The insulin-like effects of vanadiumin vivo are likely to be achieved at micromolar concentrations. Demonstrated effects of vanadium on adipose tissue of streptozotocin-diabetic rats include inhibition of basal and stimulated rates of lipolysis and effects on fat cell protein phosphorylation. The studies described below examined the effects of vanadium (to a maximum concentration of 0.5 mM) on adipose cells or tissuein vitro. Vanadium, added as a vanadyl-albumin complex or as sodium orthovanadate, produced a marked (greater than 50%) inhibition of isoproterenol-stimulated lipolysis. Inhibition of lipolysis equivalent to that seen with insulin, was achieved with 100 M vanadium. In contrast, no insulin-like stimulation ofde novo fatty acid biosynthesis was observed with vanadium below 0.5 mM. Surprisingly, the antilipolytic effects of vanadium persisted in the presence of cilostamide, an inhibitor of the insulin-sensitive isoform of cyclic nucleotide phosphodiesterase. Studies with purified preparations of the catalytic subunit of cyclic AMP-dependent protein kinase revealed dose-dependent inhibition with vanadyl-glutathione (to a maximum of 40% inhibition). Equivalent inhibition of cyclic AMP-dependent phosphorylation of Kemptide (50%) was observed upon incubation of freshly-prepared fat-pad supernatant fractions with vanadyl-glutathione. These results suggest that effects of low concentrations of vanadium may be mediated, at least in part, by actions on the catalytic subunit of cyclic AMP-dependent protein kinase.  相似文献

A wound stimulus evoked a number of repetivite action potentials in the leaf ofBiophytum sensitivum. When the cut end of a leaf was immersed in a leaf extract, the resulting repetitive action potentials continue for a long time. Abstract Immunization with lipopolysaccharide from Klebsiella O3 as an immunological adjuvant did not cause the death of mice in systemic anaphylaxis to bovine serum albumin. On the other hand, most mice immunized with lipopolysaccharide from Escherichia coli O111, Klebsiella O4 and Salmonella minnesota did die. Klebsiella O3 lipopolysaccharide enhanced IgM and IgG antibody response to BSA more markedly than Escherichia coli O111 lipopolysaccharide, while it affected the production of IgE antibody only slightly. Therefore, it is suggested that the inhibition of systemic anaphylaxis by Klebsiella O3 lipopolysaccharide adjuvant might be related to its strong adjuvant action on IgM and IgG class antibody production, and that high levels of circulating IgM and IgG antibodies might act as blocking antibodies in the development of IgE-mediated systemic anaphylaxis.  相似文献

The need for rigorous methods of interpreting indirect site ordinations is discussed and the problem of oblique habitat and phytosociological trends is emphasized. A sequential approach, termed integrated interpretation, is introduced to overcome this problem. It first involves a technique termed rotational correlation a linear form of trend surface analysis, which locates the best fit between a habitat variable and the coordinates of a pluridimensional ordination. The coordinates at positions of best fit are then used to produce two-way site-species tables (seriation arrays) which summarize floristic variation in relation to each major habitat trend. The seriation arrays can also be used to identify groups of differential species. Integrated interpretation is demonstrated using semi-arid vegetation from Murcia Province, SE Spain. A two-axis site ordination of vegetation data by non-metric multidimensional scaling is shown to have two oblique trends related to aspect-induced topoclimates and types of past anthropogenic disturbance. Rotation of the configuration to maximum linear correlation achieves very high levels of explanation for each factor. Rotated ordination coordinates from the correlation analysis are used to obtain two seriation arrays and floristic noda which relate directly to the two underlying habitat trends. A floristic discontinuity is revealed on the topoclimate continuum and it is hypothesized that temperature conditions are a major determinant of floristic composition, but that moisture partly determines vegetational cover and phytomass. Several patterns of past cultivation are detected and associated with five main kinds of geomorphological unit, together with some evidence that the rate of succession is partly determined by topoclimate. The integrated approach to interpretation is compared with visual methods and other multivariate techniques. A case is made for increased interpretational rigour in site coordinate studies, especially the discontinuation of trend-seeking using individual unrotated ordination axes.Nomenclature of vascular plants follows Tutin et al. The glucose transport activity solubilized from the basal and plus insulin forms of the Golgi-rich fraction of adipocytes was partially characterized, and the results were compared with those of the activity obtained from the plus insulin form of the plasma membrane-rich fraction. The transport activity was determined in a cell-free, reconstituted, system. Prior to reconstitution, the activities in the three preparations were all (a) stable at 0°C for at least 4 h, but not at 37°C or above; (b) most stable at pH 7–9, and (c) less stable in Tes than in Tris buffer. After reconstitution, the three activities were all (d) stable at 0°C, (e) most active at pH 5.5, (f) mildly stimulated by divalent cations, (g) unaffected by insulin or 1 mM of several SH-blocking agents, (h) inhibited by heavy metal ions, 10–100 mM of monovalent salts, organic solvents, several sugar isomers, and specific sugar-transport inhibitors. The rates of d-glucose uptake by the three liposome preparations were all inhibited more strongly by 2-deoxy-d-glucose or 3-O-methyl-d-glucose than by d-glucose. These data indicate that the general properties of the glucose transport activity in the Golgi-rich fraction are similar to those of the activity in the plasma membrane-rich fraction.  相似文献

A replication-defective Simian virus 40 genome, with a deletion of about 120 nucleotides in the region encoding the N-terminal fourth of the large T antigen, has been isolated from the DNA of Simian cells transformed by SV40. Both the original transformants, and the murine transformants obtained by transfection with this cloned mutant DNA, produced a large T antigen displaying in immunofluorescence an exclusively cytoplasmic localization. The protein apparent molecular mass (83 kDa) was about 6% smaller than that of normal karyophilic large T. Restriction analysis showed that the deletion eliminated two close HinfI sites, at nucleotides 4459 and 4376 (map unit 0.50).  相似文献

Summary Amino acid transport and incorporation have been studied in vitro in rat pancreatic lobules after maximal and supramaximal hormonal stimulation with caerulein. Incorporation into proteins was increased already after 30 and 120 min of maximal stimulation, but was decreased after the infusion of a supramaximal dose. Uptake of neutral amino acids was monitored using labeled leucine and -aminoisobutyric acid (AIB). In the case of leucine the free pool was consistently reduced after maximal stimulation, while supramaximal doses led to an increase which could be potentiated by the addition of 2mM tetracaine. Using AIB, a significant increase in the intracellular pool was observed after maximal stimulation, conversely a decrease after supramaximal stimulation. Release of labeled leucine and AIB from preloaded lobules during incubation in the cold was significantly reduced after maximal secretory stimulation, but was found enhanced by 200 to 300 percent after supramaximal stimulation. No fine structural alterations at junctional complexes or at both the lateral and luminal plasma membranes were observed after maximal stimulation except an increased number of exocytotic figures at the luminal face. However, supramaximal stimulation led to progressive rarefaction of the tight junctional network and disintegration of the gap junctions. Concomitantly, an equal distribution of membrane particles on both faces of the plasma membrane together with a random occurrence of exocytotic figures were observed.Supported by a grant from the Deutsche Forschungsgemeinschaft, Bonn-Bad Godesberg (SFB 122, project C 5). Zusammenfassung In der vorgelegten Arbeit wurde die sogenante deduktive Klassifikationsmethode der anthropogenen Pflanzengesellschaften behandelt. Diese Methode geht vom System der höheren Vegetationseinheiten der Braun-Blanquet Schule aus. Sie ermöglicht die syntaxonomische Bearbeitung zahlreicher anthropogener Gesellschaften unserer Kulturlandschaft, die nur aus Arten mit breiterer ökologischer und zönologischer Amplitude zusammengesetzt sind, und zwar ohne unerwünschte Verwechslung der Bestimmungsmerkmale einzelner Rangstufen des Systems (Klassen-, Ordnungs-, Verbands- und Assoziationskenn-und trennarten). Die Grundprinzipien der Methode wurden schon früher von Kopecký & Hejný (1971, 1973, 1974) und Kopecký (1976) erläutert. Die vorliegende Arbeit bringt eine präzisierte Auslegung der Methode auf Grund neuer Erfahrungen, die bei der Bearbeitung der spontan entstandenen strassenbegleitenden Gesellschaften derMolinio-Arrhenatheretea Tx. 1937 in Nordostböhmen gewonnen wurden. Aus dem reichen Aufnahmematerial werden nur einige Aufnahmen der Basalgesellschaften mitAlopecurus pratensis und der von ihnen abgeleiteten Derivatgesellschaften mitDactylis glomerata undAgropyron repens fur die anschauliche Demonstration der Methode beigegeben (Tab. 1 und 2). Die Aufnahmen der übrigen Gesellschafts-typen werden in einer anderen Arbeit (Kopecký 1978) veröffentlicht.
Summary Use of a so-called deductive method in syntaxonomic classification of roadside plant communities in NE Bohemia.The present paper explains the so-called deductive method of syntaxonomic classification of plant communities, derived from the system of syntaxa of higher rank of the Braun-Blanquet approach. This method enables the synthesis of vegetations units of the varied anthropogenic plant communities in the man-made landscape, and their arrangement within the existing system, without changing the classification and determination criteria of both the higher and lower syntaxa in this system. Basic principles of this method have been outlined earlier by Kopecký & Hejný (1971, 1973, 1974) and Kopecký (1976). Following the newly obtained data and experience, this paper brings further details and more precise explanation of the method. Recently, the deductive method has been applied to the classification of turf communities which spontaneously develop along the roads in NE Bohemia. In order to illustrate the principles of the method, selected relevés of basal communities withAlopecurus pratensis (Table 1), and respective communities derived from these basal communities (Table 2) are presented. A complete set of vegetation relevés of all other roadside communities will be published elsewhere (Kopecký 1978).
