Antheraea mylitta, a sericigenous insect of economical importance is often infected with an intracellular parasite of the genus Nosema. This pathogen is known to cause fatal pebrine disease and is considered as an important factor that strongly influences the development of the host. Larvae developed from the eggs laid by a female infected with Nosema sp. showed extended development period. The increment in the larval weight declined significantly in infected larvae in comparison to uninfected ones. Food consumption, digestion, relative consumption rate (RCR), efficiency of conversion of ingested food (ECI), efficiency of conversion of digested food (ECD), and relative growth rate (RGR) values declined significantly, but at the same time a significant increase in approximate digestibility (AD) was also observed. Silk production declined in infected larvae. Silk gland weight and shell weight also significantly declined following infection over uninfected larvae. The reproductive potential in adults declined significantly (P<0.001) with decrease in ovary weight (31.6%), fecundity (54.1%), and fertility (34.9%). Egg chorionation was also affected in adults, which developed from infected larvae. The maternal infection level in one generation (10.4 x 10(6) spores/female) decreased significantly in the next generation (8.0 x 10(6) spores/female).  相似文献

Pheromone-binding proteins (PBPs) located in the antennae of male moth species play an important role in olfaction. They are carrier proteins, believed to transport volatile hydrophobic pheromone molecules across the aqueous sensillar lymph to the membrane-bound G protein-coupled olfactory receptor proteins. The roles of PBPs in molecular recognition and the mechanisms of pheromone binding and release are poorly understood. Here, we report the NMR structure of a PBP from the giant silk moth Antheraea polyphemus. This is the first structure of a PBP with specific acetate-binding function in vivo. The protein consists of nine alpha-helices: alpha1a (residues 2-5), alpha1b (8-12), alpha1c (16-23), alpha2 (27-34), alpha3a (46-52), alpha3b (54-59), alpha4 (70-79), alpha5 (84-100) and alpha6 (107-125), held together by three disulfide bridges: 19-54, 50-108 and 97-117. A large hydrophobic cavity is located inside the protein, lined with side-chains from all nine helices. The acetate-binding site is located at the narrow end of the cavity formed by the helices alpha3b and alpha4. The pheromone can enter this cavity through an opening between the helix alpha1a, the C-terminal end of the helix alpha6, and the loop between alpha2 and alpha3a. We suggest that Trp37 may play an important role in the initial interaction with the ligand. Our analysis also shows that Asn53 plays the key role in recognition of acetate pheromones specifically, while Phe12, Phe36, Trp37, Phe76, and Phe118 are responsible for non-specific binding, and Leu8 and Ser9 may play a role in ligand chain length recognition.  相似文献

The segments 10 (S10) of the 11 double stranded RNA genomes from Antheraea mylitta cytoplasmic polyhedrosis virus (AmCPV) encoding a novel polyhedrin polypeptide was converted to cDNA, cloned, and sequenced. Three cDNA clones consisting of 1502 (AmCPV10-1), 1120 (AmCPV10-2), and 1415 (AmCPV10-3) nucleotides encoding polyhedrin of 254, 339, and 319 amino acids with molecular masses of 29, 39, and 37 kDa, respectively, were obtained, and verified by Northern analysis. These clones showed 70-94% sequence identity among them but none with any sequences in databases. The expression of AmCPV10-1 cDNA encoded polyhedrin in Sf-9 cells was detected by immunoblot analysis and formation of polyhedra by electron microscopy, as observed in AmCPV-infected gut cells, but no expression of AmCPV10-2 or AmCPV10-3 cDNA was detected, indicating that during AmCPV replication, along with functional S10 RNA, some defective variant forms of S10 RNAs are packaged in virion particles.  相似文献

【目的】优化柞蚕Antheraea pernyi基因组注释,更好地扩展其在比较基因组学及品种改良研究中的应用。【方法】对柞蚕进行全长转录组测序分析;经全长转录本与参考基因组比对,鉴定新基因及新转录本,并对这些新基因和新转录本进行功能注释及长链非编码RNAs (lncRNAs)预测。利用大量的蛋白质编码转录本和lncRNAs对柞蚕基因组中基因结构进行修订。最后创建矫正后的柞蚕基因组基因注释。【结果】新发现1 997个蛋白编码基因和3 399个lncRNA基因,分别由2 402个和3 574个全长转录本数据支持。发现柞蚕基因组含25 021个基因,其中19 825个基因是蛋白编码基因,包括7个保幼激素酸甲基转移酶基因。【结论】本研究促进了对柞蚕基因组基因注释信息的认识,为柞蚕及相关物种功能基因组及比较基因组学研究提供了很有用的数据资源。  相似文献

Long-term datasets are required to understand the response of long-lived organisms (e.g., gopher tortoises [Gopherus polyphemus]) to management actions, such as prescribed burns. Our objective was to estimate the effects of prescribed burning on gopher tortoise population dynamics over decadal time frames at Fort Stewart Army Reserve, southeastern Georgia, USA. We captured and marked adult tortoises from 1994–2020. In addition, since the early 1990s, managers at Fort Stewart collected spatial records of prescribed burns; thus, we could compare demography of the population to prescribed burning. We used a Bayesian hierarchical model (open population Jolly-Seber model) to estimate population parameters (emigration and survival, immigration and recruitment, and adult abundance) and their relationships with years since burn. We observed opposing responses to years since burn at 2 sites: abundance and the probability of staying (survival plus not emigrating) increased within 1 site when it had been more recently burned (F zones), but abundance and probability of staying in a second site increased when it had been longer since the site was burned (E zones). Some of these effects were weak but indicative of different responses to burning between the sites. Although the sites experienced similar burning regimes, they differed substantially in other habitat features: the F zones had almost twice the tree cover and lower soil sand composition, indicating that tortoise population responses to burning depend on habitat context. We inferred that the primary mechanism for demographic responses to years since burn was likely emigrating adults, which indicates the need for more detailed movement data. Our results demonstrate that gopher tortoise population responses to prescribed burning are complex, context dependent, and primarily influenced by tortoise movements. Therefore, prescribed burn plans may best accommodate spatially dynamic tortoise populations when they create spatial heterogeneity in burn ages within the range of typical tortoise movements. © 2021 The Wildlife Society.  相似文献

Pattern recognition receptors are known to participate in the activation of Prophenoloxidase system. In this study, a 1,3-β-D-glucan recognition protein was detected for the first time in Antheraea pernyi larvae (Ap-βGRP). Ap-βGRP was purified to 99.9% homogeneity from the hemolymph using traditional chromatographic methods. Ap-βGRP specifically bind 1,3-β-D-glucan and yeast, but not E. coli or M. luteus. The 1,3-β-D-glucan dependent phenoloxidase (PO) activity of the hemolymph inhibited by anti-Ap-βGRP antibody could be recovered by addition of purified Ap-βGRP. These results demonstrate that Ap-βGRP acts as a biosensor of 1,3-β-Dglucan to trigger the Prophenoloxidase system. A trace mount of 1,3-β-D-glucan or Ap-βGRP alone was unable to trigger the proPO system, but they both did. Ap-βGRP was specifically degraded following the activation of proPO with 1,3-β-Dglucan. These results indicate the variation in the amount of Ap-βGRP after specific immune challenge in A. pernyi hemolymph is an important regulation mechanism to immune response. Horseshoe crab (Limulus polyphemus) eggs are a dietary staple of the red knot (Calidris canutus) during its spring stopover on the Delaware Bay. Numbers of knots stopping in Delaware Bay declined in the 1990s concurrent with a decline in horseshoe crabs, leading to the hypothesis that reduced horseshoe crab egg abundance limited the red knot population. Management efforts, including a seasonal harvest moratorium in the Delaware Bay, have been instituted to restore crab populations to levels of sustainable use by multiple users, including migratory birds. Our objective was to evaluate the sufficiency of horseshoe crab eggs in Delaware Bay in May–June 2004 and 2005 for knots to refuel for their migratory flight to the Arctic breeding grounds. We examined egg counts to determine if there were fewer high egg-density sites later than earlier in the day and season, as migrating birds might deplete this resource. We studied foraging rates at red knot locations to determine if foraging probes increased with time of day and season as birds depleted surface eggs by pecking, then began probing for subsurface eggs. Finally, we experimentally tested whether red knots and their competitors depleted horseshoe crab eggs. Crab egg numbers at knot foraging sites did not decline throughout the day or season in 2004. In both years, we found no evidence that knots switched from pecking to probing with increases in time since sunrise or start of the stopover. Egg numbers were similar in exclosed and accessible plots on crab nesting depressions and in areas of open intertidal zone, but were significantly lower in accessible than in exclosed plots in the wrack line. Our results indicate that horseshoe crab eggs in Delaware Bay were sufficient to support the refueling of the present-day stopover population of red knots. If an increase in the availability of crab eggs to foraging birds does not result in an increase in knot numbers, managers must prioritize mitigation of limiting factors at other historically important spring stopovers and on the poorly understood breeding and wintering grounds in addition to the Delaware Bay. © 2011 The Wildlife Society.  相似文献
