Summary The uptake and release of tylosin by Wyoming bentonite, Ca++ montmorillonite, illite (Fithian Illinois) and kaolin (china clay) was studied. The adsorption capacities of bentonite and montmorillonite for tylosin were found to be 190 and 65 g mg–1 respectively, while the adsorption capacities of illite and kaolinite were found to be 22 and 6.5 g mg–1. 7.6 and 7.4% respectively of the tylosin adsorbed by bentonite and montmorillonite, and 30 and 25% respectively of the tylosin adsorbed by illite and kaolinite, was released in phosphate buffer.The presence of tylosin in two different soils amended with tylosin fermentation waste was also studied, and the effect of temperature on tylosin release was examined in one of the soils. The concentration of tylosin in the soil water was found to be related more to the concentration of tylosin fermentation waste than to the incubation temperature. The amounts of tylosin present in the initial leachates of the two soils, a modified John Innes compost and a limestone waste, were found to be in excess of the minimum inhibitory concentrations for susceptible bacteria. It is hypothesised that in these particular soils if the tylosin is not broken down by microbial action it could affect susceptible populations of bacteria. 相似文献

Summary
Nicotiana sylvestris cell lines resistant to the amino acid analogues S-2-aminoethyl-cysteine (AECR), or 5-methyl-tryptophan (5MTR), were isolated in suspension culture. Assuming these resistances to be dominant, we have attempted to determine if such variant cell lines can be used to select double resistant somatic cell hybrids. A total of 1.8 × 104 control calli from mixed AECR and 5MTR protoplasts, and AECR and 5MTR homokaryotic fusions were placed on double analogue selection, but none survived. Eight somatic hybrid calli (0.8%), able to grow without inhibition on the double analogue selection medium, were obtained after AECR + 5MTR protoplast fusion. These were further determined as hybrids on the basis of resistance level, chromosome number, and chlorophyll content, all characteristics differing in the parental cell lines.This study is part of a dissertation by D.W.R.W. in partial fulfillment of the requirements for the Ph. After being proposed by Lloret et al. in 2011, the resilience components resistance, recovery, resilience and relative resilience became highly popular for studying tree growth responses to disturbances and extreme weather events. As the ‘Lloret indices’, however, have conceptual limitations, various additional indices to characterize tree resilience have been discussed since then, namely: recovery period, total growth reduction, average recovery rate, and average growth reduction. In pointRes 2.0, a major update of the R-package pointRes, these new indices have now been implemented. Besides convenient functions for calculating and plotting resilience indices, two new functions for calculating pointer years using (i) the interval trend method, and (ii) a 'z-transformation of site chronology'-approach have been included as well. The superbug infection caused by New Delhi metallo-β-lactamase (NDM-1) has grown into an emerging threat, labelling and inhibition of NDM-1 has proven challenging due to its shuttling between pathogenic bacteria. Here, we report a potent covalent scaffold, ebsulfur, for targeting the protein in vitro and in vivo. Enzymatic kinetic study indicated that eighteen ebsulfurs gained except 1a–b and 1f inhibited NDM-1, exhibiting an IC50 value ranging of 0.16–9 μM, and 1g was found to be the best, dose- and time-dependent inhibitor with an IC50 of 0.16 μM. Also, these ebsulfurs effectively restored the antibacterial activity of cefazolin against E. coli expressing NDM-1, and the best effect was observed to be from 1g, 1i and 1n, resulting in an 256-fold reduction in MIC of the antibiotic at a dose of 16 μg/mL. The equilibrium dialysis study implied that the ebsulfur disrupted the coordination of one Zn(II) ion at active site of NDM-1. Labelling of NDM-1 using a constructed fluorescent ebsulfur Ebs-R suggested that the inhibitor covalently bound to the target through SDS-PAGE analysis in vitro. Also, labelling NDM-1 in living E. coli cells with Ebs-R by confocal microscopic imaging showed the real-time distribution change process of intracellular recombinant protein NDM-1. Moreover, the cytotoxicity of these ebsulfurs against L929 mouse fibroblastic cells was tested, and their capability to restore antibacterial activity of antibiotic against clinical strains E. coli EC08 producing NDM-1 was determined. The ebsulfur scaffold proposed here is valuable for development of the covalent irreversible inhibitors of NDM-1, and also for labelling the target in vitro and in vivo. 相似文献

Phagocytic cells know exactly where an infection is by following chemotactic signals. The phagocytosis of bacteria results in a ‘respiratory burst’ in which superoxide radicals are released. We have previously compared the release of reactive oxygen species (ROS) by antibiotics, during electron transfer reactions, to this event. Antibiotics in their normal bacterial environment, and ROS, are both increasingly implicated in purposeful signalling functions, rather than their more widely known roles in bacterial killing and molecular damage. Here, we extend our comparison between antibiotics and phagocytic cells to propose that antibiotics actively accumulate at a site of pathogen infection or tumour growth. A common link being virulent cellular growth. When this occurs, new proteins are secreted, aberrant iron acquisition takes place, and lipocalins are released. Each provide a mechanism by which antibiotics can bind, and be retained, at an active site of pathogen infection or tumour growth. 相似文献

Delafloxacin, a fourth-generation anionic fluoroquinolone (FQ) was approved in 2019 for community acquired bacterial pneumonia (CARP). It has broad spectrum activity and an improved class-related toxicity profile. However, it has recently failed a Phase 3 clinical trial for treatment of N. gonorrhoeae infections due to the lack of sufficient efficacy at the dose administered.Inspired by the microbiological and safety profile of delafloxacin, we have developed and profiled the first reported delafloxacin carbon analogue whereby a Nitrogen-for-Carbon swap has been successfully carried out at the C7 position. Not only have we shown that compounds with this modification maintain activity against N. gonorrhoeae (plus other gram-positive and gram-negative bacteria) but they also demonstrate a differentiated physicochemical profile.A zwitterionic derivative of delafloxacin was also profiled and demonstrated a superior microbiological profile against gram-negative strains, whilst maintaining favorable selected ADMET properties. 相似文献

The inheritance of resistance to Plum pox virus (PPV) has been studied in 1,178 apricot hybrids. Seven hundred and eighteen F1 hybrids, obtained from controlled crosses
between the susceptible Greek cultivar “Bebecou” and the resistant PPV cultivars of American origin (“Stark Early Orange,”
‘NJA2,” ‘Veecot,” “Sunglo,” “Harlayne,” and “Orangered”) were evaluated for resistance to the PPV-M (Marcus) strain, 8 years
after artificial inoculation. The inheritance of resistance to PPV has been additionally studied for the first time in a BC1 population of 95 apricot hybrids for four vegetative periods. Reaction of each hybrid to PPV-M was scored through visual
symptoms, indexing onto GF-305 and double-antibody sandwich enzyme-linked immunosorbent assay tests. Segregation within the
hybrids, determined by Chi-squared analysis, fits a 1:1 ratio (P ≤ 0.05) of the resistant vs susceptible, indicating that resistance to PPV is controlled by a single dominant gene locus
and that the above six resistant cultivars are heterozygous for the trait. Plants carrying this gene may initially develop
disease symptoms on leaves but eventually recover and no virus can be detected in leaves. Susceptible plants show similar
symptoms initially but remain symptomatic. Inheritance of resistance to PPV also has been studied in 365 F1 hybrids by crossing
the resistant cultivar “Stella” with the susceptible “Bebecou” and the resistant cultivars “Sunglo” and “NJA2,” for 8 years
after inoculation. The segregation ratio was 1:0 (resistant/susceptible) suggesting that “Stella” is homozygous for the resistance
trait. The purpose of this work was the enhancement of the knowledge of inheritance of resistance to PPV for the selection

Computational simulation models can provide a way of understanding and predicting insect population dynamics and evolution of resistance, but the usefulness of such models depends on generating or estimating the values of key parameters. In this paper, we describe four numerical algorithms generating or estimating key parameters for simulating four different processes within such models. First, we describe a novel method to generate an offspring genotype table for one- or two-locus genetic models for simulating evolution of resistance, and how this method can be extended to create offspring genotype tables for models with more than two loci. Second, we describe how we use a generalized inverse matrix to find a least-squares solution to an over-determined linear system for estimation of parameters in probit models of kill rates. This algorithm can also be used for the estimation of parameters of Freundlich adsorption isotherms. Third, we describe a simple algorithm to randomly select initial frequencies of genotypes either without any special constraints or with some pre-selected frequencies. Also we give a simple method to calculate the “stable” Hardy–Weinberg equilibrium proportions that would result from these initial frequencies. Fourth we describe how the problem of estimating the intrinsic rate of natural increase of a population can be converted to a root-finding problem and how the bisection algorithm can then be used to find the rate. We implemented all these algorithms using MATLAB and Python code; the key statements in both codes consist of only a few commands and are given in the appendices. The results of numerical experiments are also provided to demonstrate that our algorithms are valid and efficient. 相似文献

In vitro mutation and selection of doubled-haploid Brassica napus lines with improved resistance to Sclerotinia sclerotiorum 总被引:8,自引:0,他引:8
This paper describes a new protocol to develop doubled-haploid (DH) Brassica napus lines with improved resistance to Sclerotinia sclerotiorum. In this protocol, haploid seedlings derived from microspore cultures of B. napus were used to produce haploid calli for in vitro mutation-selection. For routine screening, mutation was induced by EMS (ethylmethane sulfonate) or occurred spontaneously, and screening for resistant mutants occurred on media with added oxalic acid (OA) as a selection agent. In tests with selected lines, the optimal concentration of EMS for mutation was determined to be 0.15%, and the optimal concentration of OA for in vitro screening was 3 mmol/l (half lethal dose was 3.1 mmol/l) for the first cycle of screening. There was an accumulated effect of OA toxicity on calli over two cycles of screening, but the growth and capacity of the surviving calli for regenerating seedlings were not affected by OA. Of the 54 DH lines produced from the in vitro mutation-selection, two DH lines of resistant mutants, named M083 and M004, were selected following seedling and glasshouse tests. The resistance of M083 and M004 to S. sclerotiorum following tests with both mycelial inoculum and OA was greater than that of their donor lines and the resistant control Zhongyou 821. In both glasshouse and field disease nurseries, disease indices on M083 and M004 were less than 50% of those of the control. The time required for M083 and M004 to mature was 14 days and 10 days shorter, respectively, than that of their donor lines. Furthermore, M083 had more pods per inflorescence, a greater 1,000 seed weight and higher yield than its donor line. Random amplified polymorphic DNA characterisation showed that M083 had DNA band patterns that differed from its donor line. 相似文献

The Gcn5-related N-acetyltransferases are an enormous superfamily of enzymes that are universally distributed in nature and that use acyl-CoAs to acylate their cognate substrates. In this review, we will examine those members of this superfamily that have been both structurally and mechanistically characterized. These include aminoglycoside N-acetyltransferases, serotonin N-acetyltransferase, glucosamine-6-phosphate N-acetyltransferase, the histone acetyltransferases, mycothiol synthase, protein N-myristoyltransferase, and the Fem family of amino acyl transferases. 相似文献
