system. The best result was achieved with PEG 600/phosphate (28.8/13% w/v) which extracted 96.15% of total enzyme activity. The peptide has a molecular weight of 5.85 kD as determined by HPLC size exclusion method with an optimized pH of 6.  相似文献

The pathways of redox equilibration of quinols and quinones have been investigated. The rate-limiting reaction involves the couple QH?/QH· of the reducing quinol and the couple Q?/Q of the oxidising quinone. Three general mechanistic points may be surmised: (i) protonation/deprotonation reactions are not rate-limiting; (ii) all transfers occur in one-equivalent steps; (iii) electron transfers, but not hydrogen atom transfers, are the dominant features. In aprotic media, no rapid route of equilibration is available since the ionic species which are necessary for thermodynamically feasible routes of electron transfer cannot exist to any great extent. The relation of these results to models of biological quinone systems is discussed.  相似文献

The nature of the inorganic calcium phosphate in the casein micelle of cows' milk has been studied by high-resolution electron microscopy. No periodic lattice spacings could be imaged, and diffraction patterns were of the diffuse amorphous type. Short-range order of less than 15 Å may be present, but the results indicate that there is no long-range order in micellar calcium phosphate.  相似文献

Ca2+ induces a structural change in phosphatidylcholine-cardiolipin bilayers, which is visualised by freeze-fracturing as lipidic particles associated with the bilayer and is detected by 31P-NMR as isotropic motion of the phospholipids. The dynamic viscosities of dilute aqueous poly(ethylene glycol) and dextran, and poly(ethylene glycol)-dextran-water solutions have been measured. The poly(ethylene glycol) and dextran samples had average molecular masses of 8000 Da and 580 000 Da, respectively. To estimate the values of viscosity of poly(ethylene glycol)-dextran-water solutions, a Grunberg like equation has been proposed which takes into account the influence of poly(ethylene glycol) and dextran concentrations. The relative errors vary between 0.76 and 11.64 in absolute value.  相似文献

A micelle enhanced spectrofluorimetric method was developed for determination of Omarigliptin (OMG) based on its native fluorescence behavior. The interaction of OMG with surfactants and macromolecules was studied. In aqueous solution, the relative fluorescence intensity (RFI) of OMG was enhanced by 24% in the presence of Tween 80 at pH 3.5. The optimal conditions for the micelle enhanced fluorescence were attained by Minitab® program using Plackett–Burman factorial design. Pareto chart, contour plots and surface plots were used to exclude the insignificant variables and optimize the significant factors. The spectrofluorimeter was operated under synchronous mode using ?λ = 30 nm and recording the RFI of the intense narrow band at 267 nm for OMG in 0.5% w/v Tween 80 + 0.2 M acetate buffer (pH 3.5) system using water as diluent. Using synchronous scan mode offered many advantages including considerable reduction of spectral overlap and enhanced linearity of the calibrators. Validation parameters were satisfied over the concentration range 0.1–2 μg/ml. The developed method was the first analytical procedure for OMG assay in Marizev® tablets. Moreover, content uniformity testing and in vitro drug release of tablets were performed.  相似文献

利用滇重楼(Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.)种子外种皮和胚乳的水浸液对白菜(Brassica pekinensis(Lour.)Rupr.)、绿豆(Vigna radiata(Linn.)Wilczak)、小麦(Triticum aestivum L.)种子进行处理,研究滇重楼种子水浸液对3种植物种子萌发、幼苗生长和保护酶活性的影响,并利用GC-MS方法对滇重楼种子内源抑制物的成分进行分析。结果显示,不同浓度滇重楼外种皮、胚乳水浸液对上述3种受体植物的发芽率、苗高、根长及鲜重均产生影响,其作用强度和水浸液的浓度有关,总体上表现出低促高抑的双重浓度效应。滇重楼种子水浸液对白菜的影响作用最强,对绿豆的影响作用最弱,且胚乳水浸液的影响较外种皮强。不同浓度滇重楼种子外种皮和胚乳水浸液均能影响3种植物幼苗体内保护酶的活性,随着水浸液浓度的升高,叶片中超氧化物歧化酶(SOD)、过氧化物酶(POD)活性总体增加,与对照相比差异显著。白菜、小麦过氧化氢酶(CAT)活性减少,与对照相比差异显著;绿豆过氧化氢酶(CAT)活性增加,但与对照相比无显著差异。利用GC-MS方法从胚乳和外种皮水浸液中分别检出8种和2种物质。研究结果表明滇重楼种子中存在内源抑制物质,可能是导致种子休眠的原因;种子水浸液可能通过影响植物幼苗保护酶的活性进而影响其正常生长;有机酸类物质可能是滇重楼种子内源抑制物之一。  相似文献

The amount of casein found in the milk of the tammar wallaby increases as lactation progresses. The increase is due to increasing amounts of β-casein; the α-casein remains largely constant. The α-casein is the more highly phosphorylated; the most abundant form is the 10-P, throughout lactation. The level of phosphorylation of β-casein shifts to lower average values in late lactation, possibly indicating the enzymatic reaction is overloaded by the increasing amounts of β-casein. Unlike bovine casein micelles, the wallaby micelles are not completely disrupted at pH 7.0 by sequestration of their calcium content with ethylene diamine tetraacetic acid (EDTA). Complete disruption only follows the addition of sodium dodecyl sulphate, indicating considerably greater importance for hydrophobic bonds in maintaining their integrity. This micellar behaviour indicates that, despite the evolutionary divergence of marsupials millennia ago, the caseins of wallaby milk assemble into micelles in much the same fashion as in bovine milk.  相似文献
