In the tip-growing filamentous cell of the xanthophycean alga Vaucheria terrestris sensu Götz, a new growing tip develops in the non-growing, cylindrical region of the cell that was exposed by local illumination. The present study examined changes in the strength and extensibility of the cell wall of the new growing tip and in the matrix components of the inner surface of the cell wall. The internal pressure required to rupture the cell walls decreased remarkably during the early to middle stages of growing tip development, but the cell wall hardly extended before rupture. In contrast, during the middle and late stages of development, cell walls were extended by internal pressure. Atomic force microscopy revealed that protease-resistant, fine granular matrix components were present only at the apical portion of a normal growing tip, and were absent in the non-growing cylindrical region. In the early and middle stages of new growing tip development, these matrix components appeared in the cell walls in patches. These results suggest that first cell wall strength decreases and then cell wall extensibility increases in the development of new growing tips, and that protease-resistant, fine granular matrix components may be involved in rendering a cell wall extensible.  相似文献

Cadmium (Cd) is a widespread heavy metal pollutant and environmental and human health hazard, which may be partially resolved using green and cost-effective phytoremediation techniques. However, the efficiency of phytoremediation is often limited by the small biomass of Cd-hyperaccumulator plants. Although cattail (Typha angustifolia L.) is tolerant of heavy metals and has a high biomass, there is little information available on its detoxification mechanisms for heavy metals, especially Cd. In the present study we investigated the tolerance of cattail to Cd and mechanisms involved in its Cd detoxification. Our results show that: (a) cattail is tolerant of Cd; (b) the root Casparian band, cell wall, vacuole, glutathione (GSH), and glutathione peroxidase (GPX) play important roles in Cd detoxification; and (c) mechanisms of Cd detoxification differ in leaf cell cytoplasm (mainly a GSH-related antioxidant defense system) and root cell cytoplasm (mainly a GSH-related chelation system). In summary, cattail possesses multiple detoxification mechanisms for Cd and is a promising species for phytoremediation of Cd-polluted environments.  相似文献

A method to characterize plant cell wall polysaccharides is presented. The complexity of the polymer structures and the large number of different charged and uncharged monosaccharides that make up plant polysaccharides have previously made analysis technically demanding and laborious. Polysaccharide analysis using carbohydrate gel electrophoresis (PACE) relies on derivatization of reducing ends of sugars and oligosaccharides with a fluorophore, followed by electrophoresis under optimized conditions in polyacrylamide gels. We show that PACE is a sensitive and simple tool for studying the monosaccharide composition of polysaccharides and of cell wall preparations. In combination with specific hydrolases, it can be used to analyze the structure of polysaccharides. Moreover, the specificity and kinetics of the plant polysaccharide hydrolases themselves can be quickly and effectively studied. PACE can detect as little as 500 fmol of monosaccharides and 100 fmol of oligosaccharides, and it is fast and quantitative.  相似文献

以花粉发育为单核期的花药接种,在MS附加IAA 4ppm、K 2ppm、BA 2ppm的培养基上诱导愈伤组织,并可直接分化出“沙尔普斯”、“红岗利德”、“春香”3个品种的花药植株。稍加调整附加激素成分和浓度,可在“索非亚”、“宝交早生”、“戈雷拉”、“红衣”、“丽红”等品种的花药上直接经愈伤组织分化出植株。其中有“沙尔普斯”、“红岗利德”、“春香”、“宝交早生”、“索非亚”等5个品种的花药植株经过病毒检测确认不带SMoV、SCrV、SMYEV和SVBV4种病毒。对无病毒植株进行了田间对比试验,植株生长势和果实产量都明显超过对照品种。  相似文献

 We previously isolated three chitin synthase genes (chsA, chsB, and chsC) from Aspergillus nidulans. In the present work, we describe the isolation and characterization of another chitin synthase gene, named chsD, from A. nidulans. Its deduced amino acid sequence shows 56.7% and 55.9% amino acid identity, respectively, with Cal1 of Saccharomyces cerevisiae and Chs3 of Candida albicans. Disruption of chsD caused no defect in cell growth or morphology during the asexual cycle and caused no decrease in chitin content in hyphae. However, double disruption of chsA and chsD caused a remarkable decrease in the efficiency of conidia formation, while double disruption of chsC and chsD caused no defect. Thus it appears that chsA and chsD serve redundant functions in conidia formation.  相似文献

Crude extracts of cauliflower florets had high xyloglucan endotransglucosylase (XET) activity, but this was largely lost after partial purification and de-salting. Activity was restored (promoted up to 40-fold) by any of a wide variety of inorganic and organic salts. Optimum concentrations for Na+, K+ and NH4+ salts were typically ~300 mM. The chlorides of Ca2+, Mg2+, Al3+ and La3+ were optimally active at lower concentrations (e.g. 0.1 mM LaCl3), but became inhibitory at higher concentrations (e.g. 5 mM LaCl3). Some anionic polysaccharides at 0.04–0.2% w/v (e.g. gum arabic, pectin and hypochlorite-oxidised xyloglucan) promoted the XET activity of de-salted enzyme, especially if a sub-optimal concentration of NaCl was also present; others (e.g. homogalacturonan, 4-O-methyl-glucuronoxylan and alginate) were inhibitory. Similar ionic effects were noted on the XET activity of the Arabidopsis protein XTH24 (heterologously expressed by insect cells); in this case carboxymethylcellulose was also stimulatory. To look for endogenous modulators of XET activity, we prepared a cold-water extract of cauliflower florets; after boiling and centrifugation, the supernatant [boiled cauliflower preparation (BCP)] promoted the XET activity of de-salted cauliflower enzyme and of XTH24. About half the activator present in BCP was an ethanol-precipitable, anionic polymer of apparent Mr <5,000. After acid hydrolysis the polymer yielded much arabinose and galactose, and small amounts of galacturonic and glucuronic acids amino acids were also present. The polymer may thus contain arabinogalactan-proteins. 为了研究孢壁蛋白与家蚕微孢子虫发芽(孢原质弹出)的相关性,我们采用碳酸钾诱导微孢子虫体外发芽结合密度梯度离心的方法(简称GDGC法),收集纯化发芽后的孢子空壳(简称孢壳),对发芽液、纯化的孢壳及成熟孢子的孢壁蛋白组分进行了分析。结果表明:GDGC法可以获得高纯度孢壳,计算出其密度为1.130g/cm^3;与发芽前成熟孢子提取的孢壁蛋白相比,空孢壳可以提取到主要孢壁蛋白SWP32、SWP30、SWP25,同时发现SWP32、SWP25丰度有所降低;结合碳酸钾发芽液的蛋白电泳分析,发现孢壳上丰度降低的SWP32在发芽液蛋白样品中存在,LC—MS/MS数据分析也发现SWP32、SWP30、SWP25在碳酸钾处理液中都有存在;而用碳酸钾溶液处理冷冻孢子时,未观察到发芽现象,电泳结果显示此时K2CO,溶液中只有SWP30条带,说明在碳酸钾溶液诱导的发芽过程中SWP32和SWP25从孢壳上脱落可能与发芽相关而不是被碱性的碳酸钾溶解下来的[动物学报54(6):1068—1074,2008]。  相似文献

Carbohydrate-hybridization probes (Vreeland and Laetsch, 1989, Planta (177, 423–434) were used to localize the homogalacturonan (pectate) component of pectins in the cell walls of leaves and soybean root nodules. Leaves of two species of the dicotyledon Dubautia were compared; these species contain much pectin but differ in their tissue water relations with respect to their cell-wall properties. Maturation of the primary cell walls in nodules was studied in the Bradyrhizobium japonicum-Glycine max symbiosis. Probe labelling was based on the divalent-cation-mediated association between pectate in tissue sections and fluorescein-conjugated pectate fragments. Pectate was also labelled by mixed-dimer formation with fluorescent polyguluronate derived from alginate. The specificity of the probe for unesterified polygalacturonate was indicated by increased cell-wall labelling after chemical or enzymatic deesterification of tissue sections, in contrast to elimination of labelling by chemical esterification. Postfixation of tissue sections improved retention of soluble pectate. Pectate differences were found in the leaves among cell types, in degree of esterification, and between plant species. The cell walls of soybean nodules were strongly labelled by the pectate probe in nodules one week and three weeks after infection. Pectate was more highly esterified in the central infected zone than in the surrouding cortex. Within the infected zone, walls of uninfected cells and infected cells were similarly labelled by the pectate probe. In a study of androgenesis in 90 Norwegian genotypes of perennial ryegrass (Lolium perenne L.), heritabilities ranged from h b 2 =0.46 to 0.80. Very high or completely positive genotypic correlations were found between most characters of androgenetic response (e.g. embryo-like structures per 100 anthers, plants per 100 embryo-like structures, albino plants per 100 anthers, green plants per 100 anthers). Three genotypes, 2 Norwegian (7-5 and 9-5) and 1 Danish (245), which had significantly different androgenetic responses were selected to study the genetic control of the processes. Genotypes 7-5 and 9-5 were highly embryogenie, 7–5 and 245 were relatively high producers of green plants, while 9-5 was unable to produce green plants. The six possible reciprocal crosses between these three genotypes were made, and 10 or 11 F1 plants from each cross were used for anther culture experiments. The cross 7-5 x 245 showed average superiority over both parents for total plant regeneration and green plant production, results not previously reported. The phenotypic correlations estimated among progenies from the crosses ranged from r=-0.99*** to 0.81***. These considerable changes, relative to the results of the screening experiment, are most likely the result of changed allele frequencies caused by the strong selection of parents in these crosses, and a relatively simple genetical control. Abstract Bacteria grow by enlarging their envelope in such a way that osmotic pressure does not normally cause physical rupture. The strategy of Bacillus subtilis for both cylindrical elongation and pole formation is now substantially defined. Side-wall growth takes place by laying down new peptidoglycan, which is then displaced outwards, stretched and discarded; cross walls are laid down in the absence of stress, and then stretched and bulged outward as the septum is split and the pole is formed.  相似文献
