Summary The radiolysis of aqueous solutions (O2-free) of HCN and NH4CN was examined at very large doses of60Co gamma radiation (up to 230 Mrad). In this dose range the cyanide initially present (0.12 M) is decomposed and only its radiolytic products participate in the radiation-induced chemical process. It has been found that the weight of the dry residue containing the mixture of nonvolatile radiolytic products increases as doses increase up to 40 Mrad (up to about 4 g/l), but with further dose increases remains practically unchanged (NH4CN) or decreases slightly (HCN). Carboxylic and amino acids are present in overirradiated samples. At increasing doses their concentrations decrease, with the exception of oxalic and malonic acids, which are continually produced and accumulate. This is also the case with the abundant NH3 and CO2, as well as with several other products that were generated at lower radiation-chemical yields. The molecular weights of the radiolytic products are up to 20,000 daltons throughout the dose range studied. Their amounts gradually change with increasing doses above 30 Mrad: The compounds with Mw between 2,000 and 6,000 daltons become more abundant, while the amounts of polymers with Mw between 6,000 and 20,000 decrease. The relevance of these findings for studies of chemical evolution is considered.  相似文献

Palladium nanoparticles enzyme aggregate (PANEA) were prepared from Candida antarctica B lipase and palladium salt by precipitation and subsequent in situ Pd nanoparticle formation. This heterogeneous catalyst was successfully used for the Suzuki–Miyaura cross-coupling reaction between bromobenzene with different phenylboronic acid derivatives under mild reaction conditions and using low Pd amount. The nanocatalyst exhibited the highest catalytic activity in a mixture of methanol/water (1:1), obtaining good to excellent product yields from the cross-coupling reaction. A variety of functional groups were accepted and the catalyst was recycled 4 times without activity loss.  相似文献

Alpha 2-macroglobulin (α2-M) is a major proteinase inhibitor in human blood and tissue. Besides its antiproteolytic potential, α2-M was found to modulate antigen- and mitogen-driven immune responses and cell growth by binding and transporting distinct cytokines, growth factors and hormones. The inhibitor is cleared from circulation by binding to a multifunctional cellular receptor present on different cell types. α2-M, as well as its receptor, are capable of binding a variety of ligands. In the present study we have applied aqueous two-phase systems to analyze the interaction of IL-1β and α2-M receptor to different forms of α2-M. The partition of IL-1β was changed by addition of transformed α2-M to the two-phase systems rather than by the native inhibitor. The interaction between IL-1β and α2-M was enhanced by divalent cations. In addition, the complex formation between 125I-labelled receptor and α2-M could clearly be demonstrated by partitioning. In the presence of divalent cations, transformed α2-M, in contrast to the native inhibitor, effectively changed the partition of the receptor. However, the observed alteration of the partition coefficient was found to be less compared with the values obtained by partitioning of the receptor in the presence of whole plasma containing the inhibitor in equivalent concentrations. The results indicate that other components of the plasma exist which competitively bind to the receptor but independent of Ca2+-ions.  相似文献

Rat carrier erythrocytes prepared by hypotonic dialysis (80 mOsm/kg) are a heterogeneous cell population that can be fractionated into two-well-defined cell subpopulations by a single partition step, in charge-sensitive dextran-poly(ethylene glycol) aqueous two-phase systems. One subpopulation (65% of total cells) has a decreased cell surface charge and is partitioned at the interface in a single step and then fractionated by counter-current distribution as a low-G subpopulation. The other subpopulation (35% of total cells) has charge surface properties more like those of the untreated control rat erythrocytes. These last cells are partitioned in the top phase in a single step and then fractionated by counter-current distribution as a high-G subpopulation. Partitioning is more effective in reducing cell heterogeneity in hypotonized rat erythrocyte populations than is density separation in Ficoll-paque which only separates a small less dense cell subpopulation (5% of total cells), with the most fragile cells, from a larger and more dense cell subpopulation (95% of total cells), with a mixture of fragile and normal cells. This simple cell separation procedure quickly reduces carrier erythrocyte heterogeneity in a single partitioning step so it can be used to prepare cells for in vivo studies.  相似文献

Cell wall storage polysaccharides from Brazilian legume seeds of Dimorphandra mollis, Schizolobium parahybum (galactomannans), Copaifera langsdorffii, Hymenaea courbaril (xyloglucans) and the galactan from cotyledons of the Mediterranean species Lupinus angustifolius were extracted and their apparent molecular masses were determined by high-performance size exclusion chromatography analysis. They were, to a large degree, polydisperse, showing molecular masses that varied from 100 000 to 2 000 000. Polyethylene glycol (PEG, 1500, 4000, 6000 and 8000), sodium citrate and dextran (73 000, 60 000–90 000, 505 000 and 2 000 000) were used for investigating phase formation with the seed polysaccharides. Galactomannans and xyloglucans demonstrated phase formation with sodium citrate concentrations lower than 30%, as well as dextrans and polyethylene glycol, and formed gels in the presence of high concentrations of sodium citrate (above 30%). Galactan did not promote phase formation with any of the reagents used. On the basis of the results obtained, the possibility of using legume seed polysaccharides for the partitioning and purification of polysaccharide enzymes in aqueous two-phase systems is suggested.  相似文献

Separation and subfractionation of cells on the basis of subtle differences in surface properties by partitioning in dextran-poly(ethylene glycol) aqueous phase systems is an established method. We report here that the incorporation of fetal bovine serum into such systems permits countercurrent distribution of small quantities of cells (∼106). In the absence of serum such small quantities of cells are lost (probably by adherence) and cannot be recovered after countercurrent distribution.  相似文献

Oxidation of erythrocyte membrane SH-groups by diamide and tetrathionate induces cross-linking of spectrin (Haest, C.W.M., Kamp, D., Plasa, G. and Deuticke, B. (1977) Biochim. Biophys. Acta 469, 226–230). This cross-linking was now shown to go along with a concentration- and time-dependent enhancement of membrane permeability for hydrophilic nonelectrolytes and ions. The enhancement is specific for oxidative SH-group modifications, is reversible by reduction of the induced disulfides, can be suppressed by a very brief pre-treatment of the cells with low concentrations of N-ethylmaleimide and is strongly temperature-dependent. The pathway of the induced permeability discriminates nonelectrolytes on the basis of molecular size and exhibits a very low activation energy (Ea 3–8 kcal/mol). These findings are reconcilable with the formation of a somewhat inhomogeneous population of aqueous pores with radii probably ? 0.65 nm. Estimated pore numbers vary with the size of the probe molecule. Assuming a diffusion coefficient as in bulk water within the pore, at least 20 pores per cell have to be postulated; more realistic lower diffusion coefficients increase that number. Alterations of the lipid domain by changes of cholesterol contents and insertion of hexanol or nonionic detergents alter the number or size of the pores. Since aggregation of skeletal and intrinsic membrane proteins also occurs after the SH-oxidation, in parallel to the formation of membrane leaks, one may consider (a) defects in the disturbed bilayer interface, (b) a mismatch between lipid and intrinsic proteins or (c) channels inbetween aggregated intrinsic proteins as structures forming the pores induced by diamide treatment.  相似文献

Abstract: The ocular ciliary epithelium, the site of aqueous humor secretion in the mammalian eye, is believed to play a key function in signaling mechanisms that regulate the rate of secretion, and thus intraocular pressure. One possible way of mediating these signaling functions is through neuropeptides and hormones secreted into the aqueous humor and acting on target tissues. We recently identified a cDNA clone sharing 100% identity with carboxypeptidase E (CPE), a neuropeptide-processing enzyme. Utilizing polymerase chain reaction, we further identified and characterized another processing enzyme, the peptidylglycine α-amidating monooxygenase (PAM), and the neuropeptide secretogranin II, a molecular marker restricted to neuroendocrine tissues. Using specific probes, we found that the nonpigmented ciliary epithelial cells express CPE, PAM, and secretogranin II mRNA, and protein. We also found that CPE and secretogranin II are abundant in aqueous humor. Treatment of cultured ciliary epithelial cells with veratridine and phorbol ester up-regulates CPE and PAM. Secretogranin II was found to be induced by veratridine, whereas phorbol ester had little effect, suggesting different mechanisms for secretion. The results demonstrate that secretogranin II, CPE, and PAM represent a specialized group of neuropeptide and neuropeptide-processing enzymes secreted by the ciliary epithelial cells which may confer to them neuroendocrine functions in cell-cell communication or cell signaling.  相似文献

Monosaccharides are selectively converted to monobenzoates in a base-catalyzed reaction with benzoyl methyl phosphate (BzMP) and a lanthanum salt in water. Yields are reported in terms of formation of the ester, which competes with hydrolysis of BzMP, to give an estimate of the efficiency of the conversion of the sugar. Higher conversions can be achieved using excess reagent. Regioselectivity is influenced by the structure of the glycoside. For example, the reaction leads to different product distributions from alpha- and beta-anomers of the glycosides. The reaction combination provides a basis for efficient ester formation in specific geometric situations, providing a means of identification as well as modification.  相似文献
