A water-insoluble (1-->3)-beta-D-glucan isolated from fresh sclerotium of Poria cocos was, respectively, sulfated, carboxymethylated, methylated, hydroxyethylated, and hydroxypropylated, to afford five water-soluble derivatives. Their weight-average molecular masses (Mw) and intrinsic viscosities ([eta]) were determined by size-exclusion chromatography combined with laser light scattering (SEC-LLS), LLS, and viscometry in phosphate buffer solution (PBS) at 37 degrees C. The antitumor activities, against Sarcoma 180 tumor cell (S-180) and gastric carcinoma cell strain (MKN-45 and SGC-7901) of the native beta-glucan and the five derivatives, were tested in vitro and in vivo. The Mw values of the five derivatives in PBS were determined to be 3.8 x 10(4), 18.9 x 10(4), 16.0 x 10(4), 76.8 x 10(4), and 224.3 x 10(4), respectively. The high Mw values of the hydroxyethylated and hydroxypropylated derivatives in aqueous solution resulted from aggregation, and their true Mw values obtained in dimethyl sulfoxide were 20.1 x 10(4) and 19.1 x 10(4). The sulfated and carboxymethylated derivatives having DS of 1.0-1.3 show good water solubility, and exist as relatively expanded chains in aqueous solution. Interestingly, the native beta-glucan did not show antitumor activity, whereas the sulfated and carboxymethylated derivatives exhibit significant antitumor activities against S-180 and gastric carcinoma tumor cells. This work showed that good water solubility, relatively high chain stiffness, and moderate molecular mass of the derivatives in aqueous solution contribute beneficial to enhancement of antitumor activity.  相似文献

A series of water soluble molybdenocene complexes of general formula [(η5-C5H5)2Mo(L)]Cl2 (L=6-mercaptopurine (2), 6-mercaptopurine ribose (3), 2-amino-6-mercaptopurine (4), 2-amino-6-mercaptopurine ribose (5)) have been prepared by reacting Cp2MoCl2 (1) with the corresponding thionucleobase/thionucleoside in a (2:1) THF/MeOH solvent mixture. The complexes have been characterized by spectroscopic methods (NMR, UV-Vis, IR and MS). 1H NMR spectroscopic data (DMSO-d6) on the complexes suggest a S-Mo-N(7) coordination by the thionucleobase/thionucleoside. In buffer solution NMR data suggest that the thionucleobase/thionucleoside remains coordinated to molybdenum probably through S(6) and assisted by either N(7) or N(1) atoms. Intermediate species such as [Cp2Mo(η1-L)(H2O)]2+/1+ where the L is acting as monodentate ligand are possible in solution. Electrochemical characterization has also been pursued by cyclic voltammetry in DMSO and buffer solution. In DMSO, the complexes including the molybdenocene dichloride exhibit reversible redox behavior. On the other hand, in buffer solution, the oxidation process is irreversible for all the species.  相似文献

In an attempt to determine the relationships between the plant profiles (country of collection, taxonomy, plant part) and the compound classes isolated with cytotoxic activity against a panel of human tumor cell lines, the data compiled from a 15-year anticancer drug-discovery project were subjected to an analysis of variance (ANOVA). The results indicate significant trends in cytotoxic activity relative to collection location, taxonomy, plant part, and compound classes isolated. Plant collections were made in tropical forests in six countries, with collections from Ecuador resulting in higher activity than those from Indonesia and Peru. Interestingly, collections from Florida were not statistically different than those from the countries with higher biodiversity. One hundred and forty-five families were represented in the collections, with the Clusiaceae, Elaeocarpaceae, Meliaceae, and Rubiaceae having low ED50 (half maximal effective dose) values. Especially active genera included Aglaia, Casearia, Exostema, Mallotus, and Trichosanthes. Roots and below-ground plant materials were significantly more active than above-ground materials. Cucurbitacins, flavaglines, anthraquinones, fatty acids, tropane alkaloids, lignans, and sesquiterpenoids were significantly more active than xanthones and oligorhamnosides. The results from this study should serve as a guide for future plant collection endeavors for anticancer drug discovery.  相似文献

This work reports the synthesis, characterization, and aqueous chemistry of a series of cytotoxic [Au(polypyridyl)Cl2]PF6 complexes {(where polypyridyl = dipyrido[3,2-f:2′,3′-h] quinoxaline (DPQ), dipyrido[3,2-a:2′,3′-c] phenazine (DPPZ) and dipyrido[3,2-a:2′,3′-c](6,7,8,9-tetrahydro) phenazine (DPQC))}. The crystal structure of [Au(DPQ)Cl2]PF6 was determined as example of the series and exhibits the anticipated square planar geometry common for d8 coordination complexes. The crystals of the complex belong to the space group P21/n with a = 7.624(2) Å, b = 18.274(5) Å, c = 14.411(14) Å, β = 98.03(3)°, and Z = 4. In 1H NMR studies of these compounds in the presence of aqueous buffer, all four complexes rapidly converted to the dihydroxy species [Au(polypyridyl)(OH)2] in a stepwise fashion. However, the [Au(polypyridyl)]3+ fragment believed to impart cytotoxicity in human ovarian cancer cell lines (A2780) remained intact and appeared stable for days. It was also noted that these Au(III) complexes were readily reduced in the presence of the common biological reducing agents, reduced glutathione and sodium ascorbate. How solution and redox stability may affect the biological activity of these novel Au(III) complexes is discussed.  相似文献

Bacillus subtilis W-118, a strain that produces antifungal materials, excreted a chitinase when cultured in a medium containing shrimp- and crab-shell powder as the major carbon source. This chitinase, purified by sequential chromatography, had a molecular mass of 20,600 Da and a pI of 6. The optimum pH, optimum temperature, and pH stability of the chitinase were pH 6, 37 degrees C, and pH 5-7, respectively. The unique characteristics of the purified chitinase include low molecular mass and acidic pI. In the investigation of the inhibitory activity, it was found that the growth of Fusarium oxysporum was 100% inhibited after incubation for 1 day with sterilized W-118 chitinase solution (5.6 units/mL). The chitinase hydrolyzates of chitin with low degrees of polymerization (DP 1-6) were analyzed by HPLC. Longer reaction times led to the generation of chitin oligosaccharides with lower DP. The chitin oligosaccharides were examined for their inhibitory effects on F. oxysporum and human leukemia cell lines.  相似文献

As the increasing emergence of multi-drug resistant tumor cells, there is an urgent need for developing new chemotherapeutic agents. NK-lysin was a novel effector of cytotoxic T cells and natural killer (NK) cells and had broad antimicrobial activity. In this study, we developed a core region of NK-lysin termed NK-18, and studied its antitumor activity and possible action mode. Our results showed that NK-18 (with 18 amino acids) possesses potent antitumor activity against bladder and prostate cancer cells by disrupting the integrity of cell membrane, but has negligible hemolysis activity against mouse erythrocytes. In addition, CD spectra was employed to study its conformation in membrane mimicking environment. NK-18 takes a standard α-helical conformation in membrane mimicking environment, which could be accounted for its more potent antitumor activity compared with its low α-helical content homologous derivatives. These findings together with its shorter amino acid sequence and lower synthesis cost suggest that NK-18 could present an alternative therapeutic strategy to cancer chemotherapy and play a promising role in fighting the multi-drug resistant tumors.  相似文献

Single factor experiment and response surface methodology were conducted to optimize the production conditions for polysaccharide with higher antitumor activity from mushroom fermentation broth. Based on the results of single factor experiments, a central composite design was applied to the following optimization and second-order polynomial regression models were established. As the yield and antitumor activity of the polysaccharides were considered with equal weight, the optimal conditions were that ethanol concentration and pH of the broth was adjusted to 85.00% and 7.70 respectively, then precipitated at 12 °C for 1 h and dried the precipitates at 40 °C. The verification experiments completed under the optimal conditions gave the polysaccharides yield of 9.843 ± 0.217 g/l, and the cancer cell growth inhibitory rate of 83.69 ± 0.167% by the polysaccharides, which were in close agreement with values predicted by the models (RSD < 5%).  相似文献

The extracellular polysaccharides (EPSs) isolated from Porphyridium cruentum were degraded by hermetical-microwave and H2O2 under ultrasonic waves. Six products were obtained with molecular weights of 6.53, 256, 606, 802.6, 903.3 and 1002 kDa. The antitumor and immunomodulatory activities of different-molecular-weight (MW) polysaccharides were evaluated by the S180-tumor-bearing mouse model in vivo and peritoneal macrophage activation in vitro. The degraded EPSs all showed clear immunomodulation to different extents. The MW of the EPSs had a notable effect on their activity. The 6.53-kDa fragment had the strongest immunoenhancing activity. Different doses of EPS all inhibited the growth of the implanted S180 tumor. The tumor inhibition index at high, middle and low doses was 53.3%, 47.5% and 40.5%, respectively. In addition, three different concentrations of EPS significantly increased lymphocyte proliferation, which indicated the unique mechanism of the antitumor effect of EPS.  相似文献

The in vitro antitumor activities and in vivo immunomodulatory effects of water-soluble highly branched heteropolysaccharides isolated from Rhizoma panacis Japonici were evaluated by three cell lines and BALB/c mice implanted with Sarcoma 180 (S-180) tumors. The heteropolysaccharides showed potent tumor therapeutic effect by potentiating the animal's immune responses including an increase in white blood cell count and lymphocyte number. The heteropolysaccharides also induced apoptosis in the S-180 cells and alleviate the short-term side effect of cisplatin (DDP) to the animal during the treatment period. The bioactivity of the heteropolysaccharides might be due to their unique structures that had an α-(1 → 4)-d-glucan main chain with side chains containing mannose and galactose residues, a spherical chain conformation, as well as high water solubility, all of which facilitated their interaction with the surface receptors of the immune cells.  相似文献

A hot water-extracted polysaccharide fraction (PFCM) of Passiflora edulis was characterized by microanalysis, infrared spectroscopy, NMR and high performance size-exclusion chromatography. The major component in PFCM is (1 → 4) linked galacturonic acid (esterified and unesterified). Neutral sugars such as arabinose, glucose, rhamnose, mannose, and fucose were also present. Traces of xylose and ribose were detected. The PFCM sample had a similar molar mass to that of pectin extracted from P. edulis under acidic conditions. Sarcoma 180 tumors treated with PFCM showed a growth inhibition ratio ranging from 40.59% to 48.73% depending on the dosage and type of PFCM administration (oral or intraperitoneal). Toxicological analysis shows that PFCM increases the cell types involved in primary defense mechanisms and no significant changes in the biochemical parameters and organs (e.g., kidney and liver) were observed. However, the use of PFCM treatment increased the spleen weight when compared with the use of 5-fluorouracil.  相似文献
