The biogenic amine serotonin is a neurotransmitter and modulator in both vertebrates and invertebrates. In the CNS of insects, serotonin is expressed by identifiable subsets of neurons. In this paper, we characterize the onset of expression in the brain and suboesophageal ganglion of the honeybee during pupal development. Several identified serotonin-immunoreactive neurons are present in the three neuromeres of the suboesophageal ganglion the dorsal protocerebrum, and the deutocerebrum at pupal ecdysis. Further immunoreactive neurons are incorporated into the developing pupal brain in two characteristic developmental phases. During the first phase, 5 days after pupal ecdysis, serotonin immunoreactivity is formed in the protocerebral central body, the lamina and lobula, and the deutocerebral antennal lobe. During the second phase, 2 days later, immunoreactivity appears in neurons of the protocerebral noduli of the central complex, the medulla, and the pedunculi and lobes of the mushroom bodies. Three novel serotonin-immunoreactive neurons that innervate the central complex and the mushroom bodies can be individually identified.

Attraction of many gravid female herbivore insects to suitable host plants is mediated largely by olfactory cues. Behaviorally, synergism among odor mixtures constituents underlies this attraction in some systems. Yet, the representation of synergistic odor-mixture effects is unknown in the antennal lobe, the first processing center for olfactory information in insect brains. Using both behavioral and physiological data we demonstrate that in the oriental fruit moth, Cydia (Grapholita) molesta, a minor constituent of a plant-derived synthetic mixture plays a key role in behavioral discrimination and in neural representation of mixtures. Behaviorally, minute amounts of benzonitrile added to an unattractive 4-compound mixture resulted in a bioactive 5-compound mixture that was as attractive to mated female moths as the natural blend. Physiologically, the bioactive benzonitrile-containing mixture elicited strong activation of one additional, new type of glomerulus that showed specific synergisms for this mixture. The specific pattern of activated glomeruli elicited by the addition of benzonitrile demonstrates a physiological correlate to the behaviorally observed synergism, and emphasizes the key role of a minor component of a complex mixture. While minor constituents of mixtures are often overlooked, they may, as conclusively documented here, be determinant for successful recognition and behavioral discrimination of suitable host plants by herbivore insects.

Highlights
  • •The RJB syndrome is mainly due to a shift in nurse bee alloparental care behavior.
  • •The RJB nurses spontaneously respond more often to larval odors compared to ITB nurses but their subsequent learning ability are similar.
  • •The RJBs evolve neurophysiological adaptations that increase the nurse bees' alloparental care behavior.
  • •Major royal jelly proteins and hexamerins play crucial neurobiological roles for the nervous system.

This study is the first to examine the circadian rhythms of melatonin in Eriocheir sinensis and Palaemonetes sinensis, two economically important crustaceans. We collected haemolymph and optic lobes from both species every 4 h over a whole day cycle. Melatonin content was measured with high-performance liquid chromatography. E. sinensis haemolymph exhibited significant (p < 0.05) peaks in melatonin at 16:00 (0.180 ± 0.020 μg·mL?1) and 24:00 (0.244 ± 0.055 μg·mL?1), while eyestalks had significant peaks at 16:00 (72.377 ± 18.100 μg·eyestalk?1) and 24:00 (98.756 ± 30.271 μg·eyestalk?1). In P. sinensis, melatonin peaked significantly only at 16:00 in optic lobes (12.493 ± 1.475 μg·eyestalk?1) (p < 0.05); no significant peaks were present in haemolymph. Thus, both E. sinensis and P. sinensis exhibit species-specific melatonin rhythms. Time of day should therefore be considered when examining the physiological status of both crustaceans, given the potential influence of fluctuating daily melatonin concentrations.
